(초록) 백색 지방 조직 (White adipose tissue, WAT)이 갈색지방으로 변환되는 획기적인 발견은 비만과 다른 대사 증후군의 치료에 큰 도움이 된다. DJ-1 KO 마우스는 WAT에서 β3-adrenergic 수용체의 세포 내 신호 전달을 증가시켜 WAT의 갈색지방화 유도를 통해 이전에는 알려져 있지 않았던 미토콘드리아 합성 증가, 지질 축적 감소, 혈관 신생 증가 및 약화된 자가소화작용을 비롯한 보정적인 메커니즘을 유도하였다. 그럼에도 불구하고 DJ-1 KO 마우스는 대조군에 비해 체중, 에너지 소모 등이 동일하였다. DJ-1 KO 마우스에서 사타구니 WAT에서 갈색지방화가 발생했음에도 KO 마우스의 비만도에 영향을 나타내지 않아 DJ-1 유전자가 비만 치료에 독특한 치료 전략을 제공하는 표적이 될 것으로 기대한다. 한편 DJ-1 KO 마우스 모델과 정상 식이 또는 고지방 식이한 대조군 마우스를 통해 갈색 지방조직 (Brown adipose tissue, BAT)의 특성, 열생성능, 지질 대사 및 미세 환경 조절에 대한 DJ-1 결손의 직접적인 결과를 고찰한 결과, DJ-1 KO 마우스는 심하게 백색화된 BAT 형상을 보여주었고 기능장애를 나타내었다. 즉, 지질축적의 증가, 교감 신경 활동 감소, 미토콘드리아 기능 장애, 혈관 신생감소 및 자가소화작용의 활성화 등의 기능장애가 나타났다. 이러한 BAT 기능 이상에도 불구하고 DJ-1 결손 마우스의 체중과 에너지 소모량 등에 영향을 주지 않았다. 이 연구 결과로부터 DJ-1 KO 마우스에서 BAT 기능이상이 비만유발에 아무런 영향을 주지 않았는데, 따라서 BAT에서 DJ-1의 표적화는 비만 치료에 대한 새로운 자료를 제공 할 수 있을 것으로 기대된다. 본 연구에서 DJ-1 결손 마우스가 체중변화를 나타내지 않은 이유는 피하 백색지방 (ingWAT)의 갈색지방화로 인한 열생성 증가, 지방 대사 촉진, 혈관 신생 증가 등의 BAT 보충기능을 유발한 것에 기인된다. 이 연구 결과는 DJ-1 결손으로 유발된 BAT 기능손상만으로 DJ-1 KO 마우스의 비만을 유도하는 것은 불충분하고 갈색지방화된 백색 지방세포 (beige 지방세포)가 갈색지방과 유사한 보충 기능을 수행하는 것으로 판단된다. 이상의 결과로부터 DJ-1 단백질/유전자를 표적으로 하여 비만 관리와 치료에 대한 새로운 전략을 기대할 수 있다.

(Abstract) The seminal discovery of browning of white adipose tissue (WAT) holds great promise for the treatment of obesity and metabolic syndrome. DJ-1 is evolutionarily conserved across species, and mutations in DJ-1 have been identified in Parkinson’s disease. Higher levels of DJ-1 are associated with obesity, but the underlying mechanism is less understood. Considering the classic view of DJ-1 as a pathological factor in obesity, we crave an understanding on this research and delving further into molecular and functional basis of DJ-1, particularly in the adipose tissue metabolic outcomes, informs us of its therapeutic hope for treatment of obesity. Independent studies were conducted to shed light on the previously unappreciated role of DJ-1 in white and brown adipocyte biology in mature models of obesity. The first study that I conducted was focused on the understanding of the role of DJ-1 in white adipose tissue biology in relation to obesity. We used DJ-1 knockout (KO) mouse models and wild-type littermates maintained on normal diet or high-fat diet as well as in vitro cell models to show the direct effects of DJ-1 depletion on white adipocyte phenotype, thermogenic capacity, fat metabolism, and microenvironment profile. Global DJ-1 KO mice show increased sympathetic input to WAT and β3-adrenergic receptor intracellular signaling, leading to a previously unrecognized compensatory mechanism through browning of WAT with associated characteristics, including high mitochondrial contents, reduced lipid accumulation, adequate vascularization, and attenuated autophagy. DJ-1 KO mice had normal body weight, energy balance and adiposity, which were associated with protective effects on healthy WAT expansion by hyperplasia. Our findings revealed that browning of inguinal WAT occurred in DJ-1 KO mice that do not show increased predisposition to obesity and suggest that such potential mechanism may overcome the adverse metabolic consequences of obesity independent of an effect on body weight. Here, we provide the first direct evidence that targeting DJ-1 in adipocyte metabolic health may offer a unique therapeutic strategy to treatment of obesity. In the second study, I investigate the effects of DJ-1 deficiency on brown adipose tissue (BAT) function in mice. DJ-1 knockout (KO) mouse models and wild-type littermates placed on normal diet or high-fat diet were utilized to demonstrate the direct consequences of DJ-1 deletion on BAT characteristics, thermogenic ability, lipid metabolism, and microenvironment regulation. Global DJ-1 KO mice had defective brown adipose tissue (BAT) activity culminating in a profound whitening of BAT. Despite aberrations in inactive BAT associated with greater lipid accretion, decreased sympathetic activity, mitochondrial dysfunction, reduced vascularity, and autophagy activation, we found that the body weight and energy balance were unaffected in male mice depleted of DJ-1. Taken together, the results of this study suggest that male DJ-1 KO mice exhibit defects in BAT activity but do not gain more weight, revealing that BAT activity is not necessarily required for predisposing DJ-1 KO mice to obesity. Therefore, therapeutic targeting of DJ-1 in BAT could provide novel insights into treatment of obesity. This thesis identified that DJ-1-null mice failed to develop more weight gain, and that they have defects in BAT activity that is compensated for by browning of subcutaneous ingWAT via metabolically healthy phenotypes accompanied with an increase in thermogenic identities, fat metabolism and tissue microenvironment profile. Our findings furthermore underscore that impaired BAT activity alone is insufficient for predisposing DJ-1 KO mice to obesity and suggest that beige adipocytes play a larger role than BAT in weight maintenance and improvements in metabolic features. Overall, our findings and conclusions established an interesting and exciting therapeutic hope that targeting DJ-1 in adipocyte metabolic health may serve as a novel and potential therapeutic intervention strategy to management and treatment of obesity.
