Pattern recognition receptor (PRR) toll-like receptors (TLRs), interleukins (IL), interferon (IFN) 그리고 the IFN stimulated genes (ISGs) 들은 숙주의 항바이러스성 비특이면역의 초기반응에 있어서 중추적인 역할들을 한다. 본 연구의 in vivo 실험은 감염초기 뿐 아니라 VHSV 감염 후 회복되는 과정에서의 이러한 비특이면역 인자들의 역할에 대해 알아보기 위해 수행되었다. 감염실험용 넙치는 107.8 TCID50/ml 농도의 VHSV 를 주사 한 후 15°C 또는 20°C 에 사육하였다. 바이러스 감염 후 3, 6, 12 시간 그룹과 1 일, 2 일, 4 일, 7 일 그룹에서 무작위로 5 마리씩 샘플을 취했다. 조직으로부터 RNA 를 추출하여 cDNA 를 합성한 후 real-time PCR 을 실시하였다. 15°C 그룹에서는 바이러스 genomic RNA 의 copy 수는 바이러스 감염 2 일째에 가장 높았으며 7 일째까지 높은 수치를 유지하였다. 반면에 20°C 그룹에서는 감염 후 1 일째에 가장 높았으나 이후 급속도로 감소되었다. 15°C 그룹에서 바이러스의 mRNA 수치는 감염 후 3 시간째부터 점차 증가하기 시작하여 12 시간째에 최고수치를 기록하였으며 2 일째까지 높은 수치를 유지하였다. 반면에 20°C 그룹은 4 일과 7 일째에 감지할 수 없을 정도의 낮은 수치를 보였다. 감염 후 3 시간째에 15°C 그룹에서는 TLR 2, TLR 7, interleukin 8 (IL 8), IFN regulatory factor 3 (IRF 3), IRF 7 그리고 ISG 15 수치가 증가하였고, 같은 시간대의 20°C 그룹에서는 TLR 2, IRF 3, IRF 7 그리고 ISG 15 이 control 그룹에 비해 증가하였으나 TLR 7, ILs 그리고 Mx 은 control 그룹과 유사하게 나타났다. 바이러스 copy 수가 높을 경우, IL 1β, IRF 3, IRF 7, IFN Type I, ISG 15 그리고 Mx 면역유전자들의 발현 수치가 두 그룹 모두에서 높았으며, 20°C 그룹에서의 발현이 15°C 그룹에서의 발현보다 빠르고 강하게 나타났다. 바이러스 수가 감소하였을 것으로 예상되는 시점인 실험말기의 15°C 그룹에서는 IL 1β 그리고 Mx 를 제외한 다른 유전자 발현 수치는 감소하였지만 20°C 그룹의 경우 모든 유전자 발현수치가 감소하였다. 15°C 그룹에서는, 감염 후 12 시간째 면역 유전자와 바이러스 copy 수 사이에 밀접한 연관관계를 보였다. 15°C 의 감염 후 7 일째 샘플에서 높은 수치의 바이러스 copy 수를 보였으나 Mx 를 제외한 면역유전자들의 발현율은 낮았다. 흥미롭게도, 바이러스 copy 수가 높은 1 일, 2 일째의 15°C 그룹에서 TLR 2 와 TLR 7 은 감소되었다. 반면에 20°C 그룹에서는 TLR 2 가 지속적으로 높게 발현되었으며 TLR 7 은 21 시간째부터 2 일째까지 특히 높게 발현되었다. 따라서, 15°C 에서 TLR 과 ISGs 가 발현 감소 및 지연되고 더 높은 폐사율을 나타내는 것을 볼 때 20°C 에서 사육되는 넙치에서 신속하고 높게 발현되는 TLRs 과 ISGs 들이 숙주 생존에 중요한 역할을 할 것으로 생각된다. 두 온도 그룹에서의 온도의존적 apoptosis 관련 유전자 발현 연구를 진행하였다. Type II IFN 발현은 바이러스 copy 수가 증가하면서 같이 증가하였으며 20°C 그룹에서 15°C 그룹보다 빠르고 높게 나타났다. The MHC class I 과 CD8 발현은 두 그룹 모두에서 초기단계인 3- 6 시간째에 높게 나타났으나 후기단계 2-7 일째에서는 15°C 그룹의 경우 control 보다 낮게 나타났고 20°C 그룹의 경우 control 보다 높게 나타났다. Granzyme 의 경우 15°C 에서는 2 일째에만 발현되었으나, 20°C 에서는 지속적으로 발현되었다. Perforin 을 높게 발현하는 개체에서는 caspase 3 또한 높게 발현하였다. TNFα, FasL, p53 의 발현율은 초기단계에 15°C 그룹에서 20°C 보다 높았다. Caspase 3 발현은 20°C 에서는 증가되었으나 15°C 에서는 낮게 나타났다. 흥미롭게도 15°C 그룹의 개체들 중 caspase 3 발현율이 높은 개체들은 viral RNA copy 수가 낮았다. 본 실험을 통해서 VHSV 에 감염된 넙치는 높은 온도에서의 효율적인 apoptotic 면역 반응이 숙주의 생존에 중요한 역할을 한다는 것을 알 수 있다. Imiquimod (IQ)와 poly I:C 는 바이러스에 대한 숙주의 자연면역을 유도하기 위한 TLR ligand 로 많이 이용되는 물질이다. Poly I:C 는 VHSV 에 대한 넙치의 면역을 유도하기 위해 사용되어 왔으나 IQ 는 시도된 적이 없었다. TLR7 은 ssRNA 형태인 IQ 를 인식하여 면역 작용을 일으키기 때문에 VHSV 에 의한 TRL7 의 작용을 보기 위해서는 IQ 가 더 좋은 후보물질이 될 것으로 여겨진다. 그 효과를 알아보기 위해 넙치를 15°C 에서 사육하고 VHSV 감염실험을 수행하기 이틀 전에 50 μg IQ, 100 μg IQ 또는 100 μg poly I:C 를 주사하여 면역작용을 유도하였다. Poly I:C 를 투여한 그룹에서는 5%의 폐사율을 보이며 VHSV 에 대한 높은 저항성을 보였으나 50 μg IQ, 100 μg IQ 를 처리한 그룹에서는 각각 85%, 75%의 높은 폐사율을 보였다. IQ 와 poly I:C 는 각각 TLR7 과 TLR3 유도하는 것으로 알려져있는데 본 실험에서는 흥미롭게도 poly I:C 에 의해 TLR7 은 유도가 되었으나, IQ 에 의해서는 TLR3 가 유도되지 않았다. Poly I:C 는 Mx 와 type I IFN 을 아주 강하게 유도하였으나 IQ 는 Mx 는 높게 유도하였으나 type I IFN 은 유도하지 않았다. Caspase 3 는 두 물질에 의해 유도되지 않았다. 다른 실험으로 넙치의 신장세포에 IQ 와 poly I:C 를 처리한 후 (12 시간 후) VHSV 를 감염시켜 결과를 관찰하였다(pre-treated group). 