가축의 질병 예방과 가축 생산성을 높이기 위하여 그 동안 생장 촉진제로서의 항생제를 지속적으로 사용하여 왔다. 그러나 이러한 항생제의 남용으로 인하여 축산물에 잔류되어 인체에 전이되고 결과적으로 알레르기 등의 질병을 일으키는 문제와 인체의 내성을 증가시켜 질병 발생 시 약물의 치료 효과를 감소시키는 등의 문제가 발생된다. 국내에서는 2005년도에 항생제에 대하여 25개 품목으로 대폭 그 범위를 감소시켜 왔고, EU(유럽연합)는 이보다 더 강도 높은 조치를 취하였으며, 2006년부터 가축의 치료목적 외에 성장촉진용 항생물질의 사료 첨가를 전면 금지시키고 있는 상황이다. 미생물, 효소 및 식물 추출물 등 천연자원을 사료에 첨가하는 가축 사육은 기존의 화학 항생제와 환경오염을 줄일 수 있으며, 또한 가축 및 인체에 있어서 병원성 세균들에 대한 내성을 유발하지 않고 성장을 촉진시킬 수 있는 항생제 대체제이다. 따라서 Salmonella 및 대장균 등 병원성세균에 대한 항균효과, 육계의 성장 촉진 및 면역증강 기능 효과를 통하여 질병 예방 및 생산성 향상 등의 기능을 수행할 수 있는 천연 물질을 발굴하여 이용할 필요가 있다. 본 연구는 닭 사료 첨가용 생균제를 개발하기 위해 닭 분변으로부터 면역기능이 우수한 프로바이오틱스 미생물을 선별하고, 선별된 미생물의 프로바이오틱스로서의 균주의 특성을 다음과 같이 연구하였다. 첫 번째는 면역기능이 우수한 프로바이오틱스 미생물을 분리하기 위해 닭 분변을 채취하여 생성된 모든 집락을 순수 분리하였으며, 순수분리된 균주는 균 집락 형태 관찰을 통해 내산성이 있다고 판단되는 균주를 총 46 균주 분리하였다. 그 중 면역활성이 높은 9 균주 (FC113, FC222, FC322, FC421, FC422, FC511, FC621, FC721, FC812 균주)를 우수 균주로 정하고 실험에 사용하였다. API 50 CHL kit로 간이 동정한 결과, FC322 균주를 제외한 모든 균주가 Lactobacillus salivarius로 동정되었다. 이 균주들은 인공 위액과 인공 담즙산에 내성이 있는 것으로 나타났으며, 특히 FC113, FC222, FC421, FC522, FC621 균주는 인공 위액에 대한 내성이 강한 것으로 나타났으며, FC222, FC511, FC721 균주는 인공 담즙산에 내성이 강한 것으로 나타났다. 닭에서 분리한 8종의 병원성 세균에 대해서 모든 균주가 항균활성이 있는 것으로 나타났다. 또한 닭에서 많이 이용되고 있는 10종의 항생제에 대해 균 특이적으로 내성이 있는 것으로 나타났다. 두 번째는 분리된 균주 중 면역활성이 가장 높은 3 균주 (FC222, FC621, FC812 균주)를 선택하여 프로바이오틱스로서의 특성을 연구하였다. 이들 균주는 16S rRNA로 유전학적 분석을 한 결과, 모든 균주가 L. salivarius로 동정되었다. 면역활성을 확인하기 위해 면역세포인 대식세포가 활성화되었을 때 생성되는 사이토카인 (인터루킨-1α와 TNF-α)의 생성을 측정하였는데, 인터루킨-1α는 각각 233.7±75.86, 293.4±131.90, 404.9±98.05 pg/mL의 높은 생성량을 보였으며 TNF-α는 각각 384.6±17.81, 135.8±39.49, 140.8±20.48 pg/mL의 높은 생성량을 보이는 것으로 나타났다. 이 밖에 19가지 효소에 대한 효소 활성을 살펴본 결과, 균 특이적으로 leucine arylamidase, naphtol-AS-BI-phosphohydrolase, α-galactosidase 등의 효소를 생산하는 것으로 나타났고, 반면에 간에서 해독 작용을 하는 글루쿠론산을 분해하여 암을 발생시키는β-glucuronidase는 모든 균주에서 활성이 나타나지 않았다. 또한 세포 표면의 소수성과 장 상피 세포를 이용하여 장 부착능을 알아보는 연구를 진행하였다. 세 번째는 식물체에 들어 있는 유기태 인을 분해하는 효소인 phytase를 생성하는 균주 (FC113)을 선택하여 이 균주의 프로바이오틱스로서의 특성을 연구하였다. FC113 균주의 phytase 활성은 403.6 U으로 나타났다. 이 균주 또한 16S rRNA로 유전학적 분석을 한 결과 L. salivarius로 동정되었다. L. salivarius FC113의 면역활성은 인터루킨-1α가 175.5±36.40 pg/mL, TNF-α가 353.5±61.79 pg/mL의 생성량을 나타냈다. 이 균주는 인공 위액에 내성이 강한 것으로 나타났으며 인공 담즙산에서는 2 log 균수의 감소가 있었다. L. salivarius FC113은 12 종류의 효소를 생산하는 것으로 나타났으며, 그 중에서 leucine arylamidase, naphtol-AS-BI-phosphohydrolase를 가장 많이 생산 하였으며 acid phosphatase와 α-galactosidase도 10 nmol을 생산하는 것으로 나타났다. 반면, 장 상피세포에 부착하는 능력은 0.37%로 낮은 것으로 나타났다. 본 연구 결과, 이들 균주는 프로바이오틱스 균주로서 요구되는 조건을 충족시킨다는 것을 알 수 있었으며, 면역강화 닭 사료용 첨가제로 이용 가능성을 확인할 수 있었다.

