PepMoV-Vb1의 CI는 PepMoV-Vb1에 감염되고 액포가 팽창된 식물세포 안에서 세포벽과 가까이 위치한 세포질 부위에 부분적으로 가는실 형태의 모습으로 발현되는 것을 확인할 수 있었다. 이러한 결과들은 기존에 보고된 내용과 비교하여 PepMoV 또는 potyvirus의 각 단백질들이 식물세포 내 어떤 부위과 관련되어 발현되는지 확인할 수 있었으며, 궁극적으로 potyvirus의 식물세포 내 이동경로와 식물유전자들과의 상호작용에 대한 연구에 많은 도움을 줄 수 있을 것으로 사료된다. PepMoV-Vb1의 게놈을 기초로 식물에 감염되면 초록색 형광을 발현하는 pSP6PepMoV-Vb1/GFP 벡터를 이용하여 바이오에탄올 생산과 관련된 cellulase의 한 종류인 Clostridium cellulovorans에서 분리된 endoglucanase D (EngD)를 pSP6PepMoV-Vb1/GFP의 NIb 유전자와 CP 유전자 사이에 GFP 유전자를 제거하고 삽입을 하여 pSP6PepMoV-Vb1/EngD 벡터를 개발하였다. 개발된 pSP6PepMoV-Vb1/EngD를 SP6 promoter를 이용하여 in vitro transcripts 생산하고, N. benthamiana에 접종을 하여 대조군인 PepMoV-Vb1 감염된 식물체와 비교를 하였다. 그 결과 PepMoV-Vb1/EngD에 감염된 식물체는 대조군보다 병징이 조금 약하였고, PepMoV-Vb1/EngD의 NIb 일부분과 CP 부위를 검정하기 위한 RT-PCR 분석에서는 목표된 밴드가 나타나는 것을 확인할 수 었다. 그러나 NIb-CP 부위를 검정하는 RT-PCR과 PepMoV coat protein 항체를 이용한 western blot 분석에서는 목표된 밴드 이외에도 밴드가 나타나는 것을 확인할 수 었다. PepMoV-Vb1/EngD에 감염된 식물체를 77일 까지 관찰 한 결과 대조군과 비교하여 노화가 빠르게 진행되는 것을 확인 할 수 있었고, EngD 유전자 활성도 검정되었다. 이러한 연구는 PepMoV-Vb1 안에 외래유전자인 EngD 유전자가 잘 발현되는 것을 확인할 수 있었고, 개발된 PepMoV-Vb1/EngD는 식물을 이용한 바이오에탄올 생산과정 중 효소처리 부분에 많은 도움을 줄 수 있을 것으로 사료된다. CMV에 감염된 식물을 보호하기 위해 전사후 유전자 발현 억제 (post-transcriptional gene silencing) 원리를 이용한 두 종류의 dsRNA 발현 벡터를 개발하였다. 첫 번째, agrobacterium의 도움을 받아 식물세포 내에서 dsRNA를 유도 시킬 수 있는 CMV-Ca-P1 RNA1의 1270-1629 bp와 1-300 bp 가 각각 삽입된 hpCMV1 RNAi vector와 hpCMV2 RNAi vector를 개발하였다. 개발된 벡터를 agrobacterium을 이용한 transient assay 수행하였고, 그 결과 hpCMV1 RNAi vector는 CMV-Fny에 대해서는 100 % 감염율 보이지만, CMV-Ca-P1에 대하여 22.2 %의 감염율을 나태내었고, hpCMV2 RNAi vector는 CMV-Ca-P1에 대해서는 100 % 감염율을 보이지만, CMV-Fny에 대하여 9.1 %의 감염율을 나타내었다. 식물체에 hpCMV1 RNAi vector와 hpCMV2 RNAi vector를 함께 agroinfiltration을 수행하였을 때는 CMV-Ca-P1와 CMV-Fny에 대하여 각 10 % 와 20 %의 감염율을 나타내었다. 그리고 두 벡터의 각 목표 유전자를 하나의 벡터안으로 삽입하여 hpCMV2+1 RNAi vector를 개발하였고, 식물체에 agrobacterium을 이용한 transient assay에서 수행하였다. 그 결과 CMV-Ca-P1와 CMV-Fny 에 대하여 각 10 % 와 20 %의 감염율을 나타내었다. 이러한 결과를 재확인하기 위해 hpCMV2+1 RNAi 유전자가 발현되는 N. benthamiana 형질전환체를 개발하였고, T2 세대의 각 12개식물체에 CMV-Ca-P1 와 CMV-Fny 를 접종하였다. 그 결과 CMV-Ca-P1 접종된 12개체 중 한 식물체만 감염성을 보였고, CMV-Fny 접종된 12개체 중 4개 식물체만 감염성을 나타내었다. 두 번째, RNaseIII가 부족한 박테리아 세포 내에서 dsRNA를 생성할 수 있는 CMV-Ca-P1 RNA1의 1270-1629 bp가 삽입된 L4440::Ca-P1 1270 벡터를 개발하였다. 이 벡터가 존재하는 박테리아 세포 내에서 dsRNA를 추출하여 식물잎 표면에 도포하고, 한 시간 후에 각각 CMV-Ca-P1과 CMV-Fny을 접종하였다. 그 결과, CMV-Ca-P1과 CMV-Fny에 대하여 모두 내병성을 나타내었다. 이러한 연구결과들은 CMV대한 저항성 식물개발에 위한 좋은 목표 유전자 정보를 제공할 수 있으며, 항바이러스 물질개발에 도움을 줄 수 있을 것으로 사료된다.

Many researchers have been studying the characterizations and applications of viruses to protect plant from virus infection and to produce useful foreign protein in plants. In the study, using Pepper mottle virus (PepMoV; Potyvirus) and Cucumber mosaic virus (CMV; Cucumovirus), the plant virus-associated expression system was constructed to demonstrate the subcellular localizations of viral proteins in plant cells and overexpression of foreign gene, and resistance to plant virus. To investigate the subcellular targeting of the 11 viral proteins of PepMoV-Vb1 in plant, each viral protein was inserted into a red fluorescent protein (RFP) tagged vector, and an Agrobacterium-mediated transient expression system was used. Most viral