또 다른 그룹은 IQ 를 처리하기 전에 (12 시간) VHSV 를 감염시켰다. The viral haemorrhagic septicaemia virus (VHSV) causes high mortality of olive flounder (Paralichthys olivaceus) during winter and spring season (8-15°C) but no mortality in summer (over 20°C). VHSV is a negative sense ssRNA virus belonging to the genus Novirhabdovirus in the family Rhabdoviridae. Differential viral replication, viral transcription and host immune response in VHSV infected olive flounder reared at 15°C, at which temperature host is susceptible to VHSV infection, and 20°C, at which temperature the host is not susceptible to the viral infection, were studied by real-time PCR. Toll-like receptors (TLRs), interleukins (IL), type I interferon (IFN) and IFN-stimulated genes (ISGs) play pivotal roles in early response of antiviral innate immunity of a host. The present in vivo experiment was conducted to investigate roles of these innate immune factors in early phase as well as during recovery from VHSV infection. The olive flounder were given intra-peritoneal injection of VHSV (107.8 TCID50/fish) and were maintained at 15°C or 20°C. A mock injection was given to the respective controls. Post infection, 5 fish were randomly sampled in each group for head kidney at 3, 6, 12 hour post infection (hpi), 1, 2, 4, and 7 days post infection (dpi). Total RNA extracted from the head kidney was reverse transcribed and used as template for real-time PCR. In the 15°C group, the number of viral gRNA copies peaked after 2 dpi and remained high through 7 dpi, while in the 20°C group; the copy number was at the highest at 1 dpi but drastically declined at 2 dpi and later stages. Viral mRNA levels in the 15°C group gradually increased starting at 3 hpi to reach their maximum value at 12 hpi and remained high until 2 dpi, whereas the group at 20°C showed much lower viral transcripts that were undetectably low at 4 and 7 dpi. At 3 hpi, viral infection caused in a relative fold increase of TLR 2, TLR 7, interleukin 8 (IL-8), IFN regulatory factor 3 (IRF 3), IRF 7, and ISG 15 in 15°C group. At the same time, fish at 20°C showed expression higher than control in TLR 2, IRF 3, IRF 7 and ISG 15 but TLR 7, ILs and Mx found near to the control level. When viral copy numbers were high, a higher expression of immune genes IL-1β, IRF 3, IRF 7, IFN Type I, ISG 15 and Mx was observed in both the groups, whereas in 20°C group these genes expressed earlier and stronger than 15°C group. At the end of the experiment (7 dpi), when viral load in the host declined, expression levels of the studied immune genes were also reduced, except IL-1β and Mx in 15°C group; however, at the same time in 20°C group all the genes showed decline in their expression level. In 15°C group, a high degree of correlation was observed between immune genes and viral copy number in each of the sampled fish at 12 hpi. A VHSV infected olive flounder sampled from 15°C group, with ascites at 7 dpi displayed high viral copy but under expressed immune genes except for Mx. Interestingly, when viral copies were high at 1 and 2 dpi, both TLR 2 and TLR 7 were down-regulated in 15°C group. Conversely, in 20°C group, TLR 2 expressed consistently higher, while TLR 7 expressed significantly higher from 12 hpi to 2 dpi, during which the individuals carried high viral gRNA, perhaps indicating immune suppression of these TLRs by the virus at 15°C. Thus, a quicker and stronger expression of TLRs and ISGs in the olive flounder reared at 20°C could play crucial role in host survival, while down regulation of TLRs and delayed expression of ISGs could be the reason for larger mortality of VHSV infected olive flounder reared at 15°C. The immune gene expression study was extended to understand response of apoptosis related genes in the two temperature groups, 15°C and 20°C. Type II IFN expression increased as the viral copies increased and the 20°C group showed quicker and stronger expression than the 15°C group. The MHC class I and CD8 expression