In this, study, we developed a new effective method to separate α-tomatine and dehydrotomatine which are tomato glycoalkaloid (TGA) contained in tomato using alumina column chromatography and HPLC chromatography. 1. The TGA contained in tomato was extracted by acid solvent, added in the alumina column, and fractionated by hydrated N-butanol. α-tomatine could be separated from 1 fraction (Fr) per 5 mL at 0.5 mL/min velocity of solvent in the Fr.No.26 to 36. In the result, we found that 0.414% α-tomatine was contained in the freeze dried tomato powder. 2. Two peaks were detected by HPLC analysis of a fractionated α-tomatine using alumina column chromatography. We separated detected two peaks using HPLC and analyzed aglycone and its glycoside produced by acid hydrolysis using GC. The first peak was identified as dehydrotomatine since analyzed aglycone was tomatidenol and sugar composition was xylose, glucose, and galactose (Mol ratio 1:1:2). The second peak was identified as α-tomatine showing aglycone was tomatidine and sugar components was identical with dehydrotomatine. Taken together, dehydrotomatine and α-tomatine could be separated from tomato by simple manipulation at a low price. In Chapter 2, we extracted each tomatine from 11 growth stage-tomatoes (7 growth stage of green tomatoes and 4 stages of ripening red tomatoes), separated α-tomatine and dehydrotomatine, and measured contents of them. In addition, we examined how the extracted tomatine at different stage affects on normal cell lines (Chang, human normal liver cell; Hel299, human normal lung cell) and cancer cell lines (U937, human histiocytic lymphoma cell; A549, human lung cancer cell; Hela, human cervical carcinoma cell) using microculture tetrazolium (MTT) assay and investigated correlation between tomatine contents and inhibitory activity on cancer cell proliferation. 1. We examined the effects of extracted and dried TGA on inhibition of proliferation in the normal (Chang and Hel229 cells) and cancer cell lines (U937, A549, and Hela cells). The activity of TGA in S1 to S7 samples was weak as 1uL/mL to 10uL/mL and extracted TGA from them did not affect proliferation of normal cells. However, TGA could reduce cell proliferation in U977, A549, and Hela cancer cell lines in a dose-dependent manner and a low concentration of TGA at the level of 1uL/mL still had strong inhibitory activity on the cancer cell proliferation. 5. We examined IC_(50), 50% inhibitory concentration of TGA on normal and cancer cell lines. IC_(50) of S3 was the lowest as IC_(50)=0.89 but that of S6 was the highest as IC_(50)=8.47 on Chang, normal liver cell line. IC_(50) of all samples on Hel299 cell was over 1.55. That means IC_(50) of Hel299 was higher than that of Chang cell lines. S6 (38.8%) and S1 (54.9%) had enhanced inhibitory effect on A549 and Hela cancer cell lines compared to Chang and Hel299 normal cell lines.
