Leuconostoc 속은 김치 발효 초기에 생육하여 김치의 적절한 맛과 산도를 부여하는 유산균으로 김치 산패를 유발하는 Lb. plantarum을 경쟁적으로 억제하는 Leuconostoc을 선발하여 김치의 품질을 향상시키고 유통기간을 연장하는 스타터로 개발 가능성이 있다. 따라서 김치 내에서 균들의 생육을 조절하고 김치의 저장성과 안전성을 향상시키고자 김치로부터 항균활성이 우수한 Leuconostoc및 박테리오신 생산 유산균을 동정하고 분리 균주가 생산하는 박테리오신의 특성을 확인하고자 한다. 그 결과 김치로부터 박테리오신을 생산하는 균주를 분리하여 L. citreum BS14을 동정하였고 L. citreum에서 처음으로 박테리오신 유전자의 유무를 확인하였다. 이 후 L. citreum BS14에서 분리한 조박테리오신의 특성을 확인한 결과 높은 열에 안정하고 다양한 pH 처리 하에서 항균활성이 안정하였으며 일반적으로 박테리오신이 그람 양성균을 저해하는 것에 비해 본 연구에서 분리한 박테리오신은 그람 음성 균주까지도 저해하여 넓은 저해 범위를 갖는 장점이 있다. 또한 단백질 분해 효소에 의해 분해되어 단백질로 구성된 물질일 것으로 예상된다. 따라서 본 실험에서 분리한 박테리오신은 다양한 환경 변화에서 안정하고 체내에 섭취되었을 때 단백질 가수분해 효소에 의해 분해 됨으로 인체에 영향을 주지 않을 것으로 생각된다. 또한 본 연구에서 분리한 L. citreum BS14이 생산하는 박테리오신은 분자량이 10 kDa이며 열에 안정하고 Li. mococytogenes를 강하게 저해하는 특성에 따라 α-helix로 이루어진 Class IIa에 해당하는 박테리오신으로 추정된다. Many lactic acid bacteria (LAB) produce bacteriocins which exhibit antimicrobial activities against spoilage or pathogenic strains. Owing to the increased demand for natural food additives to keep safety and to control fermentation rate of kimchi, the development of starters that produces broad and strong antimicrobial bacteriocins is of great necessity. Leuconostoc species makes a growth in early phase of kimchi fermentation and gives an appropriate flavor and acidity to kimchi and thus the bacteriocin producing starters may have a great potential as natural food biopreservatives. The objective of this study was to screen and characterize the bacteriocin producing LAB from kimchi and to investigate the applicability of those in kimchi fermentation. For screening of bacteriocin producing LAB, gram-positive and gram-negative bacteria were used as indicators and for measurement of antimicrobial activities of isolates, agar well diffusion assay method was used. As results, Leuconostoc citreum BS14 and Lactobacillus sakei B16 were isolated with their outstanding and broad antimicrobial spectrum against many homotype-LAB, gram positive and negative pathogens in kimchi. Their antimicrobial activities were maintained over broad pH range from pH2 to 9 and high temperature levels such as 121°C at 15min, but they were dissppeared by proteolytic enzyme treatment such as trypsin, proteinase K and α-chymotrypsin revealing their bacteriocin-like protein based structure. The B14 gene was amplified to give 294 bp fragment by PCR and the gene was anticipated to encode a 98-residues bacteriocin which is classified to class IIa by phylogenetic analysis of the hypothetical genes. This is the first report on a bacteriocin coding gene from L. citreum. Beside, the B16 gene was amplified to give 587bp product encoding sakacin P-like bacteriocin and it was also classified to class IIa. The bacteriocin producing LAB strains isolated in this study are regarded as potential candidates to be used as starters for extension of shelf life and imprevment of kimchi quality. Furthermore, genetic information of bacteriocin genes discovered in this study can be used for development of food-grade expression system for biotechnological application of LAB in foods.
