흰가루병 저항성 참외품종을 육성하기위해 수집된 멜론류 유전자원들에 대해 포장에서의 흰가루병 저항성 여부 및 원예적 특성을 평가하였다. 이들 중에서 흰가루병에 대해 저항성을 나타내는 계통을 찾아 원예적으로 우량한 이병성계통과 교배하여 F2 분리집단을 육성하고 흰가루병 저항성 유전자와 연관된 DNA marker를 개발하여 흰가루병저항성 참외품종을 육성하기 위한 시스템으로 활용하고자 하였다.
수집된 멜론의 유전자원에서 흰가루병에 저항성이 있는 계통을 찾아 참외육성계통에 교잡을 하여 분리선발을 하는 과정에서 기존의 유묘기 흰가루병 접종에 의한 선발방법이 아닌 CmPMR7 marker에 의한 흰가루병 저항성 개체를 검정하여 선발함으로써 환경적 영향에 의한 시행착오를 줄이고 보다 정확하고 빠른 방법으로 흰가루병 저항성 참외 계통을 육성하고 육성된 계통을 이용하여 흰가루병 저항성 고품질 참외 시판종을 육성한 결과들을 요약하면 다음과 같다.
1. 수집된 멜론류 유전자원 166점, Netted melon 57계통, honeydew melon 45계통, Oriental melon 63계통에 대한 포장의 흰가루병 저항성 여부 및 과형지수, 과장, 과경, 과중, 과육두께, 육질의 경도, 종자무게 등 주요 원예적 특성에 대해 평가하였다. 흰가루병의 저항성검정에서 완전저항성을 나타내는 계통은 네트멜론에서 “ADS”와 “RA” 2계통이 확인되었다. 수집된 계통의 원예적 특성을 검정한 결과 네트멜론은 Honeydew멜론과 참외에 비해 과피색이 대부분 녹색을 나타내 과형도 구형에 가까운데 비하여 Honeydew 멜론과 참외는 과형이 구형에서부터 긴 타원형까지 다양하였고 과피색의 경우에도 네트멜론보다는 녹색, 황색, 백색, 황피얼룩 등으로 많은 원예적 다양성을 보여주었다. 특히 과육의 경도는 멜론의 경우 1.2 kg/㎠ 내외였으나
한국인이 선호하는 참외육성계통은 2.02 kg/㎠ 수준으로 그 차이가 심하였다. 화기 구조에 있어서 멜론과 재래종참외는 대부분 양전화로 나타났지만 참외육성계통은 단성화로 나타났다.
2. 흰가루병 저항성 분자표지를 개발하기 위하여 흰가루병 저항성친과 이병성친을 교배하여 F2 분리집단을 육성하고 흰가루병을 접종하여 분리비를 분석한 결과 1:2:1로 나타나 멜론의 흰가루병 저항성은 우성 또는 불완전 우성인 하나의 주동인자와 소수의 미동인자에 의해 조절되는 것으로 판단되었다.
3. 멜론의 흰가루병 저항성에 관여하는 주동인자와 연관된 분자표지 8점을 개발하였고, 이중 CmPMR7과 CmPMR10이 흰가루병 저항성유전자와 가장 가까이 연관된 것으로 나타났고, 이들 분자표지를 이용하여 국내 참외시판품종들에 대해 분석한 결과 모든 품종들이 이병성 유전자형인 rr로 나타나 이들 분자표지는 흰가루병 저항성 멜론 및 참외품종 육성과정에서 선발표지로 활용될 수 있음을 확인하였다.
4. 세대를 진전시키는 과정에서 흰가루병 저항성개체를 선발하고자 분자표지를 활용하였고, 저항성인 RR과 중도저항성인 Rr을 정식하여 포장검정을 병행한 결과 분자표지 분석결과와 일치하였다.

