This study was conducted to investigate physicochemical properties and functional ability of black Doraji. It was analyzed proximate composition, reducing sugar, free sugars, free amino acids, organic acids, minerals and measured Hunter color values and texture. The Doraji was extracted with hot-water and methanol for their antioxidant activities and cell cytotoxicity.
The results obtained from this study were as follows:
1. The black Doraji contained lower level of moisture with 73.96% than raw Doraji. Black Doraji has the higher level of crude lipid(1.15%), crude ash(3.05%) and crude protein(2.58%) than raw Doraji, respectively. Black Doraji contained lower level of carbohydrate with 16.32% than raw Doraji.
2. Black Doraji contained high level of reducing sugar with 9128.33 mg/100g. Arginine was detected to be the major free amino acids in Doraji regardless of processing condition, and composition of glutamic acid was the highest in Doraji. Free sugar content of black Doraji was 7270.22 mg/100g, and was higher than that of raw Doraji, and total organic acid contents of black Doraji was increased after heat treatment, because the acetic acid and lactic acid be formed during heat treatment at 60℃ and other organic acids were increased. Mineral content of the black Doraji was higher than those of raw doraji, and potassium was prominent mineral in Doraji, constituting 88% of black Doraji and 79% of raw Doraji. Black Doraji was higher crude saponin contents than raw Doraji.
3. The black Doraji showed significantly higher value compared with raw Doraji in L, a and b value (p<0.001). The hardness, chewiness and gumminess of raw Doraji were significantly higher than those of black Doraji (p<0.001).
4. Antioxidant activities of hot-water and methanol extracts from Doraji were evaluated, and the results are below;
First, methanol extract from black Doraji contained the highest levels of total polyphenols among the extracts. Total polyphenol contents of methanol extracts were higher than those of hot-water extracts, and extract from balck Doraji higher than extract from raw Doraji. As quantitation results of phenolic acid by HPLC, black Doraji extracts significantly more than raw Doraji extracts. Total flavonoids contents of hot-water extract from black Dorajiwas the highest(7.94 mgQE/g) among the samples.
Second, Electron donating ability(EDA) increased according to increasing concentrations of the extracts of the Doraji. The methanol extracts were higher EDA than hot-water extracts, and EDA of black Doraji were higher than that of raw Doraji. The methanol extract of black Doraji has the highest antioxidant effect of among extracts. The IC50 values of hot-water extract of raw Doraji(RW), hot-water extract of black Doraji(BW), methanol extract of raw Doraji(RM) and methanol extract of black Doraji(BM) were 3123.57 µg/mL, 1154.99 µg/mL, 3043.78 µg/mL and 937.38 µg/mL, repectively.
Third, ABTS radical scavenging ability increased according to increasing concentrations of the Doraji extracts. The BM was the highest ability of among the extracts. The IC50 values of RW, BW, RM and BM were 137.18 µg/mL, 70.35 µg/mL, 78.19 µg/mL and 50.07 µg/mL, respectively.
Fourth, hydroxyl radical scavenging ability of extracts from Doraji was increased according to increase of extract concentration. The hydroxyl radical scavenging ability of BM was the highest among the extracts at concentration 2000 µg/mL, and the BW was the highest at concentration 8000 µg/mL. The IC50 values of RW, BW, RM and BM were 5632.55 µg/mL, 5376.33 µg/mL, 5107.26 µg/mL and 5197.60 µg/mL, respectively.
5. Each extracts showed a little different antibacterial activity according to the tested strains. The BW revealed the strongest activity. The hot-water extracts were higher activity than methanol extracts.
6. The anti-inflammatory effects of extracts from Doraji in lipopolysaccharide- induced RAW 264.7 cells. In order to examine the cytotoxicity of Doraji, the cell viability rate was measured using MTT assay. All Doraji extracts except the BW had cell cytotoxicity at 900 µg/mL. All Doraji extracts inhibited concentration-dependently the NO production upon stimulation by lipopolysaccharide in RAW 264.7 cell. Although the RM was the highest NO inhibition, BW was the best NO inhibition due to low cell cytotocity at even high extract concentration.
As in this study, black Doraji showed higher amount of nutrients and saponin, more effect of antioxidant activity and anti-microbial activity than raw Doraji. These results will provide fundamental data for improving sitological value and black Doraji can be used a valuable resource for development of nutraceutical foods.
