Inhibitory effect of the extracts of miscellaneous cereals on enzymatic activities of a-amylase and a-glucosidase :

In our investigation, most effective a-amylase inhibitory activity was shown at glutinous sorghum and proso millet among the 80% EtOH extracts of 13 miscellaneous cereals, and the n-hexane and MC fractions had strongest inhibitory effect on a-amylase. The MC fractions of glutinous sorghum, and proso millet, 200 mg/ml of acarbose were treated with heat (15 ~ 60 min) and acid (pH 2.0; 1 ~ 2 hr) then mixed with enzyme reaction mixtures, there was no change of inhibitory activity against a-amylase. So, we could find that a-amylase inhibitors which were contained in cereals were stable against long time heating and strong acidic conditions like mammalian stomach. Among the 80% EtOH extracts of 13 miscellaneous cereals, the 80% EtOH extract of golden glutinous sorghum had strongest inhibitory effect on a-glucosidase, and the second was the 80% EtOH extract of barnyardgrass. Especially, the 80% EtOH extract of golden glutinous sorghum was stronger 1.5 times than same amount of acarbose. The 30 mg/ml of n-hexane and MC fractions of golden glutinous sorghum were strongest inhibitory activity against a-glucosidase, these inhibitory activity were similar to 340 times condensed acarbose (5 mg/ml). Total phenolic compound contents were most in EtOAc fractions golden glutinous sorghum, and barnyardgrass, glutinous sorghum, so we could confirm that phenolic acid was not a-amylase and a-glucosidase inhibitor. We can also confirm that linoleic acid was not a-amylase and a-glucosidase inhibitor by the test of inhibitory activity of linoleic acid on a-amylase and a-glucosidase. In animal experiment, we could confirm that the 80% EtOH extract of golden glutinous sorghum suppressed the rapidly increase of postprandial blood glucose level. The result indicated that the 80% EtOH extract of golden glutinous sorghum might delay the absorption of carbohydrates after meal. In the further study, we need more detailed and careful GC-MS analysis to find single molecule which inhibit effectively a-amylase and a-glucosidase. The 80% EtOH extracts of cereals will be treated to diabetes induced mice with streptozotocin, and confirm that the cereal extracts are effective to lowering postprandial blood glucose level and able to use instead of acarbose.
