LBD41 유전자가 다른 LBD 유전자와 기능적 상호작용을 하는지 알아보기 위해, 옥신반응 중 측근 형성에 관여한다고 알려진 class Ⅰ의 LBD16 과 LBD18 유전자의 단일 및 이중 돌연변이체와 교배하여 이중 및 삼중 돌연변이체를 제작하였다. 이러한 돌연변이 식물체를 가지고 주근 및 하배축의 길이생장, 측근의 형성, 굴중성 그리고 굴광성 등 옥신반응에 관련된 표현형을 관찰하여 본 결과 lbd16 과 lbd18 돌연변이로 인한 측근 형성 감소 이외에는 주근 및 하배축의 길이생장과 굴중성, 굴광성 그리고 식물의 성장과 발달 등에서 표현형의 변화를 관찰할 수 없었다. 그러나 Steroid regulator-inducible system 을 이용한 LBD41 과다발현 Pro_(35S):LBD41:GR 형질전환 식물체의 표현형을 분석한 결과 dexamethasone (DEX) 처리에 의해 뿌리의 성장 및 측근의 수가 야생종과 비교하여 억제되는 현상을 관찰하였다. 또한, green fluorescent protein (GFP)을 통한 세포내 위치를 분석한 결과 LBD41 단백질이 핵에 존재한다는 것을 확인 하였으며, 효모를 이용한 전사활성분석을 통하여 LBD41 단백질의 C-terminus 안에 transactivation domain 이 존재하는 것을 확인할 수 있었다. 이러한 결과들은 LBD41 단백질이 식물체 안에서 뿌리의 성장과 측근의 형성을 조절하는 전사 조절인사로써의 기능을 가지고 있다는 것을 시사한다.

While the mechanisms at the auxin perception level are understood in details, pathways downstream of Aux/IAA-ARF proteins during auxin response are largely unknown. To study auxin signaling downstream of Aux/IAA-ARF, an inducible system developed for plant cells using regulatory mechanism of the mammalian glucocorticoid hormone receptor (GR) has been used, demonstrating that the iaa1 mutant protein with enhanced protein stability impaired a variety of auxin responses by acting as a negative regulator of auxin-responsive pathway. To get insights into signaling downstream of Aux/IAA-ARF in auxin response, the effects of DEX-inducible iaa1 on auxin-regulated gene expression had previously been examined and LATERAL ORGAN BOUNDARIES DOMAIN41 (LBD41) had been identified as one of the putative iaa1-target genes. In the present study, biological and molecular function of LBD41 was investigated. RT-PCR analysis showed that expression of LBD41 was up-regulated at 1 hour after exogenous treatment of IAA and down-regulated by simultaneous treatment of IAA and DEX in Pro_(35S):iaa1:GR transgenic plants. RNA gel-blot analysis of a variety of arf mutants with or without auxin showed that auxin responsiveness of LBD41 was inhibited by ARF10 or ARF13 mutations. These results indicate that LBD41 was an auxin responsive gene regulated downstream of Aux/IAA-ARF system. To study biological function of LBD41, phenotype of loss-of-function lbd41 single mutants was analyzed compared to wild type. lbd41 single mutants appeared to be similar to wild type. Arabidopsis T-DNA insertion multiple mutants of lbd16, lbd18, or lbd41 were then constructed, but LBD41 mutation did not give any significant effects on the phenotypes of these lbd multiple mutants. Overexpression of LBD41 in steroid-regulator inducible system resulted in inhibition of root elongation and lateral root formation compared to wild type. GFP:LBD41 fusion proteins can be localized to the nucleus in Arabidopsis mesophyll protoplasts, indicative of LBD41 as a nuclear protein. Using the yeast system, a putative transactivation domain of LBD41 was identified to be located at the C-terminus. Taken together, these results suggest that LBD41 might play a role as a putative transcriptional regulator for modulating lateral root formation and root elongation.
