This study was conducted to investigate the antimicrobial effect, antioxidative activity and cytotoxicity of Galla Rhois extract and fermented Galla Rhois extract and to examine the development possibility about functional food for specified health use, Galla Rhois. The results were as follows; 1. To treatment of Galla Rhois extract against B. cereus, B. subtilis, S. aureus, V. parahaemolyticus, E. coli and S. typhymurinum at various concentration of 300 ppm, to 500 ppm, 750 ppm, and 1000 ppm and then process 6, 12, 24, 36, 48 hour surveyed the growth inhibition of bacteria. As the results, antibacterial activity of Galla Rhois extract showed some difference that depends on bacteria. But there showed antibacterial activity in all the declared bacteria except S. typhymurinum. 2. The survey about antibacterial activity of the Galla Rhois fermented extract showed that antibacterial ability that was fermented by L. delbrueckii and S. cerevisiae had been increased, but antibacterial activity of the other bacteria reduced or had no difference. And there were some differences according to bacteria, treatment concentration and fermentation bacteria. Antibacterial activity of fermented extract using especially L. delbrueckii and S. cerevisiae was proved that it was good with even 2 percents concentration. 3. Antibacterial activity of Galla Rhois extract within pH 3 to pH 7 had been safe regardless of pH but low over pH 9. 4. Antibacterial activity of fractions into n-hexane, EtOEt, EtOAc, n-BuOH, and water which each polar was different in order to observe the characters against activity component of Galla Rhois extract was examined. As the result, each EtOEt, EtOAc and n-BuOH showed high antibacterial activity and each fraction had a concentration dependent antibacterial activity. 5. Galla Rhois extract against F, solani and R, solani had relatively high antifungal activity but C. gloeosporioides had antifungal activity in only more than 10 percents and A. panax and P. expansum, very low. So antifungal activity showd a lot of difference in fraction that was divided into EtOEt, EtOAc , n-BuOH and water sequentially, and antifungal activity showed generally lower than that of antibacterial activity. 6. Total flavonoid content of Galla Rhois extract was low, 7㎎/g and could get a little increased by fermentation. Total polyphenol content showed about 25㎎/g, it had no change regardless of fermentation 7. Electron donating ability of Galla Rhois extract was lower than vitamin C, contrast group, but after fermentation, it had been increased twice as much as before fermentation. Superoxide dismutase-like activity showed relatively low level, less than 5 percents, it had been decreased after fermentation. 8. Nitrite scavenging ability of Galla Rhois extract expressed higher numerical value than that of BHT in pH 2.5 and pH 4.2, and nitrite scavenging ability in pH 2.5 expressed higher than in pH 4.2. 9. Processing fermented Galla Rhois extract with Raw 264.7 cell that was activated by LPS, the concentration of Galla Rhois extract was getting higher and higher, while NO growth reduced. This state expressed much clearer after fermentation, so it could be imagined to better for to use for people who have a health problem. Therefore we need to study about this more in the future. 10. Galla Rhois extract in fibroblast cell and HepG2 cell showed to inhibit lipid peroxidation in 100μg/mg concentration compare to H2O2 treatment but there was not any difference according to processing concentration. 11. As the consequence about the effect of Galla Rhois extract for cell viability, fibroblast cell expressed that the cell viability had been increased under from 10㎍/㎎ to 100㎍/㎎, but under over 200㎍/㎎, the viability appeared low up to 80 percents level. And HepG2 cell viability showed similarity to a contrast group up to 100㎍/㎎, but the viability to a concentration-dependent over 200㎍/㎎ had reduced and the cytotoxicity had appeared. However if Galla Rhois extract was used, it will be able to deal with even this problem.
