Honeysuckle (Lonicera japonica Thunb.) has been used as antidote, diuretics, tonics, antipyretics and anti-inflamatory agents for centuries. The anti-inflammatory and analgesic injection drug prepared from honeysuckle which was named as SKLJI (SK Lonicera japonica injection) and contains sweroside as an active consituent. The aim of this study was to develop and validate a HPLC method for the determination of sweroside. The finally selected isocratic systems consisted of a YMC Hydrosphere (250 × 4.6 mm I.D., 5μm) C18 column, a flow rate of 1.0 ml/min, a detection wavelength of 250 nm, a column temperature of 30℃ and a mobile phase containing acetonitrile / 0.1% (v/v) H3PO4 in water, 9/91 (v/v). The validation characteristics of the specificity, the linearity, the precision, the accuracy and the robustness were then evaluated. The specificity was determined by HPLC coupled to photodiode array detector and electrospray ionization mass spectrometer (LC-DAD-ESI-MS). Calibration curve for the linearity (at five concentration ranges, n = 3 per a concentration) showed a good linear regression (r2 > 0.9999). It was found that the relative standard deviations (% RSD) of the retention time and the peak area for the repeatability at three concentration ranges (78.8, 216.4 and 354 μg/ml, n = 5 per a concentration) were 0.03, 0.04, 0.13% and 0.19, 0.21, 0.04%, respectively. For the reproducibility 0.39, 0.45, 0.12% and 0.82, 0.31, 0.84%, respectively. The mean recovery rates at three concentration ranges described above (n=3 per a concentration) were evaluated as 100.5, 99.7, 100.6%, respectively. The information of chromatographic factors that care should be taken which was obtained by the robustness test. This fully validated method can be successfully applied to determine sweroside in SKLJI.
