Recently, it has been reported that mitochondrial dysfunction is associated with various pathological condition, especially cancer. However, the underlying mechanisms for tumor malignancy induced by mitochondrial dysfunction have not been fully understood. In this study, the effects of mitochondrial dysfunction on tumor malignancy were initially investigated using mitochondrial-depleted ρ0 cell model. Firstly, ρ0 cells were constructed from Hep3B hepatocarcinoma cell line by treating ethidium bromide. ρ0 cells showed the properties of rapid growth, high invasiveness and aggressive migration rather than their parental Hep3B. And the cells had also obtained activities for angiogenesis, sorafenib-resistance and cancer stem cell (CSC). Increased tumor growth factor-β (TGF-β) in ρ0 cells promoted tumor malignancy via canonical Smad-dependent signaling pathway. Activated TGF-β signaling pathway was also induced CSC phenotype via induction of sex determining region Y-box 2 (Sox2) expression. These results support that tumor malignancy might be accelerate by mitochondrial dysfunction. Next, the possibility of the suppression of tumor malignancy was examined through regulation by mitochondrial dysfunction. Under hypoxia, increased reactive oxygen species (ROS) can induce tumor angiogenesis by stabilization hypoxia-inducible factor-1 α (HIF-1α). In this study, melatonin, a small lipophilic molecule secreted primarily by the pineal gland, destabilized hypoxia-induced HIF-1α protein levels in the HCT116 human colon cancer cell line. This destabilization of HIF-1α resulted from the antioxidant activity of melatonin against ROS induced by hypoxia. Moreover, under hypoxia, melatonin suppressed HIF-1 transcriptional activity, leading to a decrease in VEGF expression. Melatonin also blocked in vitro tube formation and invasion and migration of human umbilical vein endothelial cells induced by hypoxia-stimulated conditioned media of HCT116 cells. These findings suggest that melatonin could play a pivotal role in tumor suppression via inhibition of HIF-1-mediated angiogenesis. Taken together, mitochondrial dysfunction plays an important role in the tumor malignancy and could be a novel therapeutic target to cancer therapy.
