Co-culture has been suggested to be an effective method for stem cell differentiation. In this work, we used porous membranes for co-culture of adipose stem cells (ASCs) and nucleus pulposus cells (NPCs), and investigated whether repeated co-cultures using porous membranes enhanced the chondrogenic differentiation of degenerative NPCs (dNPCs), which in turn stimulated the differentiation of ASCs into NPCs in a consecutive repeated co-culture. Human NPCs with confirmed low gene expression of collagen type II and aggrecan were collected, classified as dNPCs, and co-cultured with ASCs for 1 week (first co-culture). RT-PCR results showed that dNPCs co-cultured with ASCs had slightly increased collagen type II and sox-9 gene expression compared with dNPCs cultured alone. The dNPCs were then collected and co-cultured again with ASCs for 1 week (second co-culture). RT-RCR results showed that ASCs co-cultured with dNPCs from the first co-culture expressed more collagen type II than ASCs co-cultured with dNPCs expanded alone, as well as ASC controls. Additionally, dNPCs from the second co-culture expressed more collagen type II gene than dNPCs from the first co-culture. These data demonstrate that repeated co-culture can be used for efficient expansion and differentiation of cells for invertebral disc (IVD) cell therapy. In this co-culture system, some cells can be co-cultured with ASCs. They are nucleus pulposus cells, chondrocyte-like cells as well as cells that can form cartilage-like environment for chondrogenic differentiation of ASCs. As an examples, we used enchondroma cells in our experiment. As our expected, RT-PCR results showed that ASC can maintain chondrogenic differentiation for longer time. Finally, we can use three-dimensional scaffold sturcture to mimic the extracellular matrix (ECM) structure in nature tissue as a way to further promote proliferation and chondrogenic differentiation of dNPCs and ASCs. Here, we decided gelatin as natural polymer having both biocompatible and biodegradable properties for scaffold material and gelatin also have advantages with both hydrogel and the shape of the mesh characteristics. We fabricated hydrogel-based gelatin nanofibers by using electrospinning machine. We fabricated fibers of nano-diameter and micro-diameter depending on concentration of gelatin solution and investigated swelling and degradation ratio of gelatin nanofibers according to the degree of crosslinking extent. As a result, nanofibers showed differences in swelling and degradation ratio in accordance with various crosslinking condition and demonstrated that hydrogel-based gelatin nanofibers with desired properties can be fabricated for our target tissue or in vivo condition. Electrospun hydrogel-based gelatin nanofibers indicated that they have more affinity for ASCs than porous membrane(insert) used for the co-culture system. Therefore, gelatin nanofibers can be utilized as a porous membrane in co-culture system. These demonstrated that gelatin nanofibers as 3D scaffolds for co-culture of dNPCs and ASCs can positively influence cell proliferation and differentiation and they can be effective alternative for cartilage tissue engineering applications.
