논문 요약 홍삼나노분말과 홍삼가수분해물의 콜라겐유발 관절염 예방효과 동물실험 및 이를 첨가한 아시아고 치즈의 이화학적 특성 최경훈 식품공학과 세종대학교 대학원 본 연구는 홍삼 나노분말과 홍삼가수분해물의 콜라겐유발 관절염에 대한 예방효과를 동물실험을 통하여 규명하고, 이를 (0.1, 0.3, 0.5 %) 첨가한 아시아고 치즈의 숙성 중 이화학특성을 관찰하는데 목적이 있다. 인삼은 항암효과, 염증성질환 회복, 노화억제, 암세포 증식억제 등 수 많은 생리작용 및 약리작용을 가지고 있어 특히 동양에서 널리 쓰이던 약초이다. 홍삼은 백삼을 찌고 말린 것으로, 백삼에서 존재하지 않는 Rg3, Rg5, Rk1과 같은 사포닌 성분이 증숙 과정을 통해 생산되며, 부패, 손상을 막고, 장기간 저장이 가능하여 인삼을 섭취하는 가장 좋은 형태로 알려져 있다. 홍삼은 각광받는 건강식품원료 중 하나로 환, 캅셀, 엑기스 등의 다양한 형태로 가공되어 판매되고 있으며 홍삼을 원료로 하는 건강기능성 식품에 관한 연구도 꾸준히 진행되고 있다. 하지만 이러한 기능적 특성에도 불구하고 홍삼 내 주요 생리활성물질인 진세노사이드 (ginsenoside)가 체내 흡수율이 극히 제한되기 때문에 이를 극복하고자 나노분말화와 효소처리가수분해를 통하여 생체이용률을 극대화 하고자 하였다. 아시아고 치즈 (Asiago cheese)는 이탈리아 알프스 유래 경질 치즈로써, 그 맛과 향이 파마산 치즈 (Parmesan cheese), 체다 치즈 (Cheddar cheese)와 유사하고, 숙성취 등이 강하지 않아 보편적인 소비자의 입맛에 알맞다. 숙성 기간이 경과함에 따라 약한 단 맛에서 쓴 맛을 띄게 되며, 숙성 기간에 따라 그에 따른 음식의 사용도 다양하다. 이와 같은 특성은 홍삼나노분말과 홍삼가수분해물 첨가 기능성치즈 개발에 적합하다고 판단되었으며, 실험에 따른 결과는 다음과 같다. 첫 번째 연구로 홍삼나노분말과 일반분말의 이화학적 특성과 진세노사이드의 정량분석을 실시하였다. 입자의 형태를 관찰하기 위해 SEM(scanning electron microscope)을 사용하였으며, 입자의 크기 측정은 particle size analyzer로 측정하였다. 나노분말의 경우 둥근 입자들이 응집되어 있는 형태가 관찰되었으며, 입자의 크기가 고르게 분포한 것을 관찰하였다. 일반분말의 경우 입자의 크기가 고르지 않았으며 길죽한 형태를 나타내었다. Particle size analyzer를 통해 나노분말은 200nm 일반 분말의 경우 120 μm 으로 나타났다. Tap density 에서는 홍삼나노분말이 0.3030 g/ml로 일반분말에 비해 두 배 이상 작은 것으로 나타났다. pH의 경우 용액의 농도가 증가 할수록 내려갔으며, 나노분말의 경우 모든 농도에서 pH가 일반분말에 비해 낮았다. 색도에서는 L, a, b 값 모두에서 유의적으로 다른 값을 보였다. 보수성과 보유성에서도 나노분말이 일반분말에 비해 두 배 이상 큰 것으로 나타나서, 보수성과 보유성이 뛰어난 것으로 밝혀졌다. 안식각에서는 분말의 양이 증가할수록 유의적으로 나노분말은 감소폭이 작았으며, 일반분말의 경우 급격히 낮아지는 것으로 나타났다. 마지막으로, 진세노사이드 정량 분석에서 각각 추출 횟수를 달리 하여 실시하였는데, 세 번째 추출에서 홍삼나노분말의 경우 총 진세노사이드의 함량이 4913.6 mg/kg으로 일반분말에 비해 약 30 % 높았다. 홍삼 나노분말과 일반분말의 이화학적 특성은 다양한 분석을 통하여 유의적으로 달랐으며 (p<0.05), 총 진세노사이드 함량이 증가하였으므로, 나노분말화를 통하여 홍삼의 기능성 및 이화학적 특성이 개선되었다고 사료되었다. 두 번째 연구에서는, 홍삼나노분말과 홍삼가수분해물의 증가한 효능을 규명하기 위하여 관절염 유발 모델인 DBA/1J 마우스를 통해서 관절염 예방효과를 알아보았다. 관절염 유발 1주일 전부터 홍삼 (홍삼일반분말, 홍삼나노분말, 홍삼추출물, 홍삼가수분해물)을 급여하였고, 35일간에 걸쳐서 관절염의 증상 정도를 관찰하였다. 투여그룹은 일반 (saline 0.2ml), 대조군 (saline 0.2ml), methotrexate (0.3 mg/kg body weight), 홍삼일반분말 (0.15 mg), 홍삼나노분말 (0.15 mg), 홍삼추출물 (0.2 mg), 홍삼가수분해물 (0.2 mg)으로 관절염 유발 1주일 전부터 실험 종료 시까지 해당 시료를 경구투여 하였다. 관절염의 증상 정도는 관절염 중증도, 뒷발의 부종, 혈청분석, 병리조직학적 분석, 비장인덱스를 통하여 관찰하였고, 그 결과는 다음과 같다. 관절염유발로 대조군에서 체중 감소가 나타났으나, 홍삼 가수분해물은 1.18g, 홍삼 나노분말 그룹은 0.95g 증가하는 것으로 나타났다. 관절염 유발율과 뒷발의 부종 측정에서도 홍삼 나노분말과 홍삼 가수분해물이 홍삼 일반분말과 추출물에 비하여 긍정적인 결과가 관찰되었다. 혈청분석은 루미넥스 (LuminexTM) 시스템을 통해 분석하였으며, 혈청 내 IgG1과 IgM은 관절염유발로 인해 증가하였으나 홍삼나노분말과 홍삼가수분해물에서는 대조군에 유의적으로 낮았다 (p<0.05). 관절염증상에 중요한 역할을 하는 싸이토카인인 TNF-α 는 관절염유발로 인해 증가하였으며, 홍삼나노분말 (4.72 pg/ml)과 홍삼 가수분해물 (3.89 pg/ml)이 각각 홍삼일반분말 (5.83 pg/ml)과 홍삼추출물 (6.61 pg/ml)에 비해 유의적으로 낮았다. 병리조

Chapter 1. Physicochemical properties of nanopowdered red ginseng 1. Abstract This study was carried out to investigate the physicochemical properties of nanopowdered red ginseng (NRG) and to demonstrate the quantitative measurement of ginsenoside in NRG. To determine the physicochemical properties of the NRG, the size of particles, proximate analysis, pH, tap density, color, solubility, swelling power, and water and oil holding capacities of ginsenoside were measured. In result, size of NRG was ranged from 250-500 nm, proximate composition analysis revealed that the NRG and powdered red ginseng (PRG) had 5.29 and 5.66 % moisture, 2.42 and 2.14 % lipid, and 2.01 and 1.55 % ash, respectively. The pH value was decreased from 5.6 to 5.4 when the concentration were increased from 2 to 8%, and in case of PRG, the pH value was decreased from 5.7 to 5.5 with the concentration increased from 2 to 8% (p<0.05). However, with 8-10% of concentration, pHs were