본 연구는 고지방식이 후 저지방식이로의 식이의 교체에 따른 간 조직의 지질대사 효소인 acyl coenzyme A synthetase (ACS), carnitine palmitoyltransferase-1 (CPT-1), acetyl coenzyme A carboxylase (ACC) mRNA 발현수준과 혈청 및 간 조직의 지질함량에 미치는 영향을 조사하고자 하였다.
5주령 C57BL/6종 24 마리를 저지방식이군 (low-fat diet, LFD)과 고지방식이군 (high fat diet, HFD)의 2 군으로 나누고 각각의 실험 식이를 급여하며 9 주간 사육하였다. 9 주간의 고지방식이와 저지방식이를 급여한 동물은 식이의 변화 없이 기존의 식이를 유지하는 LLD군과 HHD군으로 나누고, 식이지방 함량에 변화를 주는 군으로 저지방식이에 적응한 동물에게 고지방 식이로 교체하여 공급하는 LHD군과 고지방 식이에 적응한 동물에게 저지방식이로 교체하여 공급하는 HLD군으로 나누어 3 주간 사육하였다. 실험동물을 희생시킨 후 혈액과 간 조직의 지질농도와 간 조직에서의 지질대사에 관여하는 효소의 mRNA의 변화를 관찰하였다.
9주간 고지방식이에 적응한 동물에게 저지방식이를 급여한 군에서는 고지방식이군과 비교하여 체중, 간, 체지방조직의 무게가 유의적으로 감소하는 결과를 나타내었으며(p<0.05), 혈청 중성지방, 총 콜레스테롤, LDL- 콜레스테롤의 농도가 감소하였다. 저지방식이로 교체한 군은 고지방식이군과 비교하여 간 조직내의 ACS, CPT-1의 mRNA의 발현이 유의적으로 감소한 반면, 간 조직내의 ACC의 mRNA의 발현은 고지방식이군에 비하여 유의적으로 높게 나타났다(p<0.05).
본 연구의 결과, 식이 중 지방함량을 달리하였을 때 간 조직의 ACS, CPT-1, ACC의 발현의 변화뿐만 아니라 혈청 지질농도의 개선에도 유익한 영향을 미치는 것으로 나타났다.

The purpose of this study was to investigate the effects of dietary changes from high fat diet (HFD, 45% of total calories from fat) to low-fat diet (LFD, 10% of total calories from fat) on mRNA levels of hepatic acyl coenzyme A synthetase (ACS), carnitine palmitoyltransferase-1 (CPT-1), and acetyl coenzyme A carboxylase (ACC) and on serum and hepatic lipid profile in mice. Five weeks-old male C57BL/6 mice were fed high fat diet for 9 weeks then switched to a low fat diet or continued the high fat diet for 3 weeks. Body weight, serum and hepatic lipid levels were measured. Expressions of ACS, CPT-1, and ACC mRNA were also analyzed. The results showed that after the high fat diets were switched to low fat diets, body weight, liver, epididymal and retroperitoneal fats were significantly reduced compared with those of high fat diet group (p<0.05). The lipid blood levels (TG, Total-cholesterol, and LDL-cholesterol) decreased. Hepatic ACS and CPT-1 mRNA levels were lower in the high fat diet group than other groups (p<0.05). On the other hand, hepatic ACC mRNA levels were higher in the low fat diet group than other groups (p<0.05). Changes in the composition of dietary fat from high to low decreased significantly body fats and serum TGs. Furthermore, changes of dietary fat position affected differently the hepatic mRNA levels of ACS, CPT-1 and ACC.
