In this study, we evaluated the anti-obesity effects of Nelumbo nucifera and Eriobotrya japonica extracts using 3T3-L1 adipocytes and db/db mouse. In cell-based assay, 3T3-L1 preadipocytes were stimulated with DMEM media containing 10% FBS, 0.5 mM 3-isobuthyl-1- methyxanthine (MIX), 5 μg/ml insulin, and 1 μM dexamethasone for differentiation of adipocytes. In the among various parts (leaf, seed, and root) of N. nucifera, leaf extract showed the most effective or significant effect decreasing process of adipogenesis through regulation of CCAAT enhancer binding protein α(C/EBPα) and peroxisome proliferator activated receptor γ (PPARγ) expression. Also, N. nucifera leaf extract at concentration of 125 and 250 μg/ml inhibited lipase enzyme activity to 28.7 and 81.3%. The addition of N. nucifera leaf extract inhibited the apototic morphologic changes such as small nuclei and nuclear fragmentation in 3T3-L1 adipocytes, whereas the activation of uncoupling protein-2(UCP-2) related to energy expenditure was increased compared with control group. The oral administration of N. nucifera leaf extract (100 or 400 mg/kg body weight/day) significantly reduced food efficacy, bodyweight, and serum and liver total cholesterol levels in db/db mice as well as diet-induced obese mice. In visceral and subcutaneous fat, administration of N. nucifera leaf extract significantly decreased adipocyte size and expression of C/EBPα and PPARγ in comparison with those of db/db mice. Then, leaf extract of N. nucifera was dissolved in H2O and sequentially fractionated with EtOAc and n-butanol. The EtOAc, n-butanol, and H2O fractions clearly inhibited the accumulation of lipid droplet and PPARγ expression in 3T3-L1 adipocytes. However, the expression of UCP-2 only activated to EtOAc fraction in 3T3-L1 adipocytes. Luteolin, component isolated from leaf of N. nucifera, reduced the accumulation of lipid droplet in 3T3-L1 adipocytes. Also, the seed and leaf extracts of E. japonica effectively inhibited the accumulations of lipid droplet and C/EBPα expression. E. japonica leaf extract at concentration of 500 and 1000 μg/ml inhibited lipase enzyme activity to 5.2 and 65.1%. Thus, these results indicate that N. nucifera or E. japonica leaf extract should be able to regulate the adipogenesis and lipolysis through regulation of transcription factors (C/EBPα and PPARγ), UCP-2, and fat size in 3T3-L1 adipocytes and db/db mice or inhibit the adipogenesis through the decrease of C/EBPα expression in 3T3-L1 adipocytes.
