The mechanism of learning and memory in the brain is promoted by the regulation of the activity of proteins inducing synthesis. And diet n-3 polyunsaturated fatty acid associated with synapse plasticity, membrane fluidity and signal molecule in brain; cerebral cortex, hippocampus. In this study, we investigated the effect of n-3 polyunsaturated fatty acid and expression of proteins in relation to cognitive function. Rats were divided into two groups and fed with ground seed and oil diets of Perilla frutescens for a three-weeks (young group) and three months (old group) feeding period. Each rat model from the different groups were subjected to behavioral and memory tests using the T-maze Test to compare the learning and memory development. After this, test animals were sacrificed and the brain tissues we collected, particularly cerebral cortex and hippocampus - the areas of the brain responsible for cognitive function. Proteins from the brain samples were subjected to 2D gel separation and proteins with differentially expressed were identified using MALDI-TOF and Q-TOF MS. And We identified expression of proteins from results with western blot and immunohistochemistry analysis. Based on the results, n-3 polyunsaturated fatty acids diet on enhaced cognitive function by increasing the expression proteins. Some proteins associated with cognition.
