HPLC를 이용한 로즈마리 추출물의 카페인산과 로즈마린산을 분리한 결과 로즈마리 에탄올 추출물에 함유된 카페인산 함량은 26.13 ㎎/100 g으로 나타났으며, 로즈마린산은 2090.21 ㎎/100 g 함유되어 있는 것으로 확인되었다. The present study was intended to identify the scalp and cell damage on acid dyeing and alkali dyeing of hair and to provide measures to minimize the hair damage. To this end, natural herbal Rosmarinus Officinalis was selected to examine the presence of antioxidant, anti-inflammatory, antibacterial, and cytotoxic properties before testing with 5% and 10% of Rosmarinus Officinalis extracts in acidic hair dye and an alkaline hair dye. The experimental results shows that DPPH radical scavenging ability including antioxidant activity increases in accordance with the concentration of the Rosmarinus Officinalis extracts, and ABTS radical scavenging ability also increases depending on the concentration like DPPH. The total polyphenol content contained in Rosmarinus Officinalis extracts accounts for 120.5 ± 0.056 ㎎ (GAE)/100 g and the flavonoid, 175.1 ± 0.020 ㎎ (GAE)/100 g, respectively. According to the measurement of NO production inhibition activity induced by LPS, to evaluate the inflammatory response, NO generation was decreased with increasing concentration; measurement results of the antimicrobial activity of Rosmarinus Officinalis extracts showed the antibacterial activity in 10 ㎎/㎖ and 100 ㎎/㎖ in acne germs, and high antimicrobial activity from 1 ㎎/㎖ in Staphylococcus aureus. Raw 264.7 cell was used to measure the viability of the cells to in order to identify the cytotoxicity of Rosmarinus Officinalis extracts; the result showed high cell viability in a low concentration. HPLC was used to separate caffeine acid and rosmarinic acid in Rosmarinus Officinalis extracts; caffeic acid content of 26.13 ㎎/100 g and rosmarinic acid of 2090.21 ㎎/100 g were found to be contained in the Rosmarinus Officinalis ethanol extracts. As the result of analysis of cuticle damages using methylene blue staining, the acid-dyeing with a 10% mixture of Rosmarinus Officinalis extracts showed the lowest optical density, or the least cuticle damage; protein elution was measured to show higher protein elution in alkaline dyeing group that in acidic group. In particular, a 10% mixture of Rosmarinus Officinalis group confirmed the lowest protein elution. The measurement results of moisturizing ability of hair showed a generally lower tendency compared to the control group but much higher than normal hair and the control group when acid dyed with 5% mixture of Rosmarinus Officinalis extracts, demonstrating high blocking effect of water flux of the hair containing Rosmarinus Officinalis extracts. As for the color measure, the color intensity showed increase in all Rosmarinus Officinalis extracts-added groups, decrease of green tints and increase in red tints in acid coloring depending on the increased mixture of Rosmarinus Officinalis extracts in red line dyeing. All tints increased in yellow line according to the increase of Rosmarinus Officinalis extracts. The color difference from the control group was the lowest in the acid dye group with 5% mixture of Rosmarinus Officinalis extracts, and was also low in alkali dye group with 10% mixture of Rosmarinus Officinalis extracts. Infrared spectroscopy was used to measure the oxidation reaction of the hair, showing the highest absorption in both the acid and alkali color groups without mixing Rosmarinus Officinalis extracts.
