미호종개의 서식처 및 서식처 특성을 조사한 결과 미소서식처를 가지며 0.6 mm 이하의 가는 모래 지형과 느린 유속, 양호한 수질에서 서식하였고, 동서종으로 저서성 어류들이 확인되었다.
미꾸리과와 종개과의 분자계통학적 분석을 위하여 미토콘드리아 COX1과 핵 RAG1 영역의 염기서열을 분석하여 분자계통학적 근연관계를 파악하고자 하였다. COX1과 RAG1의 염기서열은 각각 1,551과 855 bp로 분석되었다. Maximum Likelihood와 Bayesian 분석 결과 COX1과 RAG1 유전자 간에 차이를 보여 Cobitis 속과 Iksookimia 속은 다계통군으로 나타났다. Cobitis 속은 RAG1 분석에서 단계통군을 형성하였으나, 미토콘드리아 COX1에서는 다계통군을 형성하는 결과를 보였다. 최근에 Iksookimia 속에서 Cobitis 속으로 전속되었던 미호종개는 Cobitis 속과 가까운 근연관계를 보였으나, 분류학적인 구분은 여전히 모호하였다.
미호종개 3개 집단(갑천, 백곡천, 지천)의 유전 다양성 및 유전적 구조를 AFLP 분석을 통해 조사하였다. 3종의 primer 조합형을 이용한 AFLP 분석에서 각 집단으로부터 106, 107, 104개의 밴드가 생성되었으며, 집단 내 다형 밴드의 출현 빈도는 3개 집단에서 21.49~24.52%로 유사하게 나타났고, 이형접합율 (0.0674~0.0837) 및 유전적 다양도(0.0756~0.0871)는 매우 낮은 값을 보였다. 집단간 유전적 거리 및 유전적 상동성 분석 역시 유사한 결과를 나타내어 본 연구에서 분석된 미호종개 3개 집단은 유전적으로 매우 밀접한 근연관계를 나타내었다. 비록 pairwise Fst값은 매우 낮았지만 세 집단은 유전적 거리와 밀접한 관련성을 보이면서 유전적 분화가 일어나고 있었다. 본 연구는 미호종개의 유전적 다양성 및 분화에 대해 처음으로 보고된 연구이며, 미호종개의 보존을 위한 기초 유전 정보를 제공할 것으로 기대된다.

미호종개의 산란행동을 관찰하기 위하여 HCG 호르몬을 주사하여 실험실 내 100 l 유리 수조에 수용하였으며, 실험에 사용한 수컷의 전장은 82±5 mm, 암컷의 전장은 65±3 mm이었다. 산란행동 관찰 결과 주사 후 9시간까지 유영, 접근, 유혹행동을 보여 산란전 단계로 구분되었다. 9시간 경과 후부터는 유혹, 접촉을 반복하였으며, 10시간 경과 후부터는 산란을 시작하여 약 2시간 동안 산란이 이루어졌으며, 회당 80~120개의 난을 산란하였고, 산란은 약 15~25회 반복하여 산란 단계로 구분하였다 12시간 경과 후에는 산란행동이 더 이상 관찰되지 않아 산란후 단계로 구분하였고, 미호종개의 산란행동은 크게 3단계로 구분되었다. 호르몬 유도에 의한 산란행동이 보고된 미꾸리와 산란전 단계는 매우 유사하였으나, 산란 단계에서 수컷이 암컷의 몸을 감는 행동, 복부를 압박하는 행동에서 큰 차이를 보였다. 또한 미호종개의 성비를 분석한 결과 1 : 1로 나타나 방류 시 참고자료로 활용이 가능할 것으로 판단된다.

This study was aimed to investigate population structuring of I. choii and provide baseline data for its artificial propagation which are prerequisite for conservation and restoration. It includes 1) characteristics of natural habitats, 2) molecular phylogeny based on mitochondrial and nuclear genes, 3) assessment of genetic diversity among populations, 4) artificial propagation and 5) ecological ex situ restoration and monitoring of released stocks.
The extant and candidate habitats of I. choii are characterized by small and limited areas with 20-30 cm/sec in water currents, upper frequency 75%, sediment particles less than 0.6 mm and good water quality. The ichthyofauna and community of fish are composed of demersal species.
