Raw264.7 세포를 이용하였고, LPS 처리 2시간 전에 MG-132 (10μmol/L) 및 NH4Cl (10μmol/L)를 전처치 및 LPS (1μg/ml)를 처리하였다. PU.1 과 유비퀴틴의 단백질-단백질 상호작용을 확인하기 위하여 면역 침강 및 웨스턴 블럿을 시행하였으며, proteasome, lysosome 억제에 대한 대식세포에서의 염증반응의 변화를 확인하기 위해 nitric oxide assay 및 prostaglandin(PG) assay를 시행하였다. Raw264.7 세포에서 LPS 자극에 의해 유비퀴틴-PU.1의 발현이 증가되었고, MG-132 및 NH4Cl 의 전처치로 LPS에 의한 유비퀴틴-PU.1 의 발현이 더욱 증가됨을 확인할 수 있었다. Proteasome, lysosome 억제제의 전처치에 의해 nitric oxide 의 발현양에 변화가 있었으며, Rac2-/- null mice의 복막의 대식세포에서의 LPS에 대한 PGE2의 생성이 wild type에 비하여 감소되었고, 골수 대식세포에 lactacysitn을 처리한 경우 LPS에 대한 PGE2의 생성은 증가되었다. 본 연구에서는 대식세포의 염증반응의 기전에 유비퀴틴-PU.1이 역할을 하며, PU.1의 분해에 proteasome 및 lysosome이 주로 관여한다는 것과, 유비퀴틴화된 단백질들의 분해의 억제가 NO synthase, NADPH-oxidase, cyclooxygenase-2의 활성에 변화를 일으켜 대식세포의 염증과정에 중요한 역할을 함을 알 수 있었다.

However, the direct control mechanism of ubiquitin for PU.1 is not well known. This study was conducted to investigate the activation of PU.1, a transcription factor, by LPS stimulation in monocytes/macrophages and to determine the role of ubiquitin. Raw264.7 cells were pretreated with MG-132 (10 μmol/L) and NH4Cl (10 μmol/L) 2 hours before treatment with LPS (1μg/ml) for 4, 12 and 24 hours. We performed immunoprecipitation and Western blot to evaluate protein-protein interactions between PU.1 and ubiquitin and NO assay/prostaglandin (PG) assay to determine the effect of the increased ubiquitin-PU.1 in the inflammatory process of macrophages. In Raw264.7 cells, ubiquitin-PU.1 expression by LPS stimulation LPS treat was increased, and after pretreatment with MG-132 and NH4Cl ubiquitin-PU.1 expression was more increased. The PGE2 production by LPS stimulation was decreased in peritoneal macrophages from Rac2-/- null mice compared with wild type, and PG E2 production by LPS stimulation after pretreatment with lactacystin was increased in bone marrow macrophages. In this study, we confirmed that ubiquitin-PU.1 is the key transcription factor in the inflammatory mechanism of macrophages and proteasome and lysosome are related to the protein degradation of ubiquitin-PU.1. In particular, ubiquitination may be important in macrophage-induced inflammation through alteration in activation of NO synthase, NADPH-oxidase and cyclooxygenase-2.
