Caloric restriction (CR) is the most effective non-invasive regimen retarding the aging process in laboratory model organisms and possibly in human. In spite of accumulated knowledge on the aging and CR for several decades, the direct evidence is insufficient to explain interaction between cellular senescence, glucose levels, and genomic instability in chromosome level. In this study, we grew human dermal fibroblasts (HDFs) in culture medium containing different concentrations of glucose (0.55, 5.5 and 25 mM) and observed karyotype and chromosomal morphologies during cellular senescence. We found that chromosomal abnormalities were gradually increased toward late passage and toward higher glucose treatment. There were several structural abnormalities including appearance of dicentric chromosomes during cellular senescence. Interestingly, numerical abnormalities were gradually decreased toward late passage and were not associated with glucose levels. In particular, chromosome 10 trisomy was the dominant numerical abnormality observed in this specific HDF culture. In conclusion, our results indicate that cellular senescence accompanied with mechanical inaccuracy for chromosomal segregation, and then followed by diverse structural abnormalities because of impairment of DNA repair. Low glucose (CR) ameliorates structural abnormalities of chromosome, and high glucose accelerates these abnormalities.