Phylogenetic relationships of fish species belonging to the families Cobitidae and Balitoridae were investigated based on mitochondrial cytochrome c I (COX1) and nuclear-encoded recombination activation gene 1 (RAG1) genes. Nucleotide sequences of COX1 and RAG1 of 19 species consisted of 1,551 and 885 bp, respectively. The Maximum Likelihood and Bayesian inference analyses based on the two genes showed considerably different tree topologies. RAG1 tree revealed that all Cobitis species formed a monophyletic group, while COX1 tree showed their polyphyletic nature. I. choii, which was recently transferred to the newly erected genus Iksookimia from Cobitis, was phylogenetically affiliated to Cobitis species with high statistical supports, indicating its taxonomically unstable status.
Genetic diversity and population structuring within or among three Stream populations of I. choii (Gab, Baekgok and Ji Streams of the Geum River) were assessed by amplified fragment length polymorphism (AFLP) fingerprinting technique. The AFLP analysis using three primer combinations of selective primer pairs generated 104 to 106 DNA bands, and percent polymorphic bands ranged 21.5 to 24.5%. Heterozygosity and genetic diversity within or among populations were quite low for all of these populations with average values ranging from 0.067 to 0.084 and from 0.076 to 0.087, respectively. Analyses of pairwise distance and genetic similarity among populations also revealed the similar results with very low genetic differentiation one another. Although pairwise Fst values were very low, our data clearly indicated distinct genetic differentiation among the three populations. This is the first report concerning the genetic diversity and differentiation of I. choii, and will provide basic genetic information that should facilitate attempts to conserve this species.
Meanwhile, twenty polymorphic microsatellite loci were isolated and characterized in 67 individuals from three populations. The number of alleles per locus ranged from 8 to 33. Within populations, the observed and expected heterozygosities ranged from 0.59 to 1.00 and from 0.56 to 0.96, respectively, indicating its usefulness in population genetics studies. The 20 microsatellite loci were used to examine genetic variation between and within three I. choii populations. Average allele numbers were 11.3 to 12.6 and observed and expected heterozygosity were 0.822 to 0.859 and 0.818 to 0.871 respectively. Cross-species amplification of the loci was tested in six species of the family Cobitidae, and 18 loci were amplified in two or more related species.
Parentage of five mating groups were investigated using nine microsatellite loci to carry out genotyping. The result indicated that the offsprings showed the simple Mendelian inheritance patterns. Thus the microsatellite markers developed in this study will be useful for stock identification for I. choii recovery plan.
The egg development and early life history of I. choii were investigated to provide baseline data for development of its conservation strategies. The eggs from the females were obtained by injecting 10 IU/g of human chorionic gonadotropin (HCG) and inseminated by wet method in the laboratory. The fertilized eggs were 1.1-1.3 mm in diameter and had no oil globules. The embryo began to hatch about 24 h after fertilization under water temperature of 25°C. The newly-hatched larvae were 2.5-2.7 mm in total length, and their mouth and anus were not yet opened. Six days after hatching, the post larvae were 4.7-5.4 mm in total length, and tip of the caudal notochord was flexed 45° upward. They entered the juveniles stage when all fin-rays were formed at 35 days after hatching, and their total length were 20-22 mm.
The spawning behavior of I. choii was studied in aquaria (150 l) to develop an artificial breeding technique. Three mating groups (female : male 1 : 4) was induced by HCG injection for spawning in dosage of 50 and 20 IU/g, respectively. Spawning behavior was divided into 1) pre-spawning stage: swimming, approaching and chasing (0-9 h after injection), 2) spawning stage: chasing, beating and spawning (10-12 h after injection); the number of eggs was 80-100 eggs per beating and the beating repeated 15-25 times. 3) post-spawning stage (12 h after injection). This spawning behavior in the pre-spawning stage of I. choii was very similar to Misgurnus anguillicaudatus, but the spawning stage was different from M. anguillicaudatus in that males of I. choii twists their body to embrace females. Effect of temperature in egg development and hatching of I. choii was examined in different temperature ranges (22, 25 and 28°C). The required time from fertilized embryos to hatching were 2,259, 1,569 and 1,423 min respectively.
For successful ex situ restoration of I. choii ichthyofauna water quality and sediment properties for candidate release sites were surveyed prior to releasing. I. choii fries were artificially propagated and more than 4,200 individuals were released to the candidate release site. After one year later sexually matured females were recaptured, implying the successful restoration of I. choii at the released site.
