Vibrio parhaemolyticus는 수산물과 연관되어 있는 식중독균으로 잘 알려져 있다. 이 균은 식품공정중 식품과 식품접촉표면에서 바이오필름을 형성할 수 있다. 바이오필름은 식품원료와 가공시설의 식품안전에서 많은 관심을 받고 있는 이슈이나 식품과 식품접촉 표면에 형성된 식중독균의 바이오필름 제어에 관한 연구는 미비한 실정이다. 본 연구는 V. parahemolyticus 바이오필름의 온도별 형성능을 확인하고 이 바이오필름의 효과적인 제어법을 찾고자 한다. 첫번째 파트에서는 다양한 온도에서의 V. parhaemolyticus 바이오필름이 형성능을 Biofilm forming index (BFI) 방법과 바이오필름 형성 cell 계수법을 이용하였다. 결과적으로, 30℃에서 식품접촉표면에 많은 바이오필름의 형성과 protease 활성, AI-2 생성이 활발한 것을 확인함으로써 이 온도부근이 V. parhaemolyticus biofilm 형성의 최적 조건이라는 것을 추측할 수 있다. 본 연구는 또한 새우 껍데기 보다 게 껍데기처럼 표면이 거친 상태에서 더 많은 바이오필름이 형성되는 것을 확인하였다. 두번째 파트에서는 식품과 식품접촉표면에 형성된 바이오필름을 화학적 제어법인 차아염소산나트륨, 미산성전해수와 물리적 제어법인 UV 각각의 단일처리와 차아염소산나트륨(50-300ppm)과 UV(5-60 mW*s/cm²)의 병용처리, 미산성전해수(pH 5.8±0.1, 30ppm, 1-15min)와 UV의 병용처리에 의한 세균 저감화효과를 조사하였다. 단일처리 결과, 새우와 게 껍데기에 형성된 바이오필름이 미산성전해수의 낮은 유효염소농도에도 불구하고 차아염소산나트륨보다 더 높거나 동등한 제어효과를 보였다. 차아염소산나트륨과 UV병용처리 결과, 새우와 게 껍데기에서 형성된 바이오필름은 약 3 log10 CFU/cm² 감소치를 보인 반면, 미산성전해수와 UV 병용처리 결과에서는 최대 4.4 log10 CFU/cm² 감소값을 보였다. 본 연구는 hurdle technology가 항상 시너지 효과를 나타내지는 않는다는 것을 확인하였다. 차아염소산나트륨과 UV 병용처리는 planktonic cell에 대해 시너지 효과를 보인 반면, 미산성전해수와 UV 병용처리에서는 식품과 식품 접촉 표면에 형성된 바이오필름 제어에 시너지효과를 나타냈다. Vibiro parahaemolyticus is recognized as a human foodborne pathogen, which is mostly associated with seafood. This pathogen can form mature biofilm on food and food contact surface during food processing. Biofilm is considered as great concern in the food safety as it is resistance to common disinfectants and antimicrobials. It has been supported that the recent control methods did not provide an much reduction effect against pathogen biofilm on food contact surface as well as food surface. The purpose of this research was to assess biofilm formation at various temperatures and find an effective way to control biofilm problem in the food industry. In the 1st experiment, we evaluated V. parahaemolyticus biofilm formation at various temperatures for 3 days to check biofilm forming ability by using culturable cells count method and biofilm forming index (BFI) method on microtiter plate. Both BFI values and cultural counts showed that V. parahaemolyticus formed higher biofilm at higher temperature rather than lower temperature. The results were also confirmed by field emission scanning electron microscopy (FESEM). It was found that higher temperature (25 °C and 37 °C) formed three-dimensional structure while 4 °C forms only attachment. The food and food contact surface indicate that the 30 °C has been demonstrated as significantly higher biofilm formation and can be concluded as optimum conditions for biofilm formation by V. parahaemolyticus . The 2nd experiment was carried out to evaluate the effect of chemical treatments (sodium hypochlorite and slightly acidic electrolyzed water) and physical treatments including ultraviolet-C (UV-C) on V. parahaemolytcius planktonic and biofilm on stainless steel, shrimp and crab surface. Overall, it was found that planktonic cells were more sensitive than biofilms cells. The result showed that slightly acidic electrolyzed water had a more significant effect than sodium hypochlorite even though it has little concentration free chlorine. Sodium hypochlorite and UV-C had a significant but low reduction value on shrimp and crab surface whereas slightly acidic electrolyzed water and UV-C achieved a significantly (P < 0.05) greater reduction on both shrimp and crab surfaces. Our study indicated that hurdle technology gives synergistic effect. Sodium hypochlorite and UV-C can be used as bactericidal agents while slightly acidic electrolyzed water and UV-C can be used as effective biofilm removing agents on food contact surfaces. The study also revealed that V. parahaemolyticus biofilm was more strongly protected on crab surface than shrimp surface. V. parahaemolyticus biofilm attached in protected sites such as crevices or uneven areas are difficult to remove effectively using physical and chemical treatments confirmed by FESEM. In order to achieve a better control against V. parahaemolyticus biofilm on produce, future research should be focused on another hurdle technology for effective reduction of biofilm attached in crevices or uneven area in seafood.
