Turmeric(Curcuma longa L) and licorice(Glycyrrhiza uralensis) are well-known as oriental traditional medicinal plant. Curcumin, the active component of Turmeric, is known to have a wide array of bioactivity including anti-oxidant, anti-inflammatory, anti-cancer, and cardio-protective properties. Licorice containing glycyrrhizin is known for a variety of biological activities, such as anti-oxidants and anti-inflammatory properties. This study was designed to compare how effectively turmeric and licorice extracts(hot water and ethanol) with or without roasting ameliorate oxidative stress and inflammatory responses. Roasting treatment increased total antioxidant capacity and total phenol content both of licorice hot water and ethanol extracts, but did not affect of turmeric extracts. Total antioxidant capacity and total phenol content were higher in ethanol extracts than in hot water extracts. Treatment of RAW 264.7 cells with turmeric ethanol extract significantly reduced lipopolysaccharide (LPS)-stimulated nitric oxide(NO) and TNF-α production, while licorice ethanol extract significantly decreased NO production. Protein expressions of antioxidative enzymes, such as catalase, superoxide dismutase(SOD), and glutathione reductase, were significantly elevated in turmeric and licorice extracts-treated cells. Furthermore, expressions of inducible nitric oxide synthase(iNOS) and COX-2 were also significantly decreased in turmeric extracts and licorice ethanol extract-treated cells. However, licorice hot water extracts did not affect iNOS and COX-2 expressions tested at the level of 400㎍/ml. Ethanol extracts of turmeric and licorice had stronger elevation on antioxidative enzyme expressions and suppression on inflammatory enzyme expressions than their hot water extracts. These results suggest that turmeric and licorice extracts has mild antioxidant and anti-inflammatory activities to suppress intracellular oxidative stress and inflammatory responses through the up-regulation of antioxidant enzyme expressions and down-regulation of iNOS expression.
