A. hydrophila는 기회감염균으로 겨울철보다는 주로 여름철에 균혈증이나 패혈증을 일으킨다. 본 연구결과에 의하면 LB 배지에서 A. hydrophila종 내(C4-AHL and C6-AHL)의 바이오필름 형성, 쿼럼센싱(Quorum sensing, QS) 및 엑소프로테아제(exoprotease, EP)의 활성은 15-25 oC에서 유의적으로 증가하는 것으로 나타났다(P<0.05). 또한 15 oC 이상의 온도에서 게 껍데기 중 바이오필름의 형성과 종간(autoinducers 2) 커뮤니케이션시스템에 대해 알아보았다. 실험온도 범위 내에서 바이오필름은 게 껍데기 표면의 키틴을 이용하여 게 껍데기의 틈새에서 형성됨을 전계방사형 주사전자현미경(field emission scanning electron microscopy, FESEM)으로 관찰하였다. 결론적으로 균혈증과 패혈증을 일으키는 A. hydrophila의 식품 및 식품 접촉표면에서의 바이오필름 형성은 온도 증가에 따라 증가함을 알 수 있었다. Chapter 2에서는 다양한 농도의 포도당(0.05-2.5%)에 따른 바이오필름의 형성, 운동성의 저해작용, QS 및 EP의 형성을 관찰하였다. 그 결과, 0.05% 이상의 포도당 농도에서 A. hydrophila의 QS, 바이오필름 형성, EP 형성 및 운동성이 유의적으로 (P<0.05) 감소되었다. 또한 0.05%의 포도당 농도에서는 대조군(0%)과 유사한 효과를 나타냈다. Stage shift biofilm assay 결과 역시 2.5% 포도당은 바이오필름 형성 초기에만 억제작용을 나타냈다. 더욱 중요한 것은 QS 분자인 N-3-butanoyl-DL-homoserine lactone(C4-HSL)과 N-3-hexanoyl homoserine lactone(C6-HSL)는 부분적으로 EP 활성을 보였으며, 이는 포도당 농도를 변화시킴으로써 QS를 조절할 수 있음을 시사한다. 결론적으로 식품 또는 식품보존을 위해 첨가한 포도당은 바이오필름의 형성과 질병 발생의 원인이 되는 A. hydrophila의 전반적인 QS 분자(acyl-homoserine lactones, AHLs)를 조절함을 알 수 있었다. Chapter 3에서는 수산물 중 A. hydrophila 바이오필름의 생태학적 특성을 조사하기 위해 염 농도와 배양시간에 따른 이 균의 바이오필름 형성능 연구를 수행하였다. 30 oC에서 염 농도(0-3 %)와 배양시간에 (24-96h) 따른 스테인리스스틸(SUS), 유리 및 게 껍데기 표면에서의 A. hydrophila의 바이오필름 형성, QS, EP 형성능을 확인하였다. SUS와 유리표면에서 바이오필름 형성능은 R2A 배지에 접종해 확인하였고, 게 껍데기에는 다양한 세균이 존재하므로 A. hydrophila 선택배지를 사용해 바이오필름 형성을 확인하였다. EP 활성은 FluoroTM protease assay kit를 사용하여 확인하였다. Acylhomoserine lactone (AHL) 형성은 Chromobacterium violaceum CV026을 bioreporter strain을 사용하여 확인하였고, HPLC를 이용해 재확인 및 정량하였다. Autoinducers 2 (AI-2)는 Vibrio harveyi BB170 사용하여 확인하였다. 또한 다양한 염 농도에서 바이오필름의 구조를 FESEM을 통하여 관찰하였다. 연구 결과, 24시간 배양한 샘플은 0-0.25% 염 농도에서 바이오필름과 EP 생성이 촉진되었고, AHL와 AI-2는 0.5-3.0%의 염 농도에서 감소함을 확인하였다. 또한 게 껍데기 표면에서의 바이오필름은 0-0.25%의 염 농도에서 형성됨을 확인하였고 3%의 염 농도에서는 바이오필름이 형성되지 않았으며, 게 껍데기 표면에 부착돼 있었음을 확인하였다. 0.1-3% 염 농도는 바이오필름의 형성이나 표현형에 영향을 주지 않았고 오히려 억제효과를 나타냈다. 24 h 배양한 샘플은 0-0.25% 염 농도에서 바이오필름 형성과 QS 조절유전자 발현을 촉진하였으며, 반면 0.25% 이상의 염 농도에서는 억제되었다. 96 h 배양한 A. hydrophila 샘플은 모든 염 농도에서 스트레스를 받았음이 확인되었다. Chapter 4에서는 살균소독제(에탄올, 차아염소산나트륨, 페르아세틱산, 염화벤즈알코늄)별 바이오필름 및 부유세포(planktonic cells)에 대한 저감화작용을 규명해 보고자 평판계수법과 Weibull model을 사용하였다. Crystal violet(CV) assay를 통하여 바이오필름제거 효과를 확인하였으며, FESEM을 통해 그 정도를 관찰하였다. Weibull model 적용 결과, α(scale)와 β(shape)의 R2값은 0.88-0.99사이로 높은 적합도를 나타냈다. 바이오필름은 부유세포에 비하여 살균소독제에 대한 높은 저항성을 나타냈으며, 70% 에탄올 처리는 바이오필름을 제거하는데 가장 효과적이었으며, 유의적 감소효과를 나타냈다(p < 0.05). 또한 Weibull model에서 얻은 변수 중 b-value는 바이오필름 매트릭스 제거효과와 상관관계를 보였다. 본 연구 결과, Weibull model은 화학적 소독제 처리에 의한 바이오필름 제거효과를 예측하는데 적합한 것으로 평가됐으며, 식품 및 식품접촉표면에서 살균소독제 처리에 의한 바이오필름 형성 및 식중독균의 제어효과를 확인하는데 좋은 수단으로 사용될 것으로 판단된다. Chapter 5에서는 저온플라즈마의 일종인 cold oxygen plasma(COP)에 의한 양상추 중의 A. hydrophila 바이오필름 제어효과를 확인하였다. A. hydrophila 균액에 양상추를 접종한 뒤 서로 다른 온도(4-30°C)에서 저장하여 바이오필름과 부유세포의 균수를 확인하였다. COP 처리에 의한 감소효과는 Weibull model을 통하여 확인하였다. COP 처리에 의한 제어 메커니즘을 규명하고자 FESEM과 QS assay를 진행하였다. 15 °C 이하에서 양상추에 형성된 A. hydrophila 바이오필름은 COP 5분 처리 시 최대 5 log 감소하였으며(p < 0.05), 15 °C 이상에서 형성된 바이오필름은 COP 처리에 대한 저항성이 높아짐을 확인할 수 있었다. 부유세포는 모든 온도에서 COP 15초 처리에 의해 5 log 이상의 감소 값을 나타냈다. 5 log 감소의 Td-values의 평균값은 부유세포와 바이오필름에서 각각 4.4–8.1초, 1.84–25.33분으로 나타났다. 또한 높은 온도(≥ 15 °C) 에서 QS, 바이오필름 형성 및 기공으로의 침투현상(internalization)이 유의적으로 증가함을 나타냈으며, 바이오필름 형성 증가와 기공으로의 침투현상은 COP 처리에 대한 저항성을 나타내는 주요 원인으로 사료된다. 결론적으로 본 연구에서는 온도에 대한 양상추에서 QS, 바이오필름 형성, 침투현상 및 COP 처리에 대한 감소효과를 알아보고자 하였다. Acylhomosrine lactone(AHL) QS와 autoinducers-2(AI-2)의 확인은 표준평판법으로 진행하였다. HPLC와 를 통하여 게 껍데기 배양배지 중 QS 분자의 정량분석을 하였으며, FESEM을 통하여 바이오필름의 구조를 확인하였다. 