Abnormally hyperphosphorylated tau proteins that accumulate as neurofibrillary tangles are observed in the AD, which are now categorized as tauopathies. It has recently shown that glycogen synthase kinase 3beta(GSK3beta) induced the hyperphosphorylation of tau resulting in impaired learning and memory. In the present study, GSK3beta-targeting ELISA was established and GSK3beta inhibitor investigated its inhibitory effects on GSK3beta-mediated tau phosphorylation. In the current study, it was found that a flavonoid morin effectively inhibited GSK3beta activity, and blocked GSK3beta-induced tau phosphorylation in vitro. In addition, protective effects of morin against Abeta-induced toxicity in human neuroblastoma SH-SY5Y cells, and morin modulated SH-SY5Y cells against Abeta-induced tau hyperphosphorylation. Furthermore, the current study evaluated the protective effects of morin on tau hyperphosphorylation in 3xTg-AD mice. Elevated tau hyperphosphorylation and paired helical filaments were observed in the hippocampus of 3xTg-AD mice. Interestingly, morin treatment significantly decreased accumulation of tau hyperphosphorylation and paired helical filament tau in 3xTg-AD mice. Therefore, modulating GSK3beta can be the beneficial target to AD associated with hyperphosphorylated tau proteins, and the current study suggests that morin can be a potential therapeutic agent to modulate the overactivated GSK3beta??in pathophysiological condition of AD.
