In East Asia, Hovenia dulcis Thunb. has been used in traditional herbal medicine for the treatment of liver diseases and detoxification after alcoholic poisoning. Current pharmaceutical studies have revealed further pharmaceutical applications of Hovenia dulcis Thunb. based on its hepatoprotective effect, anti-diabetic effect, and ability to reduce alcohol concentration in the blood. This research was conducted to establish for investigate the anti-hangover, stress and hepatoprotective effect of Hovenia dulcis Thunb. In the first experiment, hot water and 20 % fermented ethanol extracts of Hovenia dulcis Thunb. showed highest antioxidant activity compared with 80 % fermented ethanol extracts. Furthermore, hot water and 20 % fermented ethanol extracts of Hovenia dulcis Thunb. indicated a higher polyphenol contents than the 80 % fermented ethanol extracts. In the second experiment, ICR mice treated with hot water extracts of Hovenia dulcis Thunb. and 4 h after ethanol treatment, serum alanine aminotransferase (ALT) levels were significantly decreased. Besides, at 1 and 4 h after ethanol consumption, serum levels of acetaldehyde were decreased. In the third in vitro experiment, Hovenia dulcis Thunb. extracts showed cytoprotective and cellular antioxidant activity. Compared with control cells, the treatment of Hep G2 cells with extracts of Hovenia dulcis Thunb. showed significant cell viability promotion against hydrogen peroxide, ethanol, and acetaldehyde treatment. In addition, it was effective for inhibiting peroxyl radical-induced DCFH oxidation. In the tests for alcohol dehydrogenase (ADH) and acetaldehyde dehydrogenase (ALDH) activities using S9 rat liver extracts, hot water extracts of Hovenia dulcis Thunb. enhanced the basal activities of ADH activity by almost 60 %, and ALDH activity by almost 20 % compared with control group. In vivo experiment, AST and ALT activities were significantly lowered by the supplementation of hot water extract of Hovenia dulcis Thunb. compared with whole ethanol treated group. Moreover, the acetaldehyde level in serum was significantly lowered by hot water extract of Hovenia dulcis Thunb. treatment compared with whole ethanol treated group, too. The result of enzymatic assay in liver cytosol showed up-regulation of ADH activity about 40% compared with ethanol treated group. The expressions of NF-κB and IFN-γ in hot water extracts of Hovenia dulcis Thunb. and ethanol treated group were significantly decreased compared with whole ethanol treated group. Especially, Hovenia dulcis Thunb. hot water extract and ethanol treated group showed significant down regulation of NF-κB gene expression compared with whole ethanol treated group. Furthermore, hot water extract of Hovenia dulcis Thunb. 300 mg/kg and ethanol treated group showed significant down regulation of IFN-γ gene expression compared to Silybum marianum 300 mg/kg and ethanol treated group that is positive control. In the final experiment (chronic restraint stress model), hot water extract of Hovenia dulcis Thunb. treatment significantly increased liver and spleen indexes compared with control. Besides, the levels of AST in serum were showed significantly high by hot water extract of Hovenia dulcis Thunb. supplementation compared with control.
