국내에서 생산 유통 중인 고춧가루에서 병원성 대장균 오염도 조사를 실시하였다. 병원성 대장균의 오염도 조사를 위해 지역별로 채취한 고춧가루 50점을 사용하였다. 병원성 대장균의 확인은 분자생물학적 방법인 PCR(Polymerase Chain Reaction)법을 사용하였고 real-time PCR 장비를 활용하였다. 고춧가루 시료의 전처리는 DNA Extraction Kit를 사용하여 시료 중 DNA를 분리하고 정제하여 분석용 시액으로 사용하였다. 4종류의 병원성 대장균(EAEC, EPEC, EHEC, ETEC)의 검출은 각각 target gene을 검출할 수 있는 primer와 반응시켜 시료 중 병원성 대장균의 종류를 확인하였다. 실험 결과는 시료 50점 중 1점에서 병원성 대장균 검출로 오염도는 2% 이며, 병원성 대장균의 종류는 EAEC(장관 부착성 대장균, Enteroadherent E.coli)형으로 확인되었다. 또한 대장균을 저감할 수 있는 방법으로 알콜 처리 및 UV 조사를 제시하였고 두 가지 방법으로 감소 효율을 조사하여 보았다. 무균 고춧가루 시료에 대장균 표준균주를 인위적으로 접종 후 알콜을 처리하지 않고 배양한 것을 대조군으로 사용하였으며, 대장균이 접종된 시료에 알콜을 농도별로 희석하여 처리하고 선택배지를 사용하여 대장균수를 확인한 결과, 대조군은 대장균 수가 1.8 × 106 CFU/mL 으로 검출되었고, 알콜 10% 처리하여 10분 경과하였을 경우 대장균 수는 1.1 × 106 CFU/mL의 검출로 102 CFU/mL정도의 감소 정도를 보였으며, 시료에 알콜 20% 이상의 처리한 실험군은 대장균 수가 전혀 검출되지 않았다. 이에 고춧가루에 20% 이상 농도로 10분 이상 침지 알콜 처리는 대장균 살균에 효과가 있음을 알 수 있었다. 또한, 병원성 대장균을 저감할 수 있는 또 다른 방법으로 UV 조사 시간이 대장균의 감소에 얼마나 영향을 미치는지 실험하였고 위와 같은 방법으로 무균 고춧가루 시료에 대장균 표준균주를 인위적으로 접종 후 UV를 조사하지 않은 시료를 대조군으로 하였으며, 접종한 시료에 UV를 시간별로 구분하여 조사 후 선택배지를 사용하여 대장균 수를 확인한 결과 UV를 조사하지 않은 대조군의 대장균 수는 5.0 × 105 CFU/mL 이었고, 5분간 UV를 조사 후 확인한 대장균 수는 3.0 × 103 CFU/mL, 15분간 UV를 조사 후 확인한 대장균 수는 2.0 × 103 CFU/mL, 30분간 UV를 조사 후 확인한 대장균 수는 1.0 × 103 CFU/mL, 45분간 UV를 조사 후 확인한 대장균 수는 2.0 × 103 CFU/mL이었고, 60분과 120분의 UV 조사의 실험군에서는 대장균 수가 검출되지 않았다. This study was conducted to investigate the level of contamination of pathogenic E. coli in the 50 red pepper powders collected in domestic regions. The pathogenic E. coli was confirmed using real-time PCR. DNA in samples was extracted using DNA extraction kit and the refined extracted solution was used for analysis. To confirm the 4 types of pathogenic E. coli such as EAEC(Enteroadherent E. coli), EPEC(Enteropathogenic E. coli), EHEC(Enteroheamorrhagic E. coli) and ETEC(Enterotoxigenic E. coli), the samples were reacted with primer for target gene detection. Only 1 sample among the 50 samples was contaminated with pathogenic E. coli. The level of contamination was 2%. The type of pathogenic E. coli detected in the sample was EAEC(Enteroadherent E. coli). This study was also conducted to determine the effect of alcohol treatment and UV irradiation on reduction of E. coli population in the pepper powder. Control was the sample without alcohol treatment after inoculating E. coli reference strain in the red pepper powder. The number of E. coli colonies was determined on selective agar after treating E. coli-inoculated samples with the different concentration of alcohol. The number of E. coli in control was 1.8 × 106 CFU/mL. The number of E. coli in 10 minutes immersion treatment with 10% alcohol was 1.1 × 106 CFU/mL, resulting in a hundredfold decline. In over 20% alcohol-treated samples, E. coli was not detected. This result showed that over 20% alcohol-treatment for at least 10 minutes of immersion was effective on reduction of E. coli. This study was also conducted to determine the effect of UV irradiation time on E. coli reduction. Control was samples without UV treatment after inoculating E. coli reference strain in red pepper powder. The number of E. coli colonies on selective agar was determined after treating E. coli-inoculated samples with different duration of UV irradiation. The number of E. coli in control group was 5.0 × 105 CFU/mL. The number of E. coli in the 5 min of UV irradiated sample was 3.0 × 103 CFU/mL, 2.0 × 103 CFU/mL in 15 min UV irradiated sample, 1.0 × 103 CFU/mL in 30 min UV irradiated sample, and 2.0 × 103 CFU/mL in 45 min UV irradiated sample.
