닥나무 뿌리 MeOH 추출물 및 용매별 분획물로부터 생리활성을 탐색하고 활성물질로 추정되는 물질을 분리, 정제하여 구조 동정을 하였다. 우선 닥나무 뿌리를 MeOH을 이용하여 추출하였고, 이를 극성별로 n-Hexane부터 CHCl3, EtOAc 및 n-BuOH로 순차적으로 분획을 하였다. 추출물 및 분획물로부터 생리활성 탐색실험으로 전자공여능(DPPH), Tyrosinase 저해활성실험, 항균실험을 진행하여 활성능을 비교하였다. 전자공여능(DPPH)은 CHCl3 분획물 > EtOAc 분획물 > n-BuOH 분획물 > n-Hexane 분획물순으로 나타났다. 또한 Tyrosinase 저해 활성을 측정한 결과 n-Hexane 분획물을 제외한 나머지 분획물에 골고루 미백성분이 있는 것으로 추정되었다. 항균실험 결과 Paper disk법 등을 사용하여 피부상제균 및 여드름 균주의 항균활성을 실험한 결과 분획물 대부분 S. aureus, P. aeruginosa는 효과가 없었으며 P. acnes, S. epidermis 두 개의 균주만이 clear zone을 형성하였다. 특히 CHCl3 분획물이 P. acnes에 대해서 높은 항균활성을 보였다. 항산화, 미백 및 항균활성을 실험한 결과 CHCl3 분획물에서 모두 우수하게 나타났다. 생리활성이 우수한 CHCl3 분획물에서 활성이 있는 것으로 추정되는 물질을 분리하기 위하여 silica-gel column chromatography를 행하여 5개의 분획물을 얻었다. 그 중 수득량이 많은 분획물을 Preparative HPLC를 이용하여 물질을 분리하였다. 분리된 물질들은 HPLC를 이용하여 순도를 확인하였고 GC-MS를 이용하여 compound Ⅰ은 392, compound Ⅱ는 394로 분자량을 확인하였다. 두 물질은 각각 NMR을 사용하여 구조 동정한 결과 compound Ⅰ, compound Ⅱ의 구조를 isoprenylated flavan인 kazinol B, kazinol A로 확인 되었다.

In this study, Broussoneria kazinoki was investigated for the biological activity as antioxidants, skin-whitening agents and antibacterial agents through the isolation and structural elucidation of interested substances. Parts of plants used for testing of biological activities among paper mulberry (B. kazinoki) were roots. The roots of B. kazinoki were extracted with MeOH, and the concentrated extract was partitioned with n-Hexane, CHCl3, EtOAc and n-BuOH. In Electron Donating Ability (EDA), 33% of EDA was revealed in 10㎍/mL of MeOH extract, and the fractions of CHCl3, Ethyl acetate, n-Butanol fraction showed 45.3%, 19.1%, 9.2% of EDA. Among the fractions, CHCl3 fraction had high EDA. Tyrosinase inhibitory effect, which is related to skin-whitening, was 79% in B. kazinoki root (MeOH extract) at 10㎍/mL. Antibacterial effects by B. kazinoki against the human skin-resident microflora such as Staphylococcus aureus, S. epidermis, Pseudomonas aeruginosa and Propionibacterium acnes indicated that the MeOH extract and CHCl3 fraction were higher than that of n-Hexane, EtOAc and n-BuOH fraction. The CHCl3 fraction showed high antibacterial activities against P. acnes showing 14 ㎜ of clear zone in 2mg/disk, it showed both antioxidative and antibacterial activity. Active substances from CHCl3 fraction were purified through silica gel column chromatography and HPLC. The eluting solvent of column chromatography to isolate biological activity compounds from CHCl3 fraction consisted of CHCl3 and EtOAc. Biological activity compounds were analyzed using NMR. Their chemical structures were determined as Kazinol B and Kazinol A by comparing NMR data with those of the reported compounds from relative plants.
