Bar strip을 이용하여 확인된 T0 형질전환 25개 이벤트는 격리온실 내에서 이앙과 재배 후 제초제(0.3%, 상표명 바스타) 처리에 의한 생물검정을 통하여 형질전환체를 확인하였을 때, 22개 이벤트는 다량의 종자(T1)를 획득하였고, 3개의 이벤트는 불임으로 종자를 얻지 못했다. 형질전환 T1 이벤트의 정상종자 형성율은 대조구 82%에 비하여 T23 이벤트(90.7%)를 제외하고 평균 48.8%로 낮은 경향이었으며, 발아율은 대조구 87.5%에 비하여 T13 이벤트(62%)가 가장 낮았지만 대부분의 이벤트가 80% 이상이었다. 선발 마커로 사용하는 제초제(0.3%, 상표명 바스타)로 형질전환체를 선발한 고정율은 평균 75.7%이었으며, 특히 T22 이벤트는 98.3%의 높은 고정율을 보였다. We constructed an over-expression binary vector producing the functional heat-labile enterotoxin (LTB) subunit of Escherichia coli and cholera toxin of Vibrio cholerae (CTB) in transgenic rice seeds. We confirmed the insertion of two genes in the binary vector for rice transformation by restriction enzyme digestion and polymerase chain reaction. Both LTB and CTB genes showed 82.3% and 80.2% homologies in nucleotide and amino acids sequences, respectively. We produced the transgenic rice plants of twenty-fives events including 104 lines. The stable integration of the LTB, CTB and bar genes into the rice nuclear genome was confirmed by PCR analyses. Also, the stable integration of the LTB gene into the rice nuclear genome was verified by Southern blot analyses. Through the analyses of flanking regions, we identified the integration sites in chromosomes of transgenic rices. Bar strip test and Western blot analyses for T1 rice showed that PAT and LTB proteins were expressed in transgenic rice seeds. The number of developed seed in transgenic rices showed the decrease compared to that of wild type rice, but germination rate was similar to that of wild type rice. The survival ratio of transgenic rice plants treated by Basta herbicide was over 60%. And we made sure that twenty-two events of transgenic T1 plants were the transformants containing LTB, CTB and bar genes using analyses of PCR, reverse-transcriptase PCR, Bar strip and 0.3% basta treatment. GM1-ganglioside binding assays showed that recommbinant LTB proteins produced in transgenic rice plants could bind to the intestinal membrane GM1-ganglioside receptor, indicating correct folding and disulfide bond formation of LTB pentamers within transgenic rice. We proved transgenic T05 event that one copy of LTB and CTB genes were integrated within exon site of protein kinase-like domain containing protein gene existing in rice chromosome 3. Also, the transgenic T05 event was propertied as follows: the ratio of seeds harvested (67.6%), the rate of germination (64.6%) and the ratio of the fixed transgenic plants (69.1%).