The principal objective of this study was to screen and select acid-tolerant Lactobacillus strains from chicken feces, feeds, and other sources. Fourty six strains evidencing acid tolerance (pH 3.5) were isolated in this study. Among them, nine strains exhibited marked immunostimulatory effects. Therefore, nine candidate strains were characterized for probiotic use. In order to evaluate macrophage activation, NO production was measured using RAW 264.7 cells. In particular, three strains (FC812, FC222, and FC113) evidenced the highest levels of NO production measured at 38.39±20.01, 35.06±27.73, and 33.88±15.99 μM, respectively, at a concentration of 108 CFU/mL. The majority of strains, with the exception of strain FC322, evidenced marked resistance to artificial gastric juice (pH 2.5 with 1%(w/v) pepsin). Additionally, strains FC222, FC421, FC511, and FC721 were highly resistant to artificial bile acid (0.1%(w/v) oxgall), whereas strains FC113, FC322, FC422, FC621, and FC812 were the least resistant to bile. All nine strains exerted antimicrobial effects against chicken-related pathogens. Additionally, all nine strains were found to be resistant to several antibiotics. The isolated strains, except for strain FC322, were tentatively identified as Lactobacillus salivarius, using an API 50 CHL kit. Also, the amount of TNF-α in concentration of 107 CFU/mL from L. salivarius FC222, FC621, and FC812 was approximately 384.6±17.81 pg/mL, 135.8±39.49 pg/mL and 140.8±20.48 pg/mL, respectively. The L. salivarius strains were survived highly in gastric conditions (pH 2.5, 1% (w/v) pepsin, and 0.1% (w/v) oxgall). From the API ZYM kit, the L. salivarius strains were shown to produce enzymes including leucine arylamidase, naphtol-AS-BI-phosphohydrolase, and α-galactosidase, but β-glucuronidase was not produced. L. salivarius FC222 and FC812 had high affinity for n-hexadecane (98.2±0.48% and 90.7±0.75%), whereas L. salivarius FC621 had lowest hydrophobicity at 19.5±3.77. All L. salivarius strains exhibited low adhesion levels (0.02-4.30%). These results except adhesion level demonstrate the potential of the three L. salivarius strains as a health-promoting probiotics.
