H1N1은 0.1 M NaOH 처리 공정에서 처리되자마자 검출한계이하로 불활화 되었고, 70℃, 80℃, 90℃ 열처리 공정에서도 모두 5분 만에 검출한계이하로 불활화 되었으며, 0.1% TnBP/10% Deoxycholic acid처리 공정에서도 모두 5분 만에 검출한계 이하로 불활화 되었다. 0.3% TnBP/1% Triton X-100 농도에 따른 처리 공정에서는 농도에 상관없이 1분 만에 검출한계 이하로 불활화 되었다. 70% EtOH와 70% Propanol처리 공정에서 1분 만에 검출한계이하로 불활화 되었다. 혈액 제제에서는 알부민 제조공정 중 58℃ 열처리 공정에서 30분 만에 검출한계이하로 불활화 되었고, 에탄올 분획공정에서도 검출한계이하로 불활화 되었다. 사람면역글로불린 제조공정 중 냉에탄올 분획공정에서 검출한계이하로 불활화 되었고, 58℃ 열처리 공정에서 30분 만에 검출한계이하로 불활화 되었다. Antithrombin III 제조공정 중 58℃ 열처리 공정에서 30분 만에 검출한계이하로 불활화 되었고, 바이러스 필터 공정에서 완벽하게 제거되었다. Green plast FXIII 제조공정 중 58℃ 열처리 공정에서 120분 만에 검출한계이하로 불활화 되었고, Solvent/Detergent(0.3% TnBP /1% Tween 80) 처리 공정에서 완전히 불활화 되지 않지만, 1시간 처리 후 log reduction factor는 3.5이었다. Green plast thrombin 제조공정 중 Solvent/Detergent(0.3% TnBP/1% Tween 80) 처리 공정에서 60분만에 검출한계이하로 불활화 되었다. Fibrinogen 제조공정 중 Dryheat처리 공정에서 6% Glucose 안정제를 사용한 경우 10분 만에 검출한계이하로 불활화 되었고, 6% Saccharose, 6% Amino acid 안정제를 사용한 경우 20분 만에 검출한계이하로 불활화 되었다. Green eight 제조공정 중 Solvent/Detergent(0.3% TnBP/1% Tween 80)처리 공정에서 60분 만에 검출한계이하로 불활화 되었고, Dryheat 처리 공정에서 10분 만에 검출한계이하로 불활화 되었다. Green mono 제조공정 중 Solvent /Detergent(0.3% TnBP/1% Tryton X-100) 처리 공정에서 5분 만에 검출한계이하로 불활화 되었다. 위와 같은 결과 H1N1은 바이오의약품 제조 공정 중 바이러스 불활화/제거 공정에 의해 효과적으로 불활화/제거됨을 확인하였다.

Pandemic influenza A Virus H1N1 2009 has been identified as the cause of a widespread outbreak of febrile respiratory infection. Although the severity of this illness has ranged from mild to severe, little has been reported about how this outbreak has affected the safety of biopharmaceticals including plasma derivatives. To evaluate the safety of plasma derivatives, dedicated virus clearance processes used during the production of plasma derivatives as well as general viral inactivation processes were investigated for their effectiveness in inactivating and/or removing this virus of recent concern. In this study, influenza A virus H1N1 strain A/NWS/33 (H1N1) was chosen as the model of pandemic influenza A virus H1N1 2009. H1N1 was effectively inactivated to undetectable levels within one minute of 0.1 M NaOH, 70% EtOH, 70% propanol and solvent/detergent(S/D) treatment. AND H1N1 was inactivated to undetectable levels within five minutes of heat treatment at 70, 80, 90℃. Also H1N1 was completely inactivated by EO gas treatment. H1N1 was completely inactivated by cold ethanol fractionation process during the manufacture of albumin. H1N1 was also completely partitioned by cold ethanol fractionation and completely inactivated by low pH treatment and pasteurization during the manufacture of immunoglobulins. H1N1 was completely inactivated and removed by pasteurization and virus filtration process during the manufacture of antithrombin III. H1N1 was completely inactivated by pasteurization and partially inactivated by solvent/detergent(0.3% TnBP/1% Tween 80) treatment during the manufacture of factor XIII. H1N1 was completely inactivated by the 60 min treatment of S/D(0.3% TnBP/1% Tween 80) during the manufacture of thrombin. H1N1 was completely inactivated during the manufacture of thrombin. H1N1 was completely inactivated by dryheat treatment at 100℃ for 30 min using 6% glucose, 6% saccharose and 6% amino acids as the stabilizer during the manufacture of fibrinogen. H1N1 was completely inactivated by S/D(0.3% TnBP/1% Triton X-100) treatment and deryheat during the manufacture of factor VIII. These results indicate that H1N1 was effectively removed and/or inactivated during the manufacture of plasma-derived biophamaceuticals.