was high in both the groups at early stage of infection (3-6 hpi) but at later stages (2-7 dpi) in 15°C group expression reduced below control levels, while they expressed higher to control in 20°C group. The expression of granzyme in 15°C group showed a single peak at 2 dpi, but was consistently expressing in 20°C group. Individuals expressed high levels of perforin expressed very high levels of caspase 3. In 15°C group, TNFα FasL and p53 expressed significantly higher than 20°C only at initial stages of infection (3-6 hpi). Caspase 3 expression found to be low in 15°C fish group, whereas it was significantly elevated in 20°C group. However, IQ treated group showed high rate of mortality, where 50µg IQ and 100µg IQ group had a mortality rate of 85% and 75% respectively while the control group showed 75% mortality. To check the immune response upon IQ and poly I:C stimulation, the olive flounder reared at 15°C were given intra peritoneal injection of 100µg IQ and 100µg poly I:C and sampled at 6, 12 hour post stimulation (hps) and 1, 2 and 4 day post stimulation (dps) for head kidney. The expression of TLR 3 and TLR 7 were stimulated in pre-treated kidney cells and the expression level was similar with or without VHSV infection in the pre-treated kidney cells. In unstimulated kidney cells, TLR 7 expressed close to the control level after VHSV infection. Moreover, addition of IQ into the VHSV infected kidney cells failed to elevate TLR 7 signifying the TLR 7 inhibitory mechanism by VHSV. Expression of MDA5, IRF 3 and IRF 7 were elevated by both the ligands, the rate of transcription was very low in unstimulated kidney cells after VHSV infection. As observed in vivo, type I IFN level remained near to control level in IQ treated olive flounder as well as in the unstimulated group, while poly I:C caused robust expression of type I IFN. Pre-stimulation of olive flounder kidney cells with IQ or poly I:C resulted induction of innate immune genes, but a lower and delayed expression in response to IQ caused no host protection against VHSV. TLR 7 down regulation in VHSV infected kidney cells was noticed confirming the in vivo observation. From these studies, it can be concluded that a quicker and stronger immune response in olive flounder reared at 20°C or pre-treated with poly I:C ensured host safety whereas, at 15°C delayed immune response due to effective viral inhibitory action on host immunity led to a large scale of host mortality.
The viral haemorrhagic septicaemia virus (VHSV) causes high mortality of olive flounder (Paralichthys olivaceus) during winter and spring season (8-15°C) but no mortality in summer (over 20°C). VHSV is a negative sense ssRNA virus belonging to the genus Novirhabdovirus in the family Rhabdoviridae. Differential viral replication, viral transcription and host immune response in VHSV infected olive flounder reared at 15°C, at which temperature host is susceptible to VHSV infection, and 20°C, at which temperature the host is not susceptible to the viral infection, were studied by real-time PCR. Toll-like receptors (TLRs), interleukins (IL), type I interferon (IFN) and IFN-stimulated genes (ISGs) play pivotal roles in early response of antiviral innate immunity of a host. The present in vivo experiment was conducted to investigate roles of these innate immune factors in early phase as well as during recovery from VHSV infection. The olive flounder were given intra-peritoneal injection of VHSV (107.8 TCID50/fish) and were maintained at 15°C or 20°C. A