(국문초록) 아토피 피부염은 IgE 농도의 증가와 Th2 세포의 사이토카인 및 케모카인의 과잉생산에 의해 나타나는 만성염증성 피부질환이다. 아직까지 아토피 피부염의 정확한 발병기전은 밝혀지지 않았으며, 최근 아토피성 피부염 연구에 있어서 NC/Nga mice의 동물 모델의 사용이 적합한 것으로 알려져 있다. 본 연구에서는 아토피 피부염 동물모델인 NC/Nga mice에 DNCB를 반복적 사용으로 유도된 아토피 피부염 질환에서 어성초, 삼백초, 자초를 동량으로 배합한 HSL의 피부개선효과를 평가하였다. HSL의 효능은 조직병리학적 검사 와, 혈중 IgE 농도, TEWL의 측정, Scratching behavior 및 Clinical skin severity score를 평가하였다. DNCB를 도포하여 4주동안 경구투여한 결과, Clinical skin severity의 감소, TEWL의 감소, Scratching behavior의 감소 및 혈중 IgE 농도가 유의하게 감소한 결과를 나타냈다. 조직병리학적평가에서는 HSL에 의해 염증세포로의 침윤, 탈과립된 비만세포와 호산구수, 표피층과 각질층의 두께가 유의하게 감소되었다. HaCaT 세포를 이용한 in vitro 실험에서는 MTT assay와 gelatin zymography로 MMP-2와 MMP-9의 발현, TARC kit를 이용하여 TARC 분비를 측정하였다. HCH, SCH, LEH의 처리에 의한 HaCaT 세포의 증식능은 농도별로 유의하게 감소하였다. 유사하게, TPA와 TGF-b에 의한 MMP-9 발현도 농도별로 억제된 결과를 나타냈다. 또한 HaCaT 세포에서 INF-g (10 ng/ml)와 TNF-a (10 ng/ml)의 병용처리로 유도된 TARC 발현도 농도별로 감소한 결과를 나타냈다. 이러한 결과로 보아 HSL의 경구투여는 NC/Nga mice 병태모델에서 면역조절 작용 및 아토피 병태생리적 요인의 개선을 통한 항아토피 작용을 나타냈다. 따라서 HSL은 아토피 피부염에 유용한 천연자원으로서 기대된다. 중심단어: 아토피 피부염, 어성초, 삼백초, 자초, DNCB, NC/Nga mice

Atopic dermatitis (AD), a chronic inflammatory skin disease, is characterized by elevation of blood IgE level and over production of Th2 cytokines and chemokines. However, the precise etiology of AD has been unclear. A suitable animal model is required to explore the pathogenesis and treatment of AD. The objective of this study was to evaluate the inhibitory effects of HSL, the mixture of HCH, LEH and SCH in the equal ratio, an the development of atopic dermatitis like skin lesions induced by the repeated epicutaneous challenge of dinitrochlorobenzene (DNCB) on the dorsal skin of NC/Nga mice, an animal model of atopic dermatitis. The efficacy of HSL was evaluated by the histopathological examination, blood IgE level, measurement of transepidermal water loss (TEWL), scratching behavior and clinical severity score. The oral administration of HSL for 4 weeks from the day of application of DNCB suppressed significantly the clinical severity score, TEWL and scratching behavior and blood IgE level. Histopathologic analyses revealed that infiltration of inflammatory cells, especially degranulated mast cells and eosinophils, thickening of the epidermis and prominent hyperkeratosis, were significantly reduced in HSL treated groups. The in vitro assays were carried out using HaCaT cell line to evaluate the effects of HCH, SCH, LEH and HSL on the proliferation of HaCaT, expression of matrix metalloproteinases 2 and 9 (MMP-2 and MMP-9) and production of thymus and activation regulated chemokine (TARC). The treatment of HCH, SCH and HSL for 24 hours dose dependently inhibited the proliferation of HaCaT. Similarly, the pretreatment of them dose dependently inhibited tumor promoting agent (TPA) and transforming growth factor-b (TGF-b) induced expression of MMP-9. Also, HSL treated group suppressed the secretion of TARC in HaCaT cells stimulated with 10 ng/ml of IFN-g and 10 ng/ml of TNF-a. These results suggested that oral administration of HSL, a traditional herbal medicine, has suppressive effects on AD like skin lesions in NC/Nga mice by the modulation of the immune system. Therefor, HSL could be regarded as a natural therapeutic option for the treatment of AD.