proteins (the P1, HC-Pro, P3, PIPO, CI, 6K2, NIa and CP) of the PepMoV-V1 were detected to the cytoplasm and several viral proteins (the P1, HC-Pro, P3, 6K1 and 6K2) of them also were localized to the nucleus or chloroplast in transgenic Nicotiana benthamiana line 16C and PepMoV-Vb1/GFP-infected N. benthamiana plants. The CI protein of PepMoV-Vb1 showed thread-like structures in the cytoplasm of the PepMoV-Vb1/GFP-infected N. benthamiana plant. These results will be useful to study the cell-to-cell, long distance movement and virus-host interactions of PepMoV. To produce a foreign protein production system, a foreign gene encoding, endoglucanase D (EngD) from Clostridium cellulovorans, which has multiple enzymatic activities such as endoglucanase, xylanase and exoglucanase activities, was cloned into a viral vector derived from PepMoV-Vb1/GFP. In vitro transcripts derived from the clone named as pSP6PepMoV-Vb1/EngD were infectious and caused milder symptoms than that of wild type (PepMoV-Vb1). RT-PCR analysis showed laddered target bands of the CP, partial NIb and EngD-CP regions of the PepMoV-V1/EngD compared to those of the positive control (PepMoV-Vb1-infected plant). Western blot analysis showed non-target bands as well as target bands of the CP of PepMoV-Vb1/EngD compared to those of the positive control (PepMoV-Vb1).The PepMoV-Vb1/EngD-infected N. benthamiana plants were observed until 77 dpi and the leaves of these infected plants senesced much faster than those of the healthy and PepMoV-Vb1-infected plants. The EngD gene expression of PepMoV-Vb1/EngD was observed in upper leaves from in vitro transcripts-infected N. benthamiana plants and enzyme activity of the protein was detected by the carboxymethylcellulase assay using LB agar plates containing 0.1 % carboxymethylcellulose. This study has demonstrated the construction of pSP6PepMoV-Vb1/EngD capable of conversion of a cellulose substrate for bioethanol production. To prevent infection by CMV-Ca-P1 and CMV-Fny, two different types of dsRNA expression vectors, for use in activating the post-transcriptional gene silencing (PTGS) defense system against virus infection, were designed for expression in both plant and bacterial cells. The first type of vector was constructed for expression of dsRNA derived from viral sequence in plant cells. The two constructed vectors (hpCMV1 RNAi vector and hpCMV2 RNAi vector) were designed to produce dsRNAs containing either 360 bp (1270-1629 bp) or 300 bp (1-300 bp) fragments, respectively, from RNA 1 (a replicase gene) of CMV-Ca-P1, in leaves of N. benthamiana infiltrated with Agrobacterium tumefaciens harboring either the hpCMV1 or the hpCMV2 vectors challenged 4 days later with CMV-Fny or CMV-Ca-P1. The plants expressing the dsRNAs from the hpCMV1 vector were resistant to CMV-Ca-P1, whereas those expressing dsRNA of the hpCMV2 vector were resistant to CMV-Fny. To obtain a higher level of resistance to both CMV-Ca-P1 and CMV-Fny, a new CMV RNAi vector called ‘hpCMV2+1 vector’, containing a fusion of the viral sequences introduced into the hpCMV1 and hpCMV2 vectors, was constructed and was infiltrated into plants via A. tumefaciens. The