Acetyl-CoA carboxylase (ACC) 효소는 acetyl-CoA의 malonyl-CoA로의 carboxylation을 촉진하는 효소인데 생체내에서 지방산 합성 속도를 조절해 주는 rate-limiting 효소중의 하나이다. 본 연구에서는 Acetyl-CoA Carboxylase P1 promoter의 활성 조절을 규명함으로써 영양상태 및 호르몬 상태에 따른 ACC 유전자 발현 조절 기전을 밝히고자 한다. 1. 인슐린 처리에 의해 유도된 ACC promoter I의 활성도는 10$^-^7$M 인슐린에서 약 4.5배로 가장 크게 증가하였고 이 보다 더 높은 농도의 인슐린 첨가시 ACC I 활성도가 오히려 감소되는 것으로 나타났다. 2. 인슐린을 단독으로 처리했을 때 보다 1$\mu$M의 dexamethasone을 첨가했을 때 ACC promoter I 활성도가 전체적으로 약 1.5배 증가하여 인슐린에 의한 ACC promoter I 활성도 상승작용에 dexamethasone의 첨가 효과가 있음을 보여주었다. 3. 호르몬의 상호작용에 의한 ACC promoter I 활성도의 조절은 인슐린, dexamethasone 및 T3를 처리했을 때 약 10배 증가하여 ACC promoter I 활성도가 3개의 호르몬을 모두 처리하였을 때 가장 많이 증가하였다. 4. 25mM 포도당 첨가시 ACC promoter I 활성도가 약 1.7배 증가되었다. 5. PUFA (linolenic acid, arachidonic acid, DHA 및 EPA) 처리는 ACC promoter I 활성도를 감소시켰으며 그 중에서 EPA가 가장 큰 효과를 나타내어 약 65%의 감소를 나타냈다. 이상의 연구 결과들을 종합하면 ACC promoter I의 활성도는 인슐린, T3, dexamethasone 간의 상호작용에 의해 증가되는 것으로 나타났으며, ACC promoter I의 -1.2 kb 영역에 인슐린, T3, dexamethasone, 포도당 및 불포화지방산 등의 response element 염기서열이 있음을 시사하고 있다. 1. ACC gene expression is regulated by nutritional and hormonal status. The present study is performed to identify the regulation mechanism of ACC gene promoter I. 1. insulin treatment, respectively. 2. In the presence of dexamethasone (1$\mu$M), the effects of insulin increased about 1.5-fold, showing the additional effects of dexamethasone. 3. The activity of ACC gene promoter I increased with insulin+dexamethasone, insulin+T3, dexamethasone+ T3, and dexamethasone+insulin+T3 treatment approximately 6-, 4-, 6.5-, and 10-fold, respectively. 4. Glucose (25mM) treatment increased the ACC promoter I activity by 1.7-fold. 5. PUFA(linolenic acid, arachidonic acid, DHA or EPA) treatment decreased ACC promoter I activity, and EPA showed the largest effect by decreasing about 65%. Therefore it can be postulated that 1) insulin, dexamethasone, and thyroid hormone coordinately regulate ACC gene expression via regulation of promoter activity, 2) the -1.2-kb region of ACC promoter I may have the response element sequences for insulin, dexamethasone, T3, glucose, and polyunsaturated fatty acids. 1.
