Rhododendron, a broad-leaved evergreen tree which belongs to Ericaceae family and Rhododendron species, is distributed widely from the temperate to the frigid zones of both northern and southern hemisphere. It grows natively on the Baekdu Mountain Range in Korea having more than 1000 m altitude mainly, while now it is on the verge of extinction. Since its leaf show green even in the winter season and it blooms beautiful flowers, it has the considerable cold tolerance and high ornamental value which can be the useful characteristics for the extension of the cultivated area to the northern part of South Korea. To elucidate the mechanism of the cold tolerance of the Korean Rhododendron species and to gain the basic characteristics for the cultivation technique, their ecological environment and their morphological, physiological and biochemical changes during winter season were investigated with the comparison to the introduced Rhododendron species which are more cold-sensitive one.
Rhododendron brachycarpum, a Korean native species, grows mainly at an altitude of 1200 to 1526 m above sea level with the slope of 30 to 45 degree in the investigated sites. Betula platyphylla, Quercus aliena, Sorbus commixta, and Acer pseudo-sieboldianum are composed of the most upper part of its forest with the high frequency and the shrubs such as Magnolia sieboldii, Thuja koraiensis, Rhododendron mucronulatum, and Tripterygium regelii for the middle part, and the herbaceous plants such as Cacalia auriculata, Veratrum oxysepalum, Dryopteris crassirhizoma, Lycopodium annotinum, and Sphagnum palustre for the most lower part, which grow favorably under the environment with relatively high humidity and half-shading.
Leaf burning after winter season was most severe in the ‘Nova Zembla’ of the 7 Rhododendron species tested for 2 years, which showed 14% in 2010 and 94% in 2011 when it was extremely cold winter. However, Rhododendron brachycarpum and R. brachycarpum var. roseum, the Korean native species, did not present the cold damage, which showed more active leaf curling and change in leaf angle for the prevention of desiccation during the cold winter weather and also earlier recovery of photosynthetic activity and stomatal conductivity after winter season.
Sucrose content of leaves measured in the mid-winter was higher in the R. brachycarpum var. roseum than ‘Nova Zembla’, suggesting that the accumulated sucrose affects its cold tolerance by prevention from the injury of the membrane and dehydration caused from freezing of intercellular water. On the other hand, activity of the antioxidative enzymes such as peroxidase and catalase was higher in ‘Nova Zembla’ than R. brachycarpum var. roseum, representing that the reactive oxygen is generated more under higher stress like desiccation.
Also proteomic analysis suggested that the cold tolerance of R. brachycarpum var. roseum is determined by the increase in the scavenge of the reactive oxygen’s generated during and after winter and the better water movement even at the low temperature.
Leaf burning of ‘Parker's Pink’ showing relatively low cold tolerance was reduced when the transpiration inhibitors were treated after winter season, suggesting that leaf burning, an indicator for the cold tolerance of evergreen trees, is developed by the dehydration of the leaves.
From the above results, it is suggested that the cold tolerance of the Korean native Rhododendron species is determined by the better adaptation against the imbalance of water status in the leaves caused by the dehydration during the cold winter season and the actively transpirating spring season. Furthermore, the development of the technique for reducing water stress and the technique for the acclimation to the low temperature, and its avoidance are necessary for the extension of the cultivating area of Rhododendron species.