mock injection was given to the respective controls. Post infection, 5 fish were randomly sampled in each group for head kidney at 3, 6, 12 hour post infection (hpi), 1, 2, 4, and 7 days post infection (dpi). Total RNA extracted from the head kidney was reverse transcribed and used as template for real-time PCR. In the 15°C group, the number of viral gRNA copies peaked after 2 dpi and remained high through 7 dpi, while in the 20°C group; the copy number was at the highest at 1 dpi but drastically declined at 2 dpi and later stages. Viral mRNA levels in the 15°C group gradually increased starting at 3 hpi to reach their maximum value at 12 hpi and remained high until 2 dpi, whereas the group at 20°C showed much lower viral transcripts that were undetectably low at 4 and 7 dpi. At 3 hpi, viral infection caused in a relative fold increase of TLR 2, TLR 7, interleukin 8 (IL-8), IFN regulatory factor 3 (IRF 3), IRF 7, and ISG 15 in 15°C group. At the same time, fish at 20°C showed expression higher than control in TLR 2, IRF 3, IRF 7 and ISG 15 but TLR 7, ILs and Mx found near to the control level. When viral copy numbers were high, a higher expression of immune genes IL-1β, IRF 3, IRF 7, IFN Type I, ISG 15 and Mx was observed in both the groups, whereas in 20°C group these genes expressed earlier and stronger than 15°C group. At the end of the experiment (7 dpi), when viral load in the host declined, expression levels of the studied immune genes were also reduced, except IL-1β and Mx in 15°C group; however, at the same time in 20°C group all the genes showed decline in their expression level. In 15°C group, a high degree of correlation was observed between immune genes and viral copy number in each of the sampled fish at 12 hpi. A VHSV infected olive flounder sampled from 15°C group, with ascites at 7 dpi displayed high viral copy but under expressed immune genes except for Mx. Interestingly, when viral copies were high at 1 and 2 dpi, both TLR 2 and TLR 7 were down-regulated in 15°C group. Conversely, in 20°C group, TLR 2 expressed consistently higher, while TLR 7 expressed significantly higher from 12 hpi to 2 dpi, during which the individuals carried high viral gRNA, perhaps indicating immune suppression of these TLRs by the virus at 15°C. Thus, a quicker and stronger expression of TLRs and ISGs in the olive flounder reared at 20°C could play crucial role in host survival, while down regulation of TLRs and delayed expression of ISGs could be the reason for larger mortality of VHSV infected olive flounder reared at 15°C. The immune gene expression study was extended to understand response of apoptosis related genes in the two temperature groups, 15°C and 20°C. Type II IFN expression increased as the viral copies increased and the 20°C group showed quicker and stronger expression than the 15°C group. The MHC class I and CD8 expression was high in both the groups at early stage of infection (3-6 hpi) but at later stages (2-7 dpi) in 15°C group expression reduced below control levels, while they expressed higher to control in 20°C group. The expression of granzyme in 15°C group showed a single peak at 2 dpi, but was consistently expressing in 20°C group. Individuals expressed high levels of perforin expressed very high levels of caspase 3. In 15°C group, TNFα FasL and p53 expressed significantly higher than 20°C only at initial stages of infection (3-6 hpi). Caspase 3 expression found to be low in 15°C fish group, whereas it was significantly elevated in 20°C group.
