LPS로 염증을 야기시킨 murine macrophage cell인 RAW 264.7 세포에 1, 10, 50 ㎍/㎖ 농도의 연잎 에탄올 추출물을 처리하여 염증매개물질의 발현과 분비를 측정하였으며, 연잎 에탄올 추출물이 NF-κB pathway에 미치는 영향을 알아보기 위하여 NF-κB의 활성 정도를 측정하였다. 연잎 에탄올 추출물은 iNOS, COX-2의 mRNA 발현을 억제하여 NO와 PGE2의 생성을 감소시켰고, pro-inflammatory cytokines인 TNF-α, IL-1β, IL-6도 mRNA의 발현이 억제되어 단백질 분비 역시 감소되었다. 이러한 염증매개물질들의 발현과 분비를 조절하는 상위 기전인 NF- κB는 연잎 에탄올 추출물에 의해 활성이 감소되었으나 염증매개물질들의 억제 효과에 비하면 그 수준이 미비한 것으로 나타났다. 따라서 연잎 에탄올 추출물은 NO, PGE2, TNF-α, IL-1β, IL-6의 발현과 분비를 억제함으로써 염증 반응을 조절하는 효과를 가지고 있음을 알 수 있었다. 이를 통해 연잎은 만성질환의 증상을 완화시키는 천연물 소재로서 가치가 있다고 사료된다.

This study was designed to evaluate the anti-inflammatory effects of lotus leaf ethanol extracts (LLE) on the LPS-induced inflammatory mediators in RAW 264.7 cells and its mechanism. This study examined the inhibitory effects of LLE on the pro-inflammatory mediators such as NO, PGE2 and pro-inflammatory cytokines(TNF-α, IL-1β, and IL-6) and NF-κB activity, a transcription factor necessary for expression of pro-inflammatory mediators. In this study, LLE significantly reduced the LPS-induced expression and production of iNOS, COX-2, TNF-α, IL-1β, and IL-6. Moreover, LLE slightly reduced the LPS-induced activation of NF-κB. These results suggest that LLE has the anti-inflammatory effects by the suppression of the pro-inflammatory mediators expressions through the suppression of NF-κB activity. Therefore, this study may provide a valuable therapeutic strategy in attenuating the symptom of various chronic diseases.
