W. cibaria JW15는 유산균으로 잘 알려진 Lactobacillus plantarum GG 균에 비해 IL-1β, TNF-α 및 IL-12 의 생산량을 높은 수준으로 유도하였다. 또한 W. cibaria JW15로 활성화된 CD4+ T cell들의 표면에 CD25와 CD69의 발현을 증가 시켰고 IFN-γ의 생산을 강하게 유도 하였다. 본 연구에서는 W. cibaria JW15가 사람면역세포에서 NF-κB, ERK, p38 및 JNK 등 여러 신호전달 경로를 활성화 시키는 것을 확인하였다. 또한 이러한 연구결과를 바탕으로 W. cibaria JW15의 면역반응 향상여부를 확인하기 위해 마우스의 L. monocytogenes 감염모델을 이용하여 실험을 진행 하였다. Recent studies demonstrated immunomodulatory effect of LAB both in human and mice and suggested that LAB can used to prevent or treat various diseases including microbial infections and inflammation-mediated diseases. The present study aims to evaluate the immunomodulatory potential of LAB isolated from Kimchi to be used as probiotics to enhance immune responses against microbial infection. Initially, we tested 7 LAB selected from previous studies for their ability to induce cytokine production, T cell activation, and regulatory T cell induction using human peripheral blood mononuclear cells (PBMC) cultures, and selected Weissella cibaria JW15 for further studies. W. JW15 is Gram positive short rod bacteria and one of the most common LAB isolated from Kimchi. Recent studies demonstrated their potential as probiotics. Modulation of host immunity is an important potential mechanism by which probiotics confer health benefits. However, very little information about immunomodulatory activities of W. cibaria is available. It is investigated the effect of W. cibaria JW15 on the immune function using human PBMC. In addition, signaling pathways affected by W. cibaria JW15 were also explored. W. cibaria JW15 induced higher levels of IL-1β, TNF-α, and IL-12 compared to Lactobacillus plantarum GG, which is the most well-known probiotics strain. Furthermore, W. cibaria JW15 activated CD4+ T cell, evidenced by increased CD25 and CD69 expression on the surface of these cells, and triggered IFN-γ production strongly. These immunostimulatory activities induced by W. cibaria JW15 are likely mediated by multiple signaling pathways, such as NF-κB, ERK, p38, and JNK. In order to determine whether W. cibaria JW15 boosts immune responses to bacterial infection, mice model of Listeria monocytogens infection was used. W. cibaria JW15 was able to partially enhance the clearance for Listeria from Liver and spleen boots cytokine production in response to Listeria infection. In conclusion, the present study demonstrated that W.cibaria JW15 exerts potent immunostimulatory effects on human PBMC and mice immune cells and protects mice from Listeria monocytogens infection.