results showed that a combination of two vectors (hpCMV1 and hpCMV2 RNAi vectors) and the hpCMV2+1 RNAi vector could provide resistance to both CMV-Fny and CMV-Ca-P1. Confirming this, transgenic N. benthmiana containing the hpCMV2+1 vector conferred resistance to both CMV-Ca-P1 and CMV-Fny. The second type of the designed vectors constructed was the L4440::Ca-P1 1270 vector, which induced bacterially expressed dsRNAs in a bacterial strain lacking the dsRNA-specific endonuclease RNaseIII, and could interfere with virus infection. This vector containing sequences corresponding to nucleotides 1270-1629 of CMV-Ca-P1 RNA1, induced dsRNAs in E.coli. The crude bacterial preparations containing the dsRNAs derived from this vector were spread on the leaf surfaces of N. tabacum plants followed by inoculation of the same surfaces 1 hr later with CMV-Ca-P1 or CMV-Fny infected sap. The expressed dsRNAs from the L4440::Ca-P1 1270 vector conferred resistance to both CMV-Ca-P1 and CMV-Fny. The results of this study will provide information for the effective selection of target sequences to inhibit CMV infection and also that bacterially-expressed dsRNAs can be used as antiviral agents against viruses. In summary, these studies were investigated for the subcellular localization of the viral proteins and a foreign proteins expression using PepMoV-Vb1 and developed an antiviral dsRNA expression vector system using CMV. The results of these studies will provide information for the characterization and application of PepMoV and the effective sequences to inhibit CMV infection.

The results showed that a combination of two vectors (hpCMV1 and hpCMV2 RNAi vectors) and the hpCMV2+1 RNAi vector could provide resistance to both CMV-Fny and CMV-Ca-P1. Confirming this, transgenic N. benthmiana containing the hpCMV2+1 vector conferred resistance to both CMV-Ca-P1 and CMV-Fny. The second type of the designed vectors constructed was the L4440::Ca-P1 1270 vector, which induced bacterially expressed dsRNAs in a bacterial strain lacking the dsRNA-specific endonuclease RNaseIII, and could interfere with virus infection. This vector containing sequences corresponding to nucleotides 1270-1629 of CMV-Ca-P1 RNA1, induced dsRNAs in E.coli. The crude bacterial preparations containing the dsRNAs derived from this vector were spread on the leaf surfaces of N. tabacum plants followed by inoculation of the same surfaces 1 hr later with CMV-Ca-P1 or CMV-Fny infected sap. The expressed dsRNAs from the L4440::Ca-P1 1270 vector conferred resistance to both CMV-Ca-P1 and CMV-Fny. The results of this study will provide information for the effective selection of target sequences to inhibit CMV infection and also that bacterially-expressed dsRNAs can be used as antiviral agents against viruses. In summary, these studies were investigated for the subcellular localization of the viral proteins and a foreign proteins expression using PepMoV-Vb1 and developed an antiviral dsRNA expression vector system using CMV. The results of these studies will provide information for the characterization and application of PepMoV and the effective sequences to inhibit CMV infection.
