Wasabi (Wasabi japonica Matsum.) is a perennial plant that is a member of the Cruciferaceae family and native to Japan. Allyl isothiocyanate (AITC) found in wasabi, which has been noted for its sterilization and insecticidal effect, stabilization of vitamin C, improvement of appetite, inhibition of platelet aggregation, inhibition of carcinogen activity, prevention of tooth-decay and inhibition of senility. AITC is a secondary metabolite containing N and S. Therefore, a diverse AITC content can be obtained according to variety, nutrient status, climate, environment, growing conditions, etc. Wasabi is difficult to cultivate due to its long-term cultivation period for rhizome production and complicated cultivation method, which makes the cultivation area decrease. This study was conducted to find out the effect of chitosan concentration, sulphur concentration and water management on growth and AITC content of wasabi leaf grown in hydroponic system.
Results of the 1st experiment to find out the effect of level of chitosan solution (0, 100, 500 and 1000 ppm) and chitosan application methods (drench or spray methods) on photosynthesis, transpiration rate and chlorophyll content of the wasabi plants showed the highest values with 1000 ppm chitosan treatment. The root activity tended to increase as chitosan concentrations increased with both chitosan application methods at 30 days after treatment. But the fresh weight of the plants with the chitosan treatment were lower than those with control. On the other hand, AITC content of wasabi leaf tended to increase with increasing chitosan concentration. AITC content of wasabi leaf with chitosan spray method was higher than that with drench method.
Results of the 2nd experiment to find out the effect of sulphur treatment (x0.5, x1, x2, x3 and x5 of sulphur level in Yamazaki nutrient solution, which correspond to 0.5, 1, 2, 3 and 5 me∙L-1) on the photosynthesis, chlorophyll content, plant growth, vitamin C and AITC contents of the wasabi cultivated on the NFT system showed increase of the photosynthesis and chlorophyll content at 1 me∙L-1 and 2 me∙L-1 of sulphur treatment. The fresh weight of wasabi decreased at 0.5 me∙L-1 and 3 me∙L-1 of sulphur treatment. The highest vitamin C and AITC contents in the leaves were obtained with the sulphur treatment of 3 me∙L-1. Whereas the lowest contents were obtained with the sulphur treatments of 0.5 me∙L-1 and 3 me∙L-1.
Results of the 3rd experiment to find out the effect of sulphur level (1, 2, 3 and 5 me∙L-1) and a fixed level of chitosan treatment (1000 ppm) by spray method on the photosynthesis, chlorophyll content and plant growth of the wasabi showed decrease of the photosynthesis, chlorophyll content and plant growth with increasing sulphur level from 1 me∙L-1 to 5 me∙L-1. AITC content of the wasabi leaves increased with increasing sulphur level from 1 me∙L-1 to 3 me∙L-1, whereas the lowest AITC content was obtained at the highest sulphur level of 5 me∙L-1.
Results of the 4th experiment to find out the effect of irrigation method on the photosynthesis, chlorophyll content and plant growth of the wasabi showed that the photosynthesis, chlorophyll content and plant growth were the highest with the twice a day-15 minute irrigation method. The lowest photosynthesis, chlorophyll content and plant growth were obtained at the twice every other day-15 minute irrigation method. The photosynthesis, chlorophyll content and plant growth irrigation length according to irrigation length per day were the same for all irrigation methods. Although the total amount of the irrigated water by the three irrigation methods (the twice a day-15 minute, six times a day-5 minute and twice every other day-15 minute irrigation methods) were the same the photosynthesis, chlorophyll content and plant growth of wasabi by the three irrigation methods were different, which was explained by difference in irrigation interval. The highest vitamin C content of the leaf was obtained by the once a day-15 minute irrigation method. On the other hand, the lowest vitamin C content was obtained with the six times a day-5 minute irrigation method.
Results of the 5th experiment to find out the effect of polyethylene glycol (PEG) level (1, 2, 3 and 5%) on the photosynthesis, root activity and plant growth of the wasabi under the DFT system showed that the photosynthesis, root activity and plant growth decreased with increasing concentration of PEG, compared with the control. However, the photosynthesis, root activity and plant growth increased again at the highest concentration of PEG of 5%. AITC content of leaf increased with increasing PEG concentrations from 1% to 3%, whereas it tended to decrease at the highest PEG concentration of 5%.
