To examine the cytotoxic effect of reactive free radicals, glucose oxidase(GO) on cultured melanocytes(B16/F10), cell viability was analysed by XTT assay after melanocytes were cultured in the media containing various concentrations of GO for 1 hours. And also, the protective effect of essential oil, Geranium oil against the cytotoxicity induced by GO was accessed. In addition, the microscopic observation was performed on melanocytes treated with GO or Geranium oil. The results were as follows : 1. GO significantly decreased cell viability in dose-dependent manner compared with control after melanocytes were treated with 5-45mU/ml of GO for 1 hours, respectively. 2. XTT90 and XTT50 values were determined at 12.6mU/ml and 37.3mU/ml of GO, respectively after cultured melanocytes were treated with GO for 1 hours. 3. The cytotoxicity value of GO was highly-toxic by the toxic criteria of Borenfreund and Puerner. 4. In the protective effect of Geranium oil on the cytotoxicity induced by GO, Geranium oil significantly increased cell viability which was decreased by GO-induced cytotoxicity after melanocytes were pretreated with Geranium oil for 2 hours before the treatment of GO. 5. In light microscopy, GO decreased cell number and cell processes in dose-dependently. While, Geranium oil increased cell number and cell processes compared with GO-treated group. From these results, it is suggested that oxygen free radicals, GO was highly-toxic effect on cultured melanocytes, and also, Geranium oil was effective in the prevention of the GO-induced cytotoxicity.
