This paper is an experimental paper regarding the Bio-MEMS(micro electro mechanical system) process technology standardization for the micro biochip fabrication. The Bio-MEMS process technology requires the micro pattern etching technology, micro bonding technology, micro fluidic technology, micro optical technology and electricity signal processing techniques. The paper was established the micro glass pattern etching standardization and the micro optical fiber cladding etching, micro glass hole etching standardization, micro glass bonding standardization and the micro fluid current and the cell control and the output voltage Vpp(peak to peak voltage ) waveform by scattering angles. The experimental materials used soda lime glass of low cost, good hydrophilic and single mode optical fiber of the good focusing on behalf of high cost polydimethyl siloxane(PDMS), polymethyl methacrylate(PMMA) and the optical lens. The Major process method of Bio-MEMS technology used the buffered oxide etchant(BOE), electro chemical discharge(ECD) and ultra violet sensitive adhesives(UVSA). All experiments were performed 10 times according to the process conditions. The first of experimental results, the stable conditions in the etching process of the glass etching and the optical fiber cladding etching is NH4F: HF ratio of the mixture to 6 : 1 and the ultrasonic agitation bath(UAB) temperature to 90℃. The glass and the optical fiber cladding was stable the surface roughness and the etching by time. Therefore, the glass etching and the optical fiber cladding etching was to establish stable etch rate conditions. The second of the experimental results, the stable conditions in the etching process of the glass hole etching is the sodium hydroxide(NaOH) concentration ratio of 35w% and the hydrogen peroxide(H2O2) ratio of the mixture to 1 : 1 and 35 V of the direct current(DC) voltage. The glass hole process used stable etching through change of the DC voltage. Therefore, the glass hole etching was to establish a stable etch rate condition. The third of the experimental results, the stable conditions in the bonding process of glass was coated uniformly Adhesive material with a thickness of 10 μm on the glass. The coated glass was aligned onto the pattern glass. The alignment glass was bonded by 10 sec of exposure using ultraviolet(365 nm, 20mW/cm2). The adhesive material is safe for the human body, mechanical characteristics, chemical characteristics, improved adhesive surface, low cost, time saving process, permeability of the adhesive surface, precision and shock. The ultraviolet sensitive adhesives method will be possible in the application of many industry fields. The fourth of experimental results, was a biochip fabricated by the standardized Bio-MEMS process technology. The cell experiment of the biochip used red blood cells. The transmitting and receiving of a laser beam used single mode optical fiber. The operating principles of the red blood cells measurement are as follows: as cells were injected into the micro channel, the red blood cells focused the He-Ne laser beam using a single mode optical fiber in the guide channel. The manufactured guide channel consists of the four inputs and the four outputs. The red blood cell was allowed through with the cylinder pump. The focusing of the fluid stream was injected into the middle channel 1 ㎕/min and the edge channel 2.5 ㎕/min. The output voltage Vpp waveforms of the scattering laser beam was measured with a photo detector. The receiving angle of the output optical fiber is 0 o, 5 o, 10 o, 15 o. The magnitude of the output voltage Vpp waveforms and the difference was measured according to the reception angle of the scattering laser beam. The magnitude of the output voltage Vpp waveforms was measured in the decrease according to increase of the reception angles. The difference of the Vpp waveforms is due to differences of the light transmittance of the red blood cells. The result of the Bio-MEMS process conditions paper was to establish standardization, simplification, quantification, low cost, short time. In addition, the paper will be expected to contribute to the application science fields of biomedical engineering, semiconductor engineering, solar cell engineering, bioinformatics engineering, genomics engineering and biochips engineering.
