Main components of licorice are glycyrrhizin, flavonoids like liquiritigenin and its glycoside liquiritin and so on. In this study, we prepared licorice flavonoids-loaded liposomes to apply cosmetics as skin drug delivery. Liposome-in-hydrogel complex systems were developed by incorporating ceramide liposomes into cellulose hydrogel. Ceramide liposome is consisted of biocompatible lipid membranes which remained stable for over 3 weeks. The encapsulation efficiency of liquiritigenin or liquiritin-loaded liposome is 44.86%, 51.65%, respectively. Cellulose hydrogel was prepared using NaOH/urea (1 ~ 10%) solution to improve the solubility of cellulose. Because cellulose is insoluble in water or organic solvent. Through investigation of viscoelasticity, water uptake and porous structure, 6% urea/NaOH solution is the optimum condition for liposome-in-hydrogel complex system. The complex system was prepared by incorporating ceramide liposome solution into cellulose hydrogel by swelling method. Using Franz diffusion cell, skin permeability of flavonoids-loaded complex system was measured. Liposome-in-hydrogel complex system (LQG 56.55%, LQ 66.99%) had greater skin permeability of liquiritigenin and liquiritin compared to phosphate buffer (control, pH 7.4)(LQG 4.92%, LQ 5.30%) or single system of liposome (LQG 43.34%, LQ 48.97%). Liquiritin generally showed higher skin penetration than liquiritigenin. Both liposomes and liposome-hydrogel complex system showed liquiritin demonstrated greater skin permeability than liquiritigenin. These results indicate that liposome-in-hydrogel complex system is a potential drug delivery system to enhance skin permeation licorice flavonoids (liquiritigenin and liquiritin) having a anti-oxidant, anti-inflammatory properties.
