Aim of this thesis is to understand molecular basis of plant innate immunity which renders plants endure and survive in the hostile environment. In the first chapter, it will be given general description of known layers of plant innate immune systems with reviewing published literatures. Plant innate immunity consisted of MAMP triggered immunity (MTI) which is begun with recognizing common structures of microbes and effector triggered immunity (ETI) which is specialized immunity starts with recognizing certain effector proteins secreted by pathogens through type III secretion systems (TTSS). In addition, systemic acquired resistance as a result of ETI or induced systemic resistance triggered by non-pathogenic microbes, and defense priming associated with induced resistance will be discussed. The following chapter II contains the first report describing bacterial RNAs among diverse bacterial genetic materials could be a novel MAMP to increase plant immune responses against pathogenic bacteria. Plants recognize non-self structure that is general cues from invading pathogens referred to as microbe associated molecular patterns (MAMPs) such as chitin from fungi, flagellin and elongation factor of bacteria. Plant immunity triggered by the MAMP perception is addressed as MAMP triggered immunity which primes plant defense responses to subsequence attack of pathogens. The basic structures of MAMPs, MAMP receptors and the signaling process of MAMP perception are conserved both in animal and plants. Nucleic acid-MAMPs such as bacterial CpG DNA, bacterial mRNAs, 16S ribosomal RNA were reported while unmethylated plasmid DNA from bacteria was only reported determinant inducing plant immunity. The pre-treatment of RNAs from Pseudomonas syringae pv. tomato DC3000 enhanced resistance to subsequent infection of the same bacteria in Arabidopsis thaliana. In addition, bacterial RNAs application on leaf disks induced superoxide anion production, callose deposition, MAPK cascade activation, and defense gene expression. The ribosomal RNA which is the most abundant RNA species from P. syringae pv. tomato DC3000 also could mount most of plant defense response as total bacterial RNAs led to the bacterial rRNA might be the determinant triggering plant immune responses and could be a novel nucleic acid MAMP in plant. In the chapter III, it will be provided new evidence that aphid phloem sucking herbivore increased plant systemic immunity against the biotrophic bacterial pathogen Xanthomonas axonopodis in chili pepper (Capsicum anuum L.). As one of the wide distributed plant pathogen, herbivore elicits plant defense responses in diverse aspects. Conversely, pre-challenge with the compatible pathogen X. axonopodis on pepper leaves significantly reduced aphid numbers. Interestingly, the bacterial populations of the beneficial plant growth-promoting rhizobacteria Bacillus subtilis GB03 was higher than control, the saprophyte Pseudomonas fluorescens Pf-5 was same as control, and the pathogenic Ralstonia solanacearum SL1931 was lower than control at 7 days after aphid infestation. This study provides a new understanding of tri-trophic (insect-plant-beneficial root bacteria) interactions and their role in the induction of defence mechanisms. These results provide new insight into the existence of VOC signal molecule produced by P. polymyxa for plant growth promotion and P. polymyxa specific long-chain VOC tridecane that serves as a bacterial determinant of induced resistance in plants. Collectively, this study provided information about layered mechanisms of plant innate immunity, proposed bacterial ribosomal RNA as a novel nucleic acid MAMP in plant and provided new evidence of multi-trophic interactions and their role in the induction of plant defence mechanisms.