혼합배양 시 새우와 게 표면에서 P. aeruginosa와 P. fluorescence의 바이오필름의 형성은 변화가 없었으나, A. hydrophila 바이오필름 생성을 억제하였고 S. In chapter 1, it was aimed to know biofilms formation, quorum sensing and exoprotease production of Aeromonas hydrophila affected by elevated temperature from 15-37 oC. A. hydrophila is an emerging foodborne pathogens and causes higher bacteremia and septicemia during summer’s seasons rather than winter seasons. In this report, we found that 15 to 25 oC significantly (P<0.05) enhanced the biofilms formation, protease activity and quorum sensing of intraspecies (C4-AHL and C6-AHL) in LB broth. In crab surface, the elevated temperatures from 15 oC to higher stimulate the biofilms formation and interspecies communication systems (autoinducers 2). Field emission electron microscopy revealed that elevated temperatures utilize chitin surface of crab and formed biofilms inside the crack surface. We concluded that higher temperatures enhance the biofilms formation on foods and foods contact surfaces by A. hydrophila and contribute the diseases of bacteremia and septicemia. In chapter 2, we investigated biofilm formation, motility inhibition, quorum sensing, and exoprotease production of this opportunistic pathogen in response to different glucose concentrations from 0.05% (wt/vol) to 2.5% (wt/vol). In this report, we found that more than 0.05% (wt/vol) glucose significantly impaired (P <0.05) quorum sensing, biofilm formation, protease production, and swarming and swimming motility while 0.05% (wt/vol) showed the activities similar to that of the as control (0% glucose). Stage shift biofilm assay also noted that the addition of glucose (2.5%) inhibited initial biofilm formation but not later stages. More importantly, the complementation of quorum sensing molecules N-3-butanoyl-DL-homoserine lactone (C4-HSL) and N-3-hexanoyl homoserine lactone (C6-HSL) partially restored the activity of protease production, which demonstrated the role control of quorum sensing in by glucose concentrations. We conclude that glucose concentrations in food or preserved by glucose could regulate acyl-homoserine lactones (AHLs) quorum sensing molecules, which mediate biofilm formation and virulence in A. hydrophila. In chapter 3, the study was aimed to find out the effect of salinity on Aeromonas hydrophila on biofilms formation on stainless steel, glass, and crab; quorum sensing, exoprotease production with cultural age. Herein, the biofilms formation was assessed with different concentration of salinity from fresh (0%) to saline water (3.0%). To differentiate the young and old culture planktonic cells were grown at 30 oC for 24h and 96h respectively. The biofilms formation was assessed in stainless steel (SS), glass and crab and viable counts were determined in R2A agar for SS and glass but Aeromonas selective media for crab samples as foods might contain other bacteria. The exoprotease activity was assessed using FluoroTM protease assay kit. Quantification of Acylhomoserine lactone (AHL) was performed with bioreporter strain Chromobacterium violaceum CV026 and further confirmed the concentration with high performance liquid chromatography. Autoinducers 2 (AI-2) was determined with Vibrio harveyi BB170. The biofilms structure at different concentrations of salinity was done by field emission scanning electron microscopy (FESEM). Results revealed that young culture from 0 to 0.25% salinity stimulated biofilms formation, protease productions, AHL and AI-2 while reduced from 0.5% to 3.0% salinity. The FESEM images shows that 0 to 0.25% salinity forms three dimensional structures of biofilms with breaking the surfaces while 3% formed only attachment. However, in mark contrast, any concentrations of salinity (0.1 to 3%) did not stimulate the biofilms formation and phenotypic characters rather than reduced the activities. The study conclusively proved that young culture formed enhance biofilms and other regulatory genes in quorum sensing from 0 to 0.25% salinity but reduced higher than 0.25% salinity. For old cultures, any concentration of salinity was stress for A. hydrophila. The present study highlights the food