This study was aimed to develop the method of beef tenderization in order to retain the original shape and lowering the hardness level below 50,000 N/m2 by using bromelain treatment under high pressure. The beef hardness parameters (pressure; 50-150 MPa, concentration of enzyme; 0.1-0.5%, and enzyme reaction time; 60-240 min) were optimized via response surface methodology (RSM) with a three-variable-six-level Central Composite Design (CCD). The enzyme concentration and reaction time were the factors affecting the hardness of beef. To verify the predicted optimum hardness conditions (45,000 N/m2), the experiments were performed at the three points selected within the optimal ranges, such as 1) 50 MPa, 0.1% and 231 min; 2) 100 MPa, 0.1% and 231 min; and 3) 150 MPa pressure, 0.1% concentration of enzyme and 231 min enzyme reaction time. The experimental values of hardness were 44,285 ± 2,406, 44,993 ± 2,572 and 46,117 ± 3,133 N/m2 respectively, which were well suited to the value (45,040 N/m2), predicted by the regression model. These results clearly show that the method applied to control the hardness of beef would be reliable. Despite decomposed muscle fiber, bromelain treated all meat samples under high pressure were retained its original shapes. Sensory evaluation of bromelain treated meat under high pressure was received higher scores in tenderness with no other distinguished characteristics. These results suggest that treated bromelain under high pressure would be possible to manufacture the softened meat with the hardness below 50,000 N/m2 while retaining the original shape.
