Aim of the study: Lespedeza bicolor (Fabaceae) has been used as a traditional medicine for lots of diseases like vascular, skin ailment, cough, athlete’s foot. In the present study, antioxidant, anti-inflammatory and anti-nociceptive properties of L. bicolor (MLB) were evaluated. Materials and Methods: Antioxidant potential of MLB was determined using various types of radicals such as DPPH, superoxide anion, NO radical, ABTS radical and reducing power. In this study we evaluated the anti-inflammatory activity of MLB not only in vitro but in vivo using IFN-γ and LPS-activated murine peritoneal macrophage model. We performed NO assay to check the nitrite concentration and pro-inflammatory mediators such as iNOS and COX-2 expression levels were determined by Western blotting. In order to assess the anti-inflammatory action of MLB in vivo, we used paw edema model. Results: The radical scavenging tests of MLB against DPPH radical, superoxide anion, nitric oxide and ABTS radical revealed that MLB had potent antioxidant capacity and reducing power compared to positive controls such as ascorbic acid. MLB suppressed NO production and iNOS expression via down-regulation of NF-κB activation. It also attenuated the expression of COX-2. MLB also significantly suppressed trypsin-induce paw edema. Moreover, MLB demonstrated strong and dose-dependent anti-nociceptive activities on various experimental pain models including thermal nociception and chemical nociception, compared to tramadol and indomethacin, a reference drug. In combination test using naloxone, analgesic activity of MLB was not changed indicating the anti-nociception of MLB was not due to acting as an opioid receptor agonist. Conclusion: The present results indicate that MLB can be used as valuable radical scavenging, anti-inflammatory and anti-nociceptive agent for the treatment of various diseases.