ecology of A. hydrophila in water fish and other crustaceans such as shrimp and crab. In chapter 4, the present study focuses on the inactivation kinetics of various disinfectants including ethanol, sodium hypochlorite, hydrogen peroxide, peracetic acid, and benzalkonium chloride against A. hydrophila biofilms and planktonic cells. Efficacy was determined by viable plate count and compared using modified Weibull model. The removal of biofilms matrix was determined by the crystal violet (CV) assay and was confirmed by field-emission scanning electron microscope. The results revealed that all the experimental data and calculated Weibull α (scale) and β (shape) parameters had a good fit as the R2 values were between 0.88 and 0.99. Biofilms are more resistant to disinfectants than planktonic cells. Ethanol (70%) was the most effective in killing cells in the biofilms and significantly reduced (p < 0.05) biofilms matrix. The Weibull parameter b-value correlated (R2=0.6835) with the biofilms matrix removal. The present findings deduce that the Weibull model is suitable to determine biofilms matrix reduction as well as the effectiveness of chemical disinfectants on biofilms. The study showed that the Weibull model could successfully be used on food and food contact surfaces to figure out exact contact time for killing biofilms forming foodborne pathogens. In chapter 5, disease outbreaks from fresh produce have been increasing as a result of the increasing global consumption of produce. We evaluated the efficiency of cold oxygen plasma (COP), a new technology for the decontamination of fresh produce, in reducing biofilm of A. hydrophila, an emerging food-borne pathogen on lettuce. Lettuce leaves were inoculated with A. hydrophila at different temperatures to form biofilm and planktonic populations, and the inactivation kinetics of COP was determined using a modified Weibull model. To elucidate the antimicrobial mechanism of COP, we used field emission scanning electron microscopy (FESEM) and a quorum sensing (QS) assay. We found that incubation at lower temperatures (< 15 °C) significantly reduced the biofilm populations of A. hydrophila on lettuce by 5.0-log within 5 min of treatment with COP (p 5.0-log within 15 sec of incubation at any temperature. The mean Td-values for a 5-log reduction (analogous to the traditional D-value) ranged from 4.4–8.1 sec and 1.84–25.33 min for the planktonic and biofilm populations, respectively. However, higher temperatures (≥ 15 °C) resulted in a significantly higher QS (p < 0.05) as well as biofilm formation and internalization in stomata, suggesting that higher biofilm formation and internalization might be the main factors mediating resistance to COP. In summary, this study highlights the impact of temperature on the modulation of QS, biofilm formation, internalization, and COP resistance in lettuce. The natural water contains other bacteria very frequent such as Pseudomonas aeruginosa, Pseudomonas fluorescence. The aim of the study was to determine the dual species interactions of P. aeruginosa and P. fluorescence with A. hydrophila. The interactions during cross contaminations have been determined with Pectobacterium carotovorum, Salmonella Typhimurium and Listeria monocytogenes. Biofilms formations by monoculture and dual culture have been done on shrimp and crab as chitinous surface. The acylhomosrine lactone (AHL) quorum sensing and autoinducers-2 (AI-2) have been detected by using standard procedures. The HPLC and LC-MS have been performed to identify the quorum sensing molecules secreted by monocultures and mixed cultures grown in crab broth. Field emission electron microscopy has been carried out to see the biofilms structural organization. Results showed that P. aeruginosa and P. fluorescence inhibited biofilms formation of A. hydrophila without