Diabetes mellitus is a chronic disease caused by inherited and/or acquired deficiency in production of insulin by the pancreas. In diabetic patients blood glucose levels increase and/or insulin sensitivity decrease. Insulin is key hormone produced by pancreatic beta cells, which controls blood glucose. To cure diabetes agents are used to promote insulin secretion, or improve insulin sensitivity of pancreatic beta cells. Polyamine is known as the essential element in the beta cell insulin secretion. In addition, polyamines are present in high concentrations within pancreatic beta cells. Deficiency of polyamines such as putrescine, spermine and spermidine has been reported to cause damage to the insulin production and secretion. In particular, it has been reported that triamine, spermine has a capability to promote insulin secretion from pancreatic beta cells. In this study, we investigated the insulin secreting activities of diamines such as ethylenediamine, trimethylenediamine, putrescine and cadaverine using islet β-cell line and normal or diabetic mice and found they have ability to promote insulin secretion. First, we examined insulin secretion from mouse islet β-cell line(MIN-6) in response to addition of various concentrations of diamines(0.1 μM to 10 μM). Addition of all diamines tested at concentrations above 10 μM for 20 min significantly enhanced basal insulin secretion levels. However, an increase in glucose-stimulated insulin secretion was found only at 100 μM diamines except trimethylenediamine, which enhanced the insulin secretion above 1 μM. All diamines tested except ethylenediamine were found to exert positive effects on both basal and glucose-stimulation insulin secretion concentrations above 10 μM when the cells were treated with these agents for 24 hr. The insulin secretion potentials of cells treated with these diamines for 24 hr were approximately 1.5 times higher than those for 20 min. A maximal insulin secretion level of cells treated with each diamine was approximately 3.5 to 5 times higher than that of untreated cells. Among the transcription factors tested MafA and Nkx2.2 were highly expressed in cells treated with diamines. OGTT was performed after treating diamines in normal mouse. We found that only trimethylenediamine decreases blood glucose levels at only time point of 15 min after oral administration of glucose. However, other diamines did not significantly affect the blood glucose level in normal mice. STZ-induced diabetic mice were injected with trimethylenediamine or ethylenediamine for 3 wk and revealed that these diamines slightly decrease blood glucose levels at the end of injection time point compared to control. Further study on the influence of diamines in type 2 diabetic mouse model is needed. Moreover, the study to elucidate the relationship between the enhanced insulin secretion and the up-regulation of transcription factors such as MafA and Nkx2.2 in daimine-treated pancreatic cells is needed. In conclusion, we report here that diamines promote insulin secretion from pancreatic beta cells for the first time. After further examination, we may consider these diamines as potential insulin secretagogues.
