Among thousands visualized spots on 2-DE gels, improvement of the detection of low abundant proteins (LAPs) is essential for enhancing the experimental quality. A new method for prefractionation of major abundant proteins such as glycinin, β-conglycinin, and ribulose-1,5-bisphosphate carboxylase / oxygenase (RuBisCO) in soybean seeds and leaves, respectively, was developed to enrich LAPs from plant extracts, by means of the Tris-Mg/NP-40 extraction buffer with protamine sulfate precipitation (PSP), named PSP method. Fractionated proteins with gradient PS concentrations were analyzed by 13% SDS-PAGE to confirm the minimum concentration used for fractionation for different soybean tissues. Reducing of high abundant proteins (HAPs) was accompanied with the increase of PS percentage, and suggesting that 0.05% and 0.1% PS in Tris-Mg/NP-40 buffer were determined as the minimum concentration in seed and leaf samples, respectively. Most glycinins and RuBisCO large and small subunits (LSU and SSU) were fractionated into PS-precipitation. 2-DE analyses revealed that LAPs of soybean seeds and leaves were more displayed and enriched in the PS-supernatant than that of total extract proteins.
