ABSTRACT Effects of Eriobotryae Folium (EF) on Gene Expression in Human HaCaT keratinocyte Kim Shin Dept. of Oriental Medicine Graduate School of Dongshin University (Directed by Prof. Park Hye-sun, O.M.D., Ph. D.) In the theory of traditional medicine, Eriobotryae Folium (EF) has been used to treat patients with cough in oriental countries. EF can clear away lung-heat, resolve phlegm, alleviate cough and lower the adverse-rising qi and arrest vomiting. Recently, EF is known to have anti-oxidantic action. For this reason, the present author designed to investigate the effects of EF on cell viability and proliferation rates of HaCaT keratinocyte, then effects on changes in genetic profile by EF. So, the genetic profile for the effect of EF on human derived HaCaT keratinocyte was measured using microarray technique, and the functional analysis on these genes was conducted. Total 536 genes were up-regulated and 587 genes were down-regulated in cells treated with EF. Genes induced or suppressed by EF were all mainly concerned with cellular metabolic process, primary metabolic process, protein binding, ion binding and cell part. Comparison study showed that expression levels of genes related regulation of molecular function and microfilament motor activity were elevated, on the other hand, those related membrane bound organelle were lowered significantly. The network of total protein interactions was measured using cytoscape program, and some key molecules, such as DAB2, JAK2, SOCS1, NRIP1, IRS1, APBB1, SIAH1 and MUC1 that can be used for elucidation of therapeutical mechanism of medicine in future were identified. These results suggest the possibility that EF can repair skin damage and also suggest that related mechanisms are involved in MET, MYO5A, MYO5B and JAK2 related signalling pathways.
