Although the pathogenesis of this disease remains still obscure, several researchers have report that death of RPE cells caused by excessive accumulation of A2E, a component of ocular lipofuscin, and alteration to photoxidative forms of A2E is crucial determinants of AMD. At the present time, there is no medical or surgical treatment for person with AMD. Some clinical studies demonstrated the benefits of nutritional supplements, such as beta carotene and omega-3 for prevention of AMD. However, understanding of the role of supplements is not perfect yet. In this study, extract of Vaccinium uliginosum L.(VU) that has potent antioxidant activity was evaluated as effective preventive supplement for AMD using cultured human RPE cells (ARPE-19 cells) and AMD animal model induced by high fat diet and UV A irradiation. In the in vitro test, VU reduced significantly RPE cell loss induced by A2E photooxidation in a dose-dependent manner, and inhibited significantly accumulation of A2E within ARPE-19 cells. Also, the same tests were performed on three fractions (flavonols, anthocyanins, carbohydrates) isolated from VU to find the most active fraction. As the results, protective effect against ARPE-19 cells loss showed the most effect in the flavonols fraction and inhibitory effect on accumulation of A2E was the highest in the anthocyanins. In the in vivo test, retina of mice was examined histologically. In observation on photoreceptors using light microscopy, it was found by counting the number of photoreceptor nuclei in the outer nuclear layer(ONL) and measuring thickness of the layer that VU protects photoreceptor cells. Transmission electron microscopy analysis revealed that VU blocked the accumulation of lipofuscin pigment-granules induced by high fat diet and UV A light irradiation. In conclusion, we might know that VU protect RPE cells against cell loss by inhibiting photooxidation and accumulation of A2E in the in vitro test using ARPE-19 cells. In the in vivo test, it was demonstrated that VU markedly reduced accumulation of lipofuscin within RPE cell and the loss of photoreceptor. In the study, it was presented that VU may be developed as a health functional food or medicine.