6. 과피색은 BC2F1에서 모계(SW3)와 유사한 진한 황색을 나타내었고 과육의 경도는 BC2F3 이후 세대에서 “SW3” 계통과 유사하게 나타났으며, BC2F6에서 표현형과 과 품질이 참외 “SW3” 계통과 유사한 “San1”, “San2”, “SanY” 등 3계통을 개발하였고, “San2” 계통의 과육경도를 더욱 향상시키기 위해 BC2F5 세대에서 “SW3” 계통을 1회 더 여교배하여 “San2” 계통의 과육경도를 2.0 kg/㎠ 수준으로 향상시킨 “SanB” 계통을 개발하였다.
7. 분자표지를 활용하여 육성된 흰가루병 저항성 4 계통을 이용하여 교배조합
을 작성하였고 농우바이오 육종연구소에서 F1 조합성능검정을 실시하여 우수한 결과를 얻은 2 조합(G2Gink×San1, G2Gink×SanY)을 참외 주산지인 경북 성주, 칠곡 등에서 농가실증시험을 실시한 결과 흰가루병에 대해 저항성
을 보이는 고품질 참외로 인정되어 각각 ‘국보꿀‟참외(G2Gink×San1), „오복플
러스꿀‟참외(G2Gink×SanY)라는 명칭으로 생산.판매 신고 하여 상업화하였다.

Powdery mildew caused by Podosphaera xanthii is a major disease that can reduce yield (fruit yield and/or size) and market quality (flavor, color, storability, etc) in oriental melons. Successful control of powdery mildew in oriental melon is critical to ensure leaves remain healthy until fruit maturity and obtain high sugar content, which results in good fruit quality. Oriental melon growers still rely on chemical treatments to control powdery mildew. However, the use of fungicides over many decades has resulted in the development of P. xanthii resistance to many chemical compounds, which have therefore lost their efficacy. Also, increasing concerns for public health have motivated breeders and growers to seek alternative strategies to control plant diseases. The development of new resistant cultivars appears to be the most eco-compatible
method for powdery mildew disease control. Hence, diverse genetic resources of C. melo genotypes were collected to select and characterize new resistance resources. Among these collected genotypes, only two showed complete resistance to powdery mildew disease in field screening. One of these two genotypes was used to transfer resistance genes into elite breeding line of oriental melon. Complete resistant line "ADS" was bred from the resistant European netted melon variety and this line was used to develop resistant oriental melon cultivars.
The melon breeding line "ADS" was used not only as a resistance source for oriental melon breeding but also as a suitable genotype to study the genetic and molecular mechanisms underlying resistance to the pathogens. Nevertheless, several factors limit studies on the genetic control of powdery mildew resistance because of several reasons like the genetic variability in pathogen populations, the lack of consistency in experimental assays, and the influence of environmental conditions on the expression of resistance. Moreover, artificial inoculation methods used to assess powdery mildew resistance are laborious and time-consuming, thus hampering the development of new commercial cultivars.
For breeding of powdery mildew resistant oriental melon, DNA markers tightly linked to a given resistance gene are useful tools to avoid the above mentioned drawbacks and also provide significant advantages over traditional phenotypic screening. Marker-assisted selection (MAS) is a rapid, relatively inexpensive method, and it is not hampered by pathogen unavailability. In addition, there are no environmental limitations since MAS can be performed off-season, and it also permits the simultaneous screening for several characteristics. To breed powdery mildew resistant oriental melon cultivars efficiently, the author tried to develop DNA markers linked to the resistance gene and used these markers as a tool for MAS. Following are outlines for this
research to develop DNA markers and breed resistant oriental melon cultivars.
1. Genetic resources of 166 C. melo genotypes (57 netted melon, 45 honeydew melon, 63 oriental melon) were collected and evaluated for powdery mildew resistance and other horticultural characteristics (fruit weight, fruit length, fruit diameter, FL/FD ratio, flesh firmness, fruit skin color, flesh color, thickness of flesh, seed weight, flower type). Among these 166 genotypes, only two genotypes (ADA and RA) showed complete resistance to powdery mildew disease.
