A high performance liquid chromatography with diode array detection (HPLC-DAD) method was developed and validated for the simultaneous analysis of 3 marker compounds (peucedanol-7-O-β-D-glucopyranoside, hyuganin C, and 3',4'-disenecioylkhellactone) of Peucedani japonici Radix, which is the root of Peucedanum japonicum Thunberg (Umbelliferae). Extraction conditions were optimized by sonication with methanol for 40 minutes at room temperature. The chromatographic separation was performed on an Optimapak C18 column (250 mm × 4.6 mm, 5 μm) with a gradient elution of water and acetonitrile/methanol (9/1, v/v). The extract was detected by DAD at the wavelength of 322 nm. The method was validated by assessing specificity, linearity, accuracy & precision (intra- and inter-day), limit of quantitation (LOQ), recovery and robustness. All the results were within the required criteria. Also, a UHPLC-MS/MS using Q Exactive orbitrap mass spectrometer was conducted to identify many of coumarins in Peucedani japonici Radix. As a results, Full MS and MS/MS spectrum of coumarins were obtained by UHPLC-MS/MS system. A total of 60 medicinal herbs (33 samples of Peucedani japonici Radix, 2 of Saposhnikoviae Radix, 25 of Glehniae Radix) were analyzed by the developed method, and it was found that the contents of marker compounds differed greatly. Multivariate statistical methods, such as principal component analysis (PCA) and cluster analysis (CA), were used to characterize the medicinal herbs origin. As a result, Peucedani japonici Radix was clearly distinguished from Saposhnikoviae Radix and Glehniae Radix. This study was successfully applied to monitoring and quality control of Peucedani japonici Radix as compared with Saposhnikoviae Radix and Glehniae Radix.
