The VSF was present as a macromolecule of over 100 kDa in a natural state, which is a minimum unit capable of displaying antiviral activity, and the highest antiviral activity of the VSF was found in a fraction of over 500 kDa. The purified VSF was treated with several proteases to find the proteolytic resistance. The sensitivity pattern of VSF to various proteases indicatethat it partially differ from the typical mouse IgG. The VSF maintained the activity for 3 days at 25 ℃ and 37 ℃. However, incubation of the purified VSF at 65 ℃ resulted in a time-dependent loss of antiviral activity, In addition, the purified VSF was found to be stable in a range of pH 7.0 to 8.0. Amino acid sequence was analyzed by N-terminal and internal sequencing, and MALDI-TOF. 5' capfishing and RACE PCR methods were performed to verify the nucleotide sequence of VSF. The results of amino acid and nucleic acid sequences for VSF reveal that VSF is a novel protein with some homology to mouse Ig (γ3, κ), Though structually VSF protein showes some resemblance with IgG in biochemical & sequencial characters, but functionally it differs drastically from the convensional IgG. Our results suggest that hybridoma cells produce an antiviral factor that protects several other viruses in vitro and in vivo and may have therapeutic potential.