2. Marker assisted breeding system was adapted to breed powdery mildew resistant cultivars efficiently and accurately. A segregating F2 population was obtained by crossing resistant inbred line "ADS", derived from European netted melon variety, with susceptible inbred line "SW3" to elucidate the inheritance nature of the resistance and to develop DNA markers linked to the resistance locus. This F2 population was evaluated for response to the pathogen P. xanthii under the natural field condition. Distribution of powdery mildew incidence in 262 F2 plants agreed with the segregation pattern of monogenic inheritance (incomplete dominant gene) indicating that the resistance to powdery mildew is controlled mainly by one major gene.
3. DNA fingerprinting techniques combined with bulked segregation analysis identified eight markers linked to the resistance locus. These eight markers associated with powdery mildew resistance were converted into SCAR markers and named as CmPMR. Out of these markers, two markers, CmPMR7 and CmPMR10 were identified as most closely linked to the resistance locus within 2cM interval and showed susceptible homozygous genotype "rr" in all the commercial oriental melon cultivars used in this study. These SCAR markers were used as reliable selection markers to evaluate powdery mildew resistance accurately in the subsequent breeding system.
4. DNA marker linked to the powdery mildew resistance was used as a selection tool during all the steps for selection of resistant plants in each generation. Those selected plants having resistance homozygous or heterozygous genotype for DNA markers in seedling stage were selected, and the plants were transplanted in a plastic house and compared with the incidence of powdery mildew disease under the field conditions (artificially inoculated with collected spores from infected leaves of susceptible control plants). All the plants selected by DNA marker genotype data showed complete or incomplete resistance in field condition, indicating that results from marker genotype and field screening were identical.
5. Crosses between netted melon and oriental melon to introduce powdery mildew resistance of netted melon into oriental melon resulted in normal seed production and germination, indicating that there were no problems in cross pollination between these subspecies in C. melo .
6. Fruit color in BC2F1 generation showed deep yellow similar to oriental melon breeding line "SW3", and flesh firmness was similar to "SW3" after BC2F3 generation. All the horticultural characteristics and fruit quality were similar to "SW3" in BC2F6 generation, so that 3 new powdery mildew resistant breeding lines were selected in this generation and named as "San1", "San2" and "SanY". However, new breeding line "San2" exhibited a slighty lower value of flesh firmness compared to "SW3". One plant in BC2F5 generation was backcrossed to "SW3" one more time to improve flesh firmness. From this subsequent crossings, one more breeding line named as "SanB" with improved flesh
firmness up to the level 2.0 kg/ ㎠similar to "SW3" was bred.
7. Four oriental melon cultivars resistant to powdery mildew disease were developed by crossing inbred lines developed in this study using DNA marker system and elite inbred lines developed in previous breeding program. Among these 4 lines, 2 showed excellent horticultural characteristics and powdery mildew resistance in field test. These two lines were also evaluated as powdery mildew resistant and high quality by the farmers in major oriental melon production areas, so these cultivars were finally registered in 'Korea Seed and Variety Service' by the name of „Gukbo‟(G2Gink×San1) and „ObokPlus‟. (G2Gink×SanY) in 2010 and commercialized in 2011.

Out of these markers, two markers, CmPMR7 and CmPMR10 were identified as most closely linked to the resistance locus within 2cM interval and showed susceptible homozygous genotype "rr" in all the commercial oriental melon cultivars used in this study. 7. Four oriental melon cultivars resistant to powdery mildew disease were developed by crossing inbred lines developed in this study using DNA marker system and elite inbred lines developed in previous breeding program. Among these 4 lines, 2 showed excellent horticultural characteristics and powdery mildew resistance in field test. These two lines were also evaluated as powdery mildew resistant and high quality by the farmers in major oriental melon production areas, so these cultivars were finally registered in 'Korea Seed and Variety Service' by the name of „Gukbo‟(G2Gink×San1) and „ObokPlus‟. (G2Gink×SanY) in 2010 and commercialized in 2011.
