Telomeres, play an important role in the protection of human chromosomes, consist of a repetitive DNA sequence (5’-TTAGGG-3’) and are located at the ends of chromosomes. With even division of a leukocyte, telomere shortens. Additionally leukocyte telomere length is influenced by the balance between oxidative stress and anti-oxidative response. Therefore, environmental and genetic factors can affect telomere length. Thus, the rate at which telomeres are shortened differs among individuals. Foods, nutrients, and multiple environmental factors play important roles in the regulation of telomere length. Investigations of the associations between leukocyte telomere length, and foods and nutrients are ongoing. However, published studies reported the associations between leukocyte telomere length and only a few foods and nutrients. Therefore, the present study focused on 3 types of dietary information - nutrients, foods or food groups, and dietary patterns - in an overall dietary analysis in a Korean population. To date, only the association between the Mediterranean dietary pattern and telomere length has been investigated. To find other dietary patterns influencing telomere length, this study derived dietary patterns in Korean adults and analyzed their relationships with leukocyte telomere length. It was hypothesized that nutrients with anti-oxidative properties are positively associated with leukocyte telomere length. Furthermore, foods and food groups, which have been considered to have anti-oxidative nutrients, were also hypothesized to positively influence leukocyte telomere length. This study is a prospective cohort study to collect dietary information from residents of Ansan City, Korea at beginning of the study. Their blood samples were collected after 10 years from the baseline. Collected DNA mixed with sybergeen, and primers and telomeric DNA was measured by Real-time polymerase chain reaction. Telomere length was calculated to determine the relative telomere length (TS) as the ratio of telomeric DNA (T) to single copy gene 36B4 (S). Data included leukocyte telomere length and semi-quantitative food frequency questionnaire (semi-quantitative FFQ) of 1,971 participants. Folic acid, fiber, vitamin C, and retinol intake were positively associated with TS in all participants after adjusting for confounding factors such as sex, body mass index (BMI), energy intake, leisure activity, smoking status, alcohol status, income status, and the presence of diabetes, hypertension and hyperlipidemia (all p < 0.05). In men, carbohydrate intake was positively associated with TS (p < 0.01), whereas fat was inversely associated with TS (p < 0.05). In women, retinol, vitamin B2, and folic acid intake were positively associated with TS (all p < 0.05). Regarding consumption of foods or food groups, legumes (p < 0.05), nuts (p < 0.05), seaweed (p < 0.01), fruits (p < 0.01), dairy products (p < 0.01), and coffee (p < 0.05) intake were positively associated with TS in all participants after adjusting for confounding factors, whereas red meat (p < 0.01) and sweetened carbonated beverages (p < 0.05) were inversely associated with TS. There were sex-specific associations for particular nutrients and foods in analysis stratified by sex. Dietary patterns were not significantly associated with TS. Thus, these results support the hypothesis that leukocyte telomere length is associated with dietary factors, which may regulate the balance between oxidative stress and anti-oxidative response.