about 5.4 in both NRG and PRG. Tap densities were 0.3030 and 0.6667 g/cm3 in NRG and PRG, respectively. Color analysis revealed that L* values were 92.40 and 86.00, a* values 4.72 and 2.89, b* values 16.70 and 21.89, respectively, with significant differences (p<0.05). Water and oil holding capacities were nearly 2-3 times higher in NRG than in PRG (p<0.05). In quantitative analysis, total ginsenosides content was about 30 % higher in NRG (4609.30 mg/kg) than PRG (3335.71 mg/kg). In conclusion, NRG was significantly different from PRG in almost all parameters that were measured (p<0.05). In conclusion, NRG was significantly different from PRG in almost all properties and nanosizing markedly increased the total ginsenosides of red ginseng. To determine the effects, the symptoms and severities in CIA mice treated with NRG, PRG, RGH, RGE, and methotrexate (rheumatoid arthritis medicine, MTX) were measured by the body weight, clinical score, paw thickness, serum analysis (IL-6, TNF-α, IgM, and IgG1), histological changes, and spleen indexes. The results of ginseng treatments were observed significant different in the assessments such as changes of body weight, clinical score, and paw thickness of the CIA mice compared with control and it was likely considered to delay or prevent the induction with type II collagen. In analysis of cytokines, the RGH and NRG contained 3.58 and 4.72 pg/ml TNF-α and 33.75 and 34.73 pg/ml IL-6 respectively with significant differences with the control (p<0.05). NRG and RGH were lower than PRG and RGE respectively. IgM were 5532.52, 6753.02, 5938.84, and 6848.64 ng/ml in serum of NRG, PRG, RGH, and RGE, respectively, with significant differences (p<0.05) and IgG1 was also observed similar in IgM. In histological changes, it was scored by erosin, fibrosis and inflammation of bone and cartilage respectively, NRG, PRG, RGH, and RGE were significantly lower than that of control (p<0.05). In color, the additions of NRG and RGH effected on color of Asiago cheese with significant differences during ripening period at 14°C for 4 month (p<0.05). LAB were decreased from 3.50×109 to 2.95×107 CFU/ml in 0.5 NRGAC during ripening period. While, the addition of RGH in Asiago cheese had no effect on LAB counts. In texture analysis, the additions of NRG and RGH into cheese influenced texture of cheese. NRG and PRG made cheese harder than control while, RGH and RGE made cheese softer than control with significant difference. In appearance, yellowness, and cheese eye were significantly higher in NRGAC than control during 4 months (p<0.05). In flavor and taste, ginseng, astringency, and bitterness were significantly higher in NRGAC and RGHAC than in control due to unique flavor and taste of red ginseng (p<0.05). In overall acceptability, decreasing concentrations of NRG and RGH in cheese were higher than that of control. In conclusion, the addition of NRG, PRG, RGH, and RGE into Asiago cheese influenced some factors of physicochemical properties, but, in sensory evaluations, lowest concentrations of NRGAC and RGHAC were better than control in preferences. In flavor and taste, ginseng, astringency, and bitterness were significantly higher in NRGAC and RGHAC than in control due to unique flavor and taste of red ginseng (p<0.05). In overall acceptability, decreasing concentrations of NRG and RGH in cheese were higher than that of control. In conclusion, the addition of NRG, PRG, RGH, and RGE into Asiago cheese influenced some factors of physicochemical properties, but, in sensory evaluations, lowest concentrations of NRGAC and RGHAC were better than control in preferences.