affecting their own biofilm formation capatbility on/in crab and shrimp. Dual biofilms of S. Typhimuirum, L. monocytogenes and Pectobacterium carotovorum showed neutral effect with A. hydrophila. Exoprotease productions, AHL quorum sensing as well as AI-2 were found significantly reduced for the dual culture of A. hydrophila with P. aeruginosa and P. fluorescence. The dual cultures (A. hydrophila with P. aeruginosa and A. hydrophila with P. fluorescence) formed less biofilms rather than monoculture biofilm of A. hydrophila, P. aeruginosa and P. fluorescence. Overall, the study highlighted the factors related to biofilms formation and reduction such as temperature, glucose concentrations, NaCl, chemical disinfectants, quorum sensing, and presence of other bacteria.
In chapter 1, it was aimed to know biofilms formation, quorum sensing and exoprotease production of Aeromonas hydrophila affected by elevated temperature from 15-37 oC. A. hydrophila is an emerging foodborne pathogens and causes higher bacteremia and septicemia during summer’s seasons rather than winter seasons. In this report, we found that 15 to 25 oC significantly (P<0.05) enhanced the biofilms formation, protease activity and quorum sensing of intraspecies (C4-AHL and C6-AHL) in LB broth. In crab surface, the elevated temperatures from 15 oC to higher stimulate the biofilms formation and interspecies communication systems (autoinducers 2). Field emission electron microscopy revealed that elevated temperatures utilize chitin surface of crab and formed biofilms inside the crack surface. We concluded that higher temperatures enhance the biofilms formation on foods and foods contact surfaces by A. hydrophila and contribute the diseases of bacteremia and septicemia. In chapter 2, we investigated biofilm formation, motility inhibition, quorum sensing, and exoprotease production of this opportunistic pathogen in response to different glucose concentrations from 0.05% (wt/vol) to 2.5% (wt/vol). In this report, we found that more than 0.05% (wt/vol) glucose significantly impaired (P <0.05) quorum sensing, biofilm formation, protease production, and swarming and swimming motility while 0.05% (wt/vol) showed the activities similar to that of the as control (0% glucose). Stage shift biofilm assay also noted that the addition of glucose (2.5%) inhibited initial biofilm formation but not later stages. More importantly, the complementation of quorum sensing molecules N-3-butanoyl-DL-homoserine lactone (C4-HSL) and N-3-hexanoyl homoserine lactone (C6-HSL) partially restored the activity of protease production, which demonstrated the role control of quorum sensing in by glucose concentrations. We conclude that glucose concentrations in food or preserved by glucose could regulate acyl-homoserine lactones (AHLs) quorum sensing molecules, which mediate biofilm formation and virulence in A. hydrophila. In chapter 3, the study was aimed to find out the effect of salinity on Aeromonas hydrophila on biofilms formation on stainless steel, glass, and crab; quorum sensing, exoprotease production with cultural age. Herein, the biofilms formation was assessed with different concentration of salinity from fresh (0%) to saline water (3.0%). To differentiate the young and old culture planktonic cells were grown at 30 oC for 24h and 96h respectively. The biofilms formation was assessed in stainless steel (SS), glass and crab and viable counts were determined in R2A agar for SS and glass but Aeromonas selective media for crab samples as foods might contain other bacteria. The exoprotease activity was assessed using FluoroTM protease assay kit. Quantification of Acylhomoserine lactone (AHL) was performed with bioreporter strain Chromobacterium violaceum CV026 and further confirmed the concentration with high performance liquid chromatography. Autoinducers 2 (AI-2) was determined with Vibrio harveyi BB170. The biofilms structure at different concentrations of salinity was done by field emission scanning electron microscopy (FESEM). Results revealed that young culture from 0 to 0.25% salinity stimulated biofilms formation, protease productions, AHL and AI-2 while reduced from 0.5% to 3.0% salinity. The FESEM images shows that 0 to 0.25% salinity forms three dimensional structures of biofilms with breaking the surfaces while 3% formed only attachment. However, in mark contrast, any concentrations of salinity (0.1 to 3%) did not stimulate the biofilms formation and phenotypic characters rather than reduced the activities. The study conclusively proved that young culture formed enhance biofilms and other regulatory genes in quorum sensing from 0 to 0.25% salinity but reduced higher than 0.25% salinity. For old cultures, any concentration of salinity was stress for A. hydrophila. The present study highlights the food ecology of A. hydrophila in water fish and other crustaceans such as shrimp and crab. In chapter 4, the present study focuses on the inactivation kinetics of various disinfectants including ethanol, sodium hypochlorite, hydrogen peroxide, peracetic acid, and benzalkonium chloride against A. hydrophila biofilms and planktonic cells. Efficacy was determined by viable plate count and compared using modified Weibull model. The removal of biofilms matrix was determined by the crystal violet (CV) assay and was confirmed by field-emission scanning electron microscope. The results revealed that all the experimental data and calculated Weibull α (scale) and β (shape) parameters had a good fit as the R2 values were between 0.88 and 0.99. Biofilms are more resistant to disinfectants than planktonic cells. Ethanol (70%) was the most effective in killing cells in the biofilms and significantly reduced (p < 0.05) biofilms matrix. The Weibull parameter b-value correlated (R2=0.6835) with the biofilms matrix removal. The present findings deduce that the Weibull model is suitable to determine biofilms matrix reduction as well as the effectiveness of chemical disinfectants on biofilms. The study showed that the Weibull model could successfully be used on food and food contact surfaces to figure out exact contact time for killing biofilms forming foodborne pathogens. In chapter 5, disease outbreaks from fresh produce have been increasing as a result of the increasing global consumption of produce. We evaluated the efficiency of cold oxygen plasma (COP), a new technology for the decontamination of fresh produce, in reducing biofilm of A. hydrophila, an emerging food-borne pathogen on lettuce. Lettuce leaves were inoculated with A. hydrophila at different temperatures to form biofilm and planktonic populations, and the inactivation kinetics of COP was determined using a modified Weibull model. To elucidate the antimicrobial mechanism of COP, we used field emission scanning electron microscopy (FESEM) and a quorum sensing (QS) assay. We found that incubation at lower temperatures (< 15 °C) significantly reduced the biofilm populations of A. hydrophila on lettuce by 5.0-log within 5 min of treatment with COP (p 5.0-log within 15 sec of incubation at any temperature. The mean Td-values for a 5-log reduction (analogous to the traditional D-value) ranged from 4.4–8.1 sec and 1.84–25.33 min for the planktonic and biofilm populations, respectively. However, higher temperatures (≥ 15 °C) resulted in a significantly higher QS (p < 0.05) as well as biofilm formation and internalization in stomata, suggesting that higher biofilm formation and internalization might be the main factors mediating resistance to COP. In summary, this study highlights the impact of temperature on the modulation of QS, biofilm formation, internalization, and COP resistance in lettuce.